The method was compared with commercially sensitive porcine DNA detection kits (Biotecon and R-Biopharm) which are commonly used in food control laboratories for Halal and kosher authentication. The results indicated that a newly developed method called TübiGel was at least 10 times more sensitive for porcine DNA detection. The TübiGel method was found to have a detection limit of 0.01% porcine gelatin, whilst the Biotecon method had 0.1% and R-Biopharm method detected >5% porcine gelatin. The forensic DNA isolation approach of the TübiGel method was found to be a critical step. In addition, real-time PCR of TübiGel method was also found to detect porcine DNA better than real-time PCR of commercial kits.This study examined the influence of perfectionistic strivings and perfectionistic concerns on athlete burnout and two key indicators of overtraining syndrome (training distress and subjectively perceived sports performance) using cross-sectional (N = 228), short-term (a 3-month interval, N = 93) and long-term (a 1-year interval, N = 83) longitudinal designs on a sample of adolescent athletes. In the cross-sectional analyses, sequential regressions revealed that perfectionism was a significant predictor of athlete burnout and both indicators of overtraining. In the three-month longitudinal perspective, both dimensions of perfectionism (strivings and concerns) contributed to the prediction of change in burnout and sports performance, but not training distress. When the one-year longitudinal relationships were regarded, only perfectionistic strivings significantly predicted decrease in burnout, and, for sports performance, the predictive power of both dimensions of perfectionism was even more pronounced when compared to the three-month longitudinal data.Objective The present study investigated the role of the two theoretically derived mediators in the treatment of Prolonged Grief Disorder (PGD). Mediators were changes in avoidance and maladaptive cognitions. An additional hypothesis tested whether these candidate mediators are specific to CBT-based Complicated Grief Treatment (CGT) compared to Interpersonal Therapy (IPT). Method We performed secondary analyses with assessment completers (n = 131) from a randomized-controlled trial with older adults with PGD. Patients received 16 sessions of CGT or IPT. Outcomes were treatment response and reductions in grief symptoms and grief-related related impairment. Results Reductions in avoidance between baseline and week 16 mediated reductions in grief symptoms and grief-related impairment. Reductions in maladaptive grief-related cognitions over the same period mediated treatment response, reductions in grief symptoms and grief-related impairment. There were no significant treatment-mediator interactions. We could not establish that mediators changed before the outcomes. Conclusion Results are consistent with theoretical models of PGD, including the CGT treatment model. GGTI 298 clinical trial Despite different therapeutic procedures, we found no significant interaction effect, but CGT produced larger effects. Future research needs to establish a timeline of change through the use of multiple measurements of mediators and outcomes.Trial registration ClinicalTrials.gov identifier NCT01244295.This study aimed to replicate Muir and Moray that demonstrated operators' trust in automated machines developing from faith, then dependability, and lastly predictability. Following the procedure of Muir and Moray, we asked undergraduate participants to complete a training program in a simulated pasteuriser plant and an experimental program including various errors in the pasteuriser. Results showed that the best predictor of overall trust was not faith but dependability, and that dependability consistently governed trust throughout the interaction with the pasteuriser. Thus, the obtained data patterns were inconsistent with those reported in Muir and Moray. We observed that operators in the current study used automatic control more frequently than manual control to successfully produce performance scores contrary to the operators in Muir and Moray. The results imply that dependability is a critical predictor of human-machine trust, which automation designer may focus on. More extensive future research using more modern automated technologies is necessary for understanding what factors control human-autonomy trust in modern ages. Practitioner Summary The results suggest that dependability is a key factor that shapes human-machine trust across the time course of the trust development. This replication study suggests a new perspective for designing effective human-machine systems for untrained users who do not go through extensive training programs on automated systems.The understanding of mechanism during conversion from sepsis to sepsis-related ARDS remains limited. In this study, we collected gene expression matrix from the Gene Expression Omnibus (GEO) database and constructed networks using weighted gene co-expression network analysis (WGCNA) to identify the consensus and opposite modules between sepsis and sepsis-induced ARDS and obtained 27 consensus modules associated with sepsis and sepsis-related ARDS, including one model (160 genes) with opposite correlation and 1 sepsis-ARDS specific model with 34 genes. Differentially expressed genes analysis, functional enrichment and protein-protein interactions analyses of candidate genes were performed; 15 of these genes showed different expressions in sepsis-induced ARDS patients, compared with sepsis patients; genes were enriched in processes associated with ribosome, tissue mechanics and extracellular matrix. Feature selection analysis revealed that three genes, TLCD4, PRSS30P, and ZNF493, showed moderate performance in identifying sepsis-induced ARDS from sepsis.
