The Saccharomyces cerevisiae MBOAT O-acyltransferase Gup1 is involved in many processes, including cell wall and membrane composition and integrity, and acetic acid-induced cell death. Gup1 was previously shown to interact physically with the mitochondrial membrane VDAC (Voltage-Dependent Anion Channel) protein Por1 and the ammonium transceptor Mep2. By co-immunoprecipitation, the eisosome core component Pil1 was identified as a novel physical interaction partner of Gup1. The expression of PIL1 and Pil1 protein levels were found to be unaffected by GUP1 deletion. In ∆gup1 cells, Pil1 was distributed in dots (likely representing eisosomes) in the membrane, identically to wt cells. However, ∆gup1 cells presented 50% less Pil1-GFP dots/eisosomes, suggesting that Gup1 is important for eisosome formation. The two proteins also interact genetically in the maintenance of cell wall integrity, and during arsenite and acetic acid exposure. We show that Δgup1 Δpil1 cells take up more arsenite than wt and are extremely sensitive to arsenite and to acetic acid treatments. The latter causes a severe apoptotic wt-like cell death phenotype, epistatically reverting the ∆gup1 necrotic type of death. Gup1 and Pil1 are thus physically, genetically and functionally connected.Electronic health records (EHR) use is often considered a significant contributor to clinician burnout. Informatics researchers often measure clinical workload using EHR-derived audit logs and use it for quantifying the contribution of EHR use to clinician burnout. However, translating clinician workload measured using EHR-based audit logs into a meaningful burnout metric requires an alignment with the conceptual and theoretical principles of burnout. In this perspective, we describe a systems-oriented conceptual framework to achieve such an alignment and describe the pragmatic realization of this conceptual framework using 3 key dimensions standardizing the measurement of EHR-based clinical work activities, implementing complementary measurements, and using appropriate instruments to assess burnout and its downstream outcomes. We discuss how careful considerations of such dimensions can help in augmenting EHR-based audit logs to measure factors that contribute to burnout and for meaningfully assessing downstream patient safety outcomes.This letter discusses the limitations of the use of filters to enhance the accuracy of the extraction of parenthetic abbreviations from scholarly publications and proposes the usage of the parentheses level count algorithm to efficiently extract entities between parentheses from raw texts as well as of machine learning-based supervised classification techniques for the identification of biomedical abbreviations to significantly reduce the removal of acronyms including disallowed punctuations.Laeverin (LVRN) was first detected on the outer layer of the chorion laeve and migrating extravillous trophoblasts (EVTs). It is an enzyme that plays an important role in the placentation and pathophysiology of preeclampsia (PE). Previous studies have indicated that LVRN may be required for the invasion of human trophoblast cells. Paradoxically, LVRN was found to be highly expressed in the trophoblasts of PE patients with impaired invasive capacities. In this study, we detected the expression of LVRN in the placentas of PE patients (n=5) and normal term pregnancy women (n=5) as a control group by immunohistochemistry. LVRN was elevated in decidua (P=0.0083) and villi (P=0.0079) of PE patients. Next, LVRN was overexpressed via adeno-associated virus-mediated gene transfer in trophoblastic cell lines HTR8, Swan71, and JAR. Matrigel transwell assay and wound healing assay showed that overexpression of LVRN impeded the invasion of these three cell lines. Western blot analysis showed that LVRN overexpression caused downregulation of N-cadherin and vimentin and upregulation of E-cadherin, suggesting the inhibitory role of LVRN in epithelial-mesenchymal transition (EMT). Moreover, our data indicated that long noncoding RNA NONSTAT103348 (lnc10-7) was elevated in PE patients. Silencing lnc10-7 led to decreased LVRN expression. Taken together, although the basal level of LVRN may be crucial for cell invasion, overexpression of LVRN may abrogate the cell invasiveness, suggesting a multifaceted role of LVRN in the pathogenesis of PE.Executive functions refer to a set of higher-order cognitive processes involved in the control and organization of information to serve goal-directed behaviors. Skills in executive functioning are developed throughout childhood and adolescence and have been shown to be predictive of academic achievement. The coordination of these complex processes is critically dependent on brain maturation and connectivity, including key neurodevelopmental processes like myelination and synaptogenesis. Among other factors, research highlights the influential effect of nutrition and diet on these neurodevelopmental processes, which may impact executive function performance in healthy and deficient populations. This review considers the research to date on the role of key nutrients that have been identified for executive function development and their underlying neurophysiological processes in school-aged children.
Necrotizing coronary vasculitis (NCV) is a rare entity usually associated to myocarditis which incidence, cause, and response to therapy is unreported.
Among 1916 patients with biopsy-proven myocarditis, 30 had NCV. Endomyocardial samples were retrospectively investigated with immunohistochemistry for toll-like receptor 4 (TLR4) and real-time polymerase chain reaction (PCR) for viral genomes. Serum samples were processed for anti-heart autoantibodies (Abs), IL-1β, IL-6, IL-8, tumour necrosis factor (TNF)-α. Identification of an immunologic pathway (including virus-negativity, TLR4-, and Ab-positivity) was followed by immunosuppression. Myocarditis-NCV cohort was followed for 6 months with 2D-echo and/or cardiac magnetic resonance and compared with 60 Myocarditis patients and 30 controls. VX-809 mw Increase in left ventricular ejection fraction ≥10% was classified as response to therapy. Control endomyocardial biopsy followed the end of treatment. Twenty-six Myocarditis-NCV patients presented with heart failure; four with electrical instability.
