In recent years, there has been a growing awareness of the complex dependence of herbaceous plant diversity on forest structure and management. However, among the studies presented so far, those in which the chronosequence (approach based on the assumption of space-for-time substitution) was used, dominate. On the other hand, it is rare to find results based on long-term research on permanent or semi-permanent sampling plots. The aim of this study was to recognize the changes in the vegetation composition and dynamics of various indices of herbaceous plant diversity over 40 years of forest development, and their dependence on forest structure and management. Here we analyzed the temporal dynamics of herbaceous plant diversity in Carpathian fertile beech forests, based on datasets recorded on semi-permanent plots in three censuses (the 1970s, 2000s and 2010s). We checked the temporal changes in alpha, beta, and gamma diversity. Analyses of the plant diversity were performed on the background of changes in forey and to properly assess the state of the forest biodiversity, their conservation status, or conservation action plans. In addition, we need far more data from long-term observations to fully understand the possible relationship patterns between the factors controlling the forest structure and plant diversity.(1) Backgrounds Several studies have shown that the level of 25-hydroxyvitamin D (25(OH)D) could affect urticaria. The association of Vitamin D (VitD) with urticaria has not been well established. (2) Methods The up-to-date meta-analysis was performed to synthesize the new findings. We performed a systematic search in PubMed, EMBASE, Web of Science, and Cochrane Database. We included the observational studies with the comparisons of 25(OH)D between urticarial populations and controls and clinical studies with the clinical severity of urticaria records. (3) Results A meta-analysis of seventeen studies of urticaria group vs. controls revealed a mean difference of -9.35 ng/mL (95% CI -12.27 to -6.44). There was also an association of urticaria with VitD deficiency. In the subgroup analysis of age and disease type, significant effects of 25(OH)D were found among adult and chronic urticarial populations. Six VitD supplementation trials showed a significant reduction in clinical urticarial score on intervention with VitD with the standard mean difference of -3.63 and -1.54 among randomized control studies and repeated measure trials, respectively. (4) Conclusions The urticarial population, especially the adult chronic urticarial patients, may be associated with a high risk for lower serum 25(OH)D. VitD supplementations could result in a reduction of urticarial clinical symptoms.The RNA-binding protein Musashi-1 (MSI1) promotes stemness during development and cancer. Due to its specific expression pattern and diverse functions, MSI1 represents an interesting target for cancer therapy in the future. In order to start the process of data alignment, we assessed the consistency of the BCR-ABL1 quantification results of the analysis of 16 RNA samples at different levels of disease. The results were obtained by two different laboratories that relied on The Qx100/Qx200 Droplet Digital PCR System (Bio-Rad) and Quant Studio 3D dPCR System (Thermofisher) platforms. We assessed the compatibility between the estimated values by linear regression, Bland-Altman bias-plot, and Mann-Whitney nonparametric test. The results confirmed the compatibility of the measures, allowing us tocompute an 'alignment factor' (AF), equal to 1.41, which was further validated by a different series of experiments. We conclude that the performed measurements by the two laboratories are comparable, and also equalized through the introduction of an alignment factor.A single ulcerative colitis (UC) is a chronic inflammatory bowel disease (IBD) that causes inflammation of the colonic mucosa at the distal colon and rectum. The mainstay therapy involves anti-inflammatory immunosuppression based on the disease location and severity. The disadvantages of using systemic corticosteroids for UC treatment is the amplified risk of malignancies and infections. Therefore, topical treatments are safer as they have fewer systemic side effects due to less systemic exposure. In this context, pH sensitive and enzymatically triggered hydrogel of pectin (PC) and polyacrylamide (PAM) has been developed to facilitate colon-targeted delivery of budesonide (BUD) for the treatment of UC. The hydrogels were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), swelling ratio, and drug release. FT-IR spectroscopy confirmed the grafting as well loading of BUD in hydrogel. XRD showed the amorphous nature of hydrogel and increment in crystallinity after drug loading. On the other hand, SEM showed that the hydrogels exhibited a highly porous morphology, which is suitable for drug loading and also demonstrated a pH-responsive swelling behaviour, with decreased swelling in acidic media. The in-vitro release of BUD from the hydrogel exhibited a sustained release behaviour with non-ficken diffusion mechanism. The model that fitted best for BUD released was the Higuchi kinetic model. It was concluded that enzyme/pH dual-sensitive hydrogels are an effective colon-targeted delivery system for UC.
