At the same time, switches in adhesion molecule expression result in cellular rearrangements that define the morphology of the emerging neural tube. It is becoming increasingly clear that these two processes influence each other; adhesion molecules do not simply operate downstream of or in parallel with changes in cell identity but rather actively feed into cell fate decisions. Why are differentiation and adhesion so tightly linked? It is now over 60 years since Conrad Waddington noted the remarkable "Constancy of the Wild Type" (Waddington in Nature 183 1654-1655, 1959) yet we still do not fully understand the mechanisms that make development so reproducible. Conversely, we do not understand why directed differentiation of cells in a dish is sometimes unpredictable and difficult to control. It has long been suggested that cells make decisions as 'local cooperatives' rather than as individuals (Gurdon in Nature 336 772-774, 1988; Lander in Cell 144 955-969, 2011). Given that the cadherin family of adhesion molecules can simultaneously influence morphogenesis and signalling, it is tempting to speculate that they may help coordinate cell fate decisions between neighbouring cells in the embryo to ensure fidelity of patterning, and that the uncoupling of these processes in a culture dish might underlie some of the problems with controlling cell fate decisions ex-vivo. Here we review the expression and function of cadherins during early neural development and discuss how and why they might modulate signalling and differentiation as neural tissues are formed.The northeastern United States receives elevated mercury (Hg) deposition from United States and global emissions, making it critical to understand the fate of Hg in watersheds with a variety of aquatic habitats and land use types, such as the Finger Lakes region of New York State. Bats are valuable and important organisms to study chronic Hg exposure, because they are at risk of sublethal effects from elevated Hg exposure. The objectives of this study were to (1) determine total Hg (THg) and methylmercury (MeHg) concentrations in big brown bats (Eptesicus fuscus) of the Finger Lakes region; (2) assess whether morphometric, temporal, or spatial factors predict bat Hg concentrations; and (3) investigate the role of trophic position and diet represented by stable isotopes of carbon and nitrogen in explaining variations in bat Hg concentrations. We found comparable THg and MeHg concentrations to previous studies (THg range 1-45 ppm, MeHg range 0.5-38 ppm) in big brown bat fur collected throughout the Finger Lakes region. On average, MeHg made up 81% of THg in bat fur. Fifteen percent of our samples showed higher THg than a proposed toxicity threshold of 10 ppm. Together, dominant land cover and % wetland cover explained bat THg in the Finger Lakes. Trophic position (i.e., δ15N) was strongest in predicting bat THg in forests but was a weaker predictor of Hg bioaccumulation in bats from agricultural and urban areas. The range of Hg concentrations found in this study warrants further examination into the potential toxicological impacts of Hg to wildlife and the role of land use in Hg exposure to terrestrial organisms of the Finger Lakes.
Preventive home visits are part of communal programs for older citizens. The city of Hamburg promoted the pilot project "The Hamburg home visit for older citizens" in two city districts. The voluntary program is offered to persons on their 80th birthday, without comprehensive assessment and is performed by visitors with a background in healthcare or social work. The aim is to provide information, to identify need of help and to initiate contact with officials for support if wanted.
The structural and administrative prerequisites were defined together with officials of the city of Hamburg and the two city districts. The data safety concept, provision of addresses, birthday letter, information to be provided, recruitment, qualification and training of the visitors, timing and performance of visits, guiding how to communicate during the home visit, documentation and quarterly reports are described in detail.
From September 2018 to December 2019, 1636 (35%) out of 4716 persons contacted were visited by 59vil and operational prerequisites are considered. In 2020, they were offered to all districts of the city, i.e. to about 15,000 persons per year.Four independent mAb-producing CHO cell lines were grown in media supplemented with one of seven protein hydrolysates of animal and plant origin. This generated a 7x4 matrix of replicate cultures which was analysed for viable cell density and mAb productivity. In all cultures, a consistent growth rate was shown in batch culture up to 4 to 5 days. Differences between cultures appeared in the decline phase which was followed up to 7 days beyond the start of the cultures. There was a marginal but significant overall increase (x1.1) in the integral viable cell density (IVCD) in the presence of hydrolysate but a more substantial increase in the cell-specific mAb (qMab) productivity (x1.5). this website There were individual differences between hydrolysates in terms of enhancement of mAb productivity, the highest being a 166% increase of mAb titre (to 117 mg/L) in batch cultures of CHO-EG2 supplemented with UPcotton hydrolysate. The effect of one of the most active hydrolysates (HP7504) on antibody glycosylation was investigated. This showed no change in the predominant seven glycans produced but a significant increase in the galactosylation and sialylation of some but not all the antibodies. Overall, the animal hydrolysate, Primatone and two cotton-derived hydrolysates provided the most substantial benefit for enhanced productivity. The cotton-based hydrolysates can be viewed as valuable supplements for animal-derived component-free (ADCF) media and as a source for the investigation of chemically defined bioactive components. KEY POINTS • Protein hydrolysates enhanced both IVCD & qMab; the effect on qMab being consistently greater. • Cotton-based hydrolysates showed high bioactivity and potential for use in serum-free media. • Enhanced galactosylation and sialylation was shown for some of the Mabs tested.
