Somatic mutations can have critical prognostic and therapeutic implications for cancer patients. Although targeted methods are often used to assay specific cancer driver mutations, high throughput sequencing is frequently applied to discover novel driver mutations and to determine the status of less-frequent driver mutations. The task of recovering somatic mutations from these data is nontrivial as somatic mutations must be distinguished from germline variants, sequencing errors, and other artefacts. Consequently, bioinformatics pipelines for recovery of somatic mutations from high throughput sequencing typically involve a large number of analytical choices in the form of quality filters.
We present vcfView, an interactive tool designed to support the evaluation of somatic mutation calls from cancer sequencing data. The tool takes as input a single variant call format (VCF) file and enables researchers to explore the impacts of analytical choices on the mutant allele frequency spectrum, on mutational signatures and on annotated somatic variants in genes of interest. The purpose of this study was to develop and test a family-based curriculum with a parenting focus aimed at influencing child obesity risk. Qualitative data in the pilot implementation (cohort 1, n = 6 families with elementary-aged children) suggested parent value for positive messages, hands-on learning, and group sharing. However, Family Nutrition and Physical Activity (FNPA) scores for cohorts 2-6 (n = 18 families) did not change significantly from pre- to post-program. Future implementations will consider more robust ways of assessing change and recruiting participants at various stages of change.
Flexible pes planovalgus in children may be corrected with a calcaneal lengthening osteotomy (CLO).However, CLO surgery may displace the distal calcaneal fragment dorsally and affect the motion in the calcaneal-cuboid joint (CCJ). We used radiostereometric analysis (RSA) to evaluate CCJ motion and CLO stability in children one year after CLO.
Ten patients (10 feet) with symptomatic flexible pes planovalgus were investigated one year after CLO. Mean age was 11.5 years (range 8.2-14.2). Standardised RSA measurements of the foot were obtained one year after surgery without and with weight-bearing (single leg stance). Tantalum markers inserted during surgery, were used to described the CCJ motion as cuboid bone motion with respect to the distal calcaneus, and the CLO stability as distal calcaneal migration with respect to the proximal calcaneus.
One year after surgery the CLO was stable. The motion in the CCJ upon full weight-bearing was mean -1.04mm (CI95% -1.40; -0.67) joint distraction, mean 2.27mm (CI95% 1.57; 2.96) cuboid dorsal translation, mean -1.94mm (CI 95% -2.68; -1.20) cuboid medial translation, and mean 7.43° (CI 95% 3.97; 10.88) adduction.

This study sheds light on this issue by analysing membership influence in a representative sample of approximately 2000 CSOs from five European countries and the European level. As members generally have a strong influence on CSOs' policy positions, our analysis demonstrates that the pessimistic tone in much contemporary scholarly work is largely unwarranted. On the contrary, hiring professionals does not invariably decrease membership influence and can, when members are closely engaged in advocacy work, even facilitate it.This paper aims at giving an account of the philosophy of norms of Georges Canguilhem in the framework of his philosophical vitalism. Canguilhem's perspective relocates human activities within a vitalistic conception of life, which redefines the meaning of human reason by putting it in relation to values and norms.
