L. monocytogenes causes relatively few outbreaks linked to whole fresh produce but triggers recalls each year in the US. There are limited data on the influence of wet vs. dry methods on pathogen growth on whole produce. A cocktail of five L. monocytogenes strains that included clinical, food, or environmental isolates associated with foodborne outbreaks and recalls was used. Cultures were combined to target a final wet inoculum concentration of 4-5 log CFU/mL. The dry inoculum was prepared by mixing wet inoculum with 100 g of sterile sand and drying for 24 h. Produce investigated belonged to major commodity families Ericaceae (blackberry, raspberry, and blueberry), Rutaceae (lemon and mandarin orange), Roseaceae (sweet cherry), Solanaceae (tomato), Brassaceae (cauliflower and broccoli) and Apiaceae (carrot). Intact, whole inoculated fruit and vegetable commodities were incubated at 2, 12, 22 and 35±2°C. Commodities were sampled for up to 28 days, and the experiment was replicated 6 times. The average maximum growth increase was obtained by measuring the maximum absolute increase for each replicate within a specific commodity, temperature, and inoculation method. Data for each commodity, replicate and temperature was used to create primary growth or survival models, describing the lag phase and growth or shoulder and decline as a function of time. Use of a liquid inoculum (vs. dry inoculum) resulted in markedly increased L. monocytogenes growth rate and growth magnitude on whole produce surfaces. This difference was highly influenced by temperature with a greater effect seen with more commodities at higher temperatures (22 and 35°C), versus lower temperatures (2 and 12 °C). These findings need to be explored for other commodities and pathogens. The degree to which wet or dry inoculation techniques more realistically mimic contamination conditions throughout the supply chain (e.g., production, harvest, post-harvest, transportation, or retail) should be investigated.The human noroviruses are major causes of nonbacterial gastroenteritis, and are transmitted by both food and water, as well as via a person-to-person route. Asymptomatic norovirus infection of food handlers may play a role in transmission. The outbreak of noroviruses infections has been recognized in PyeongChang Winter Olympics, starting with security staffs from February 3, 2018. The Ministry of Food and Drug Safety in Korea conducted a noroviruses surveillance from asymptomatic food handlers of food catering facilities related to the Olympics to prevent the spread of the noroviruses. A total of 707 rectal swab samples from food handlers were collected and examined for norovirus using real-time RT-PCR and conventional RT-PCR. Five of 707 detected noroviruses. Genotypes of the norovirus-positive samples were determined with sequencing analysis. Identified genotypes of norovirus in asymptomatic food handlers included GI.3, GII.4, and GII.17. The GII.17 was prevalent among the genotypes, accounting for 3 of 5 detections. Food handlers with noroviruses detected in rectal swab are excluded from cooking and all foods handled by infected food handlers were discarded. Surveillance of norovirus infection for food handlers contributed to preventing noroviruses spread.In Nepal, the number of diarrhoea hospitalizations in all ages is seriously high. According to the World Health Organization, diarrheal diseases can be substantially prevented through safe drinking water sources. In the Kathmandu Valley, because of the shortage of piped water, local residents use alternative water sources, such as groundwater, jars and tanker water. However, these alternative water sources can be contaminated. This study aimed to clarify the relationship between diarrhoea risk and the combinations of drinking water sources. A survey using multiple questionnaires on diarrhoea occurrence, water sources and water treatment was conducted three time between 2015 and 2016. The odds ratios (ORs) of developing diarrhoea were significantly high for drinking jar (OR 6.1) and tanker water (OR 8.4) compared with not drinking. The combined drinking of jar and tanker water obtained the 1 log higher OR compared with drinking only piped water. Conversely, drinking groundwater had a low OR, implying that the residents refrained from drinking polluted groundwater. Navitoclax In conclusion, diarrhoea occurrence was related not only to the level of water contamination, but also to a behavioural factor, i.e. people's careful management of the choice of multiple water sources.Plant and animal intracellular nucleotide-binding and leucine-rich repeat (NLR) receptors play important roles in sensing pathogens and activating defense signaling. However, the molecular mechanisms underlying the activation of host defense signaling by NLR proteins remain largely unknown. Many previous studies have determined that the coil-coil (CC) or Toll and interleukin-1 receptor/resistance protein (TIR) domain of NLR proteins and their dimerization/oligomerization are critical for activating downstream defense signaling. In this study, we demonstrated that the nucleotide-binding (NB) domain of the tomato Sw-5b NLR alone can activate downstream defense signaling, leading to elicitor-independent cell death. Sw-5b NB domains can self-associate, and this self-association is crucial for activating cell death signaling. The self-association was strongly compromised after the introduction of a K568R mutation into the P-loop of the NB domain. Consequently, the NB K568R mutant induced cell death very weakly. The NB CΔ20 mutant lacking the C-terminal 20 amino acids can self-associate but cannot activate cell death signaling. The NB CΔ20 mutant also interfered with wild-type NB domain self-association, leading to compromised cell death induction. By contrast, the NB K568R mutant did not interfere with wild-type NB domain self-association and its ability to induce cell death. Structural modeling of Sw-5b suggests that NB domains associate with one another and likely participate in oligomerization. As Sw-5b-triggered cell death is dependent on helper NLR proteins, we propose that the Sw-5b NB domain acts as a nucleation point for the assembly of an oligomeric resistosome, probably by recruiting downstream helper partners, to trigger defense signaling.
