Results of the disc diffusion assay showed a stronger antibacterial effect of ZnO NPs compared with Se NPs. The bacterial susceptibility to the antibacterial films was as follows Listeria monocytogenes > Escherichia coli > Staphylococcus aureus > Pseudomonas fluorescens. The G/CNF films incorporated with Se nanoparticles possessed the higher property of scavenging free radicals in comparison films containing ZnO nanoparticles. In conclusion, gelatin-based edible films containing CNFs, ZnO NPs, and Se NPs can be used in the development of active food packaging products.Compression-decompression surface pressure (π)-specific molecular area (A) isotherm cycle of human serum albumin (HSA) monolayer is performed on water surface at four different subphase pH conditions, i.e., below and above the isoelectric point (pI ≈ 4.7) of HSA molecule. For all pH conditions, the decompression curve nearly follows the compression curve, however, at pH ≈ 5.0, hysteresis is observed at higher surface pressure. Out-of-plane structures and in-plane morphologies obtained from the X-ray reflectivity and AFM studies show that only the film thickness variation takes place with the change in surface pressure, which is also evidenced from the BAM images. With increase in surface pressure, the oblate-shaped HSA molecules start tilting making an angle with the water surface and as the monolayer is decompressed the molecules regain their initial untilted monomolecular configuration. Depending upon the subphase pH and local surface charge of the specific protein molecule, electrostatic repulsive interaction dominates over the van der Waals attraction and as a result decompression curve follows the compression curve as the molecules repel each other, however, closer to the isoelectric point as strength of the interactions reverses, a hysteresis is obtained at higher surface pressure and accordingly monolayer behaviour modifies on the water surface.Here, we report an analysis method for determining PHA (polyhydroxyalkanoates) contents and their monomer composition in microbial cells based on pyrolysis gas chromatography combined with mass spectrometry (Py-GC/MS). Various kinds of microbial cells accumulating different PHA contents and monomer compositions were prepared through the cultivation of Ralstonia eutropha and recombinant Escherichia coli. Py-GC/MS could analyse these samples in a short time without complicated pretreatment steps. Characteristic peaks such as 2-butenoic acid, 2-pentenoic acid, and hexadecanoic acid regarding PHA compositions and cell components were identified. Considering constituents of cells and ratios of peak areas of dehydrated monomers to hexadecanoic acid, a simple equation for estimation of PHA contents in microbial cells was derived. Also, monomer compositions of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) in R. eutropha could be successfully determined based on peak area of 2-butenoic acid and 2-pentenoic acid of Py-GC/MS, which are the corresponding species of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) in PHBV. Correlation of results between GC-FID and Py-GC/MS could be fitted very well. This method shows similar results for the samples obtained from same experimental conditions, allowing rapid and reliable analysis. Py-GC/MS can be a promising tool to rapidly screen PHA-positive strains based on polymer contents along with monomer compositions.To realize the application of xylan based film in food and drug packaging, the poor mechanical property and film-forming property of xylan based film must be overcome. Herein, a good oxygen barrier composite film with desired mechanical properties was prepared based on carboxymethly xylan (CMX), chitosan (CS), and graphene oxide (GO). see more The results of scanning electron microscope revealed the composite film had a dense and continuous structure, which will endow the composite film with excellent mechanical property. As expected, the composite film with the 0.5% mass fraction of GO exhibited best mechanical property, among which the tensile stress, tensile strain, and Young's modulus of the composite film reached 50.81 MPa, 47.61%, and 1.39 GPa, respectively. The oxygen barrier properties of the composite films significantly increased with the addition of graphene oxide due to the dense, stacked multilayer structure. In addition, these composite films exhibited good antibacterial properties. Therefore, these films show great promise in the field of food packaging and wound dressing due to their excellent mechanical, oxygen barrier and antibacterial properties.Influenza A virus (IAV) occasionally cross-species transmission among humans, swine and avian. The ectodomain of matrix protein 2 (M2e) is highly conserved in IAV, and multi-copy M2e from different species are usually displayed on the surface of nanoparticles to improve immunogenicity and constitute universal IAV nanovaccines. In our previous study, three M2e were inserted into the C-terminal of Cap protein of porcine circovirus type 2 (PCV2) to form a universal nanovaccine that provides protection against PCV2 and different subtypes of IAV. However, M2e adopts at least two converted conformations, and the intermolecular linker of M2e enhances the conformational instability, which limits the recognition by B cell receptors and production of high-level antibodies. Here, we report that the permutation of M2e affects effectiveness of nanovaccines. Three M2e derived from humans, swine and avian IAV were inserted into the C-terminal of Cap protein to form nanovaccines. Immunoprotective effects of different M2e arrangements were explored in mice. Results showed that the M2e closest to the surface of nanoparticle induced the most efficient protection against IAV derived from corresponding species. The results will contribute to develop more effective PCV2 and universal IAV bivalent nanovaccines for pigs, as well as species-specific universal IAV vaccines.
