Recent studies involving gonadectomy and sex hormone replacement experiments in mice have shed some light onto the molecular basis of sexual dimorphism in the adrenal cortex. Indeed, it has been shown that gonadal hormones influence many aspects of adrenal physiology, ranging from stem cell-dependent tissue turnover to steroidogenesis and X-zone dynamics. This article reviews current knowledge on adrenal cortex sexual dimorphism and the potential mechanisms underlying sex hormone influence of adrenal homeostasis. Both topics are expected to contribute to personalized and novel therapeutic approaches in the future.The maintenance of intracellular NAD+/NADH homeostasis across multiple, subcellular compartments requires the presence of NADH-shuttling proteins, which circumvent the lack of permeability of organelle membranes to these cofactors. Very little is known regarding these proteins in the methylotrophic yeast, Pichia pastoris. During the study of the subcellular locations of these shuttling proteins, which often have dual subcellular locations, it became necessary to develop new ways to detect the weak peroxisomal locations of some of these proteins. We have developed a novel variation of the traditional Bimolecular Fluorescence Complementation (BiFC), called divergent BiFC, to detect intraorganellar colocalization of two noninteracting proteins based on their proximity-based protein crowding within a small subcellular compartment, rather than on the traditional protein-protein interactions expected for BiFC. This method is used to demonstrate the partially peroxisomal location of one such P. pastoris NADH-shuttling protein, malate dehydrogenase B, only when cells are grown in oleate, but not when grown in methanol or glucose. We discuss the mode of NADH shuttling in P. pastoris and the physiological basis of the medium-dependent compartmentalization of PpMdhB.The incident second harmonic wave is a problematic issue for the precise measurement of the acoustic nonlinearity parameter. This paper proposes a compensation method to remove the effect of the incident second harmonic component in the measurement of the absolute acoustic nonlinearity parameter using the calibration method. For this, the second harmonic component detected by the receiving transducer is considered as the sum of the component due to material nonlinearity and the component included in the incident signal and a numerical calculation model is developed as a function of the propagation distance. In the model, the factors related to the material nonlinear parameter and the magnitude of the incident second harmonic component are unknown and these are determined by finding a value that best matches the experimental data according to the change in the propagation distance; compensation for the incident second harmonic component is then achieved. The case where the phase of the second harmonic wave due to material nonlinearity is opposite to that of the fundamental wave is also considered. To verify the validity of the proposed method, fused silica and aluminum alloy Al6061-T6 specimens with different thicknesses corresponding to the propagation distance are tested. The experimental results show that the nonlinear parameters changed significantly according to the propagation distance before compensation but were very stable after compensation. Additionally, the average values of the nonlinear parameter are 11.04 in the fused silica, which is within the literature value range (10.1 to 12.4), and that for the Al6061-T6 is 6.59, which is close to the literature value range (4.5 to 6.12).Gravel is used in railway infrastructure to reduce environmental impacts and noise, but gravel on tracks must be replaced continuously because it deforms due to wear and weathering. selleck chemicals It is therefore necessary to review the entire railroad life cycle. Our team has previously demonstrated a higher serum level of Gal-1 in patients with differentiated thyroid cancers versus healthy patients, and explored, by a knockdown strategy, the effect of Gal-1 silencing on cell proliferation and invasion in vitro, and on tumor and metastasis development in vivo. OTX008 is a calixarene derivative designed to bind the Gal-1 amphipathic β-sheet conformation and has previously demonstrated anti-proliferative and anti-invasive properties in several cancer cell lines including colon, breast, head and neck, and prostate cancer lines. In the current work, the impacts of OTX008 were evaluated in six thyroid cancer cell lines, and significant inhibitions of proliferation, migration, and invasion were observed in all lines expressing high Gal-1 levels.
