This study draws from the strategic memory regulation framework [Goldsmith, M., Koriat, A., & Weinberg-Eliezer, A. (2002). Strategic regulation of grain size memory reporting. Journal of Experimental Psychology General, 131(1), 73-95. https//doi.org/10.1037/0096-3445.131.1.73] to make predictions about how eyewitness memory reporting is affected by exposure to such reports. In Experiment 1, participants (n = 226) viewed a mock crime, were exposed to a fictitious news report about eyewitness memory accuracy (memory is accurate, memory is inaccurate, or a control condition), and then recalled the mock crime. Participants who read that eyewitness memory is inaccurate were less confident in their memory accuracy and reported less information about the mock crime compared to those in the other conditions. The specificity and accuracy of recall did not vary across conditions, however. In Experiment 2, participants (n = 2,491) watched a mock crime and were asked to identify the perpetrator from a simultaneous lineup. Participants who read that eyewitness memory is inaccurate evaluated their memory for the mock crime as relatively poorer but their lineup decisions did not differ compared to other participants. This suggests that news about eyewitness memory inaccuracy affects how people evaluate their memory capability, and differentially affects memory output depending on the memory task.Phytophthora spp. secrete vast arrays of effector molecules during infection to aid in host colonization. The crinkling and necrosis (CRN) protein family forms an extensive repertoire of candidate effectors that accumulate in the host nucleus to perturb processes required for immunity. Here, we show that CRN12_997 from Phytophthora capsici binds a TCP transcription factor, SlTCP14-2, to inhibit its immunity-associated activity against Phytophthora spp. Coimmunoprecipitation and bimolecular fluorescence complementation studies confirm a specific CRN12_997-SlTCP14-2 interaction in vivo. Coexpression of CRN12_997 specifically counteracts the TCP14-enhanced immunity phenotype, suggesting that CRN mediated perturbation of SlTCP14-2 function. We show that SlTCP14-2 associates with nuclear chromatin and that CRN12_997 diminishes SlTCP14-2 DNA binding. Collectively, our data support a model in which SlTCP14-2 associates with chromatin to enhance immunity. The interaction between CRN12_997 and SlTCP14-2 reduces DNA binding of the immune regulator. This is an open access article distributed under the CC BY-NC-ND 4.0 International license.The purpose of this study was to evaluate the effects of using the critical velocity (CV) concept to prescribe two separate high-intensity interval training (HIT) exercise programs aimed at enhancing CV and load carriage performance. 20 young adult participants (male = 15, female = 5) underwent a 4-week training period where they exercised 2 d wk-1. Participants were randomly assigned into two groups (1) HIT or (2) Load Carriage-HIT (LCHIT). Pre- and post-training assessments included running 3-minute All-Out Test (3MT) to determine critical velocity (CV) and distance prime (D') and two load carriage tasks (400 and 3200 m). There were significant increases in CV (p = 0.005) and velocity at V˙ O2max (vV˙ O2max) (p = 0.037) among the sample but not between training groups. Improvements were observed in 3200 m load carriage performance time (p less then 0.001) with a 9.8 and 5.4% decrease in the LCHIT and HIT groups, respectively. Practitioner summary Critical velocity has shown efficacy as a marker for performance in tactical populations. With the addition of load carriage, there is a reduction in the individual's CV. The CV-concept-prescribed exercises (HIT and LCHIT) 2 days per week for 4 weeks showed improvements in CV, vV˙ O2max and load carriage performance. The use of the CV concept provides a method to prescribe HIT to increase running and load carriage performances in tactical populations.Using DNA affinity chromatography we demonstrate that the S. aureus regulatory proteins MgrA, Rot, SarA, and SarS bind DNA baits derived from the promoter regions associated with the genes encoding aureolysin, ScpAB, SspABC, and SplA-F. These studies confirmed that multiple regulatory loci are involved in limiting protease production to a degree that impacts all of these phenotypes, but also demonstrate that sarA plays a predominant role in this regard. Using sarA mutants unable to produce individual proteases alone and in combination with each other, we also demonstrate that the increased production of aureolysin and ScpA is particularly important in defining the biofilm-deficient phenotype of LAC and UAMS-1 sarA mutants, while aureolysin alone plays a key role in defining the reduced accumulation of alpha toxin and overall cytotoxicity as assessed using both osteoblasts and osteoclasts.
