To identify the molecular mechanisms involved, we examined the impact and the associated signaling pathways of DHA-PL and EPA-PL on TNF-α-induced lipolysis in 3T3-L1 adipocytes. The present results revealed that DHA-PL and EPA-PL inhibited the TNF-α-induced increase of glycerol release and protected lipid droplets. In addition, DHA-PL and EPA-PL increased DHA and EPA contents in the phospholipid fraction of adipocytes, respectively. Moreover, DHA-PL and EPA-PL enhanced sirtuin 1 (SIRT1) deacetylase activity and its protein expression. By activating SIRT1, DHA-PL and EPA-PL upregulated the G0/G1 switch gene 2 protein level to inhibit adipose triglyceride lipase activity, activate AMP-activated protein kinase to reverse the downregulation of perilipin expression and phosphorylation of hormone-sensitive lipase (HSL) at Ser565 and prevent the phosphorylation of HSL at Ser660. Furthermore, DHA-PL and EPA-PL improved glucose uptake and glucose transporter type 4 translocation to the plasma membrane in TNF-α-treated adipocytes. Thus, it was concluded that DHA-PL and EPA-PL inhibit TNF-α-induced lipolysis in 3T3-L1 adipocytes by activating the SIRT1 pathways.In the present study we investigated the effect of cellular integrity on microbial utilization of proteins and carbohydrates by gut microbiota. Cotyledon cells from red kidney beans with different levels of structural integrity were fermented in-vitro by microbial communities previously adapted to the conditions of ascending, transverse and descending colon. The effect of bacterial adaptation to substrate was also assessed by using microbiota exposed to a diet rich in bean cells. Microscopy analyses indicate that cell integrity was maintained during fermentation. The amount of gas generated and the rate of total gas production was higher in broken cells compared to intact cells which suggest a faster and more extensive utilization of nutrients when cell wall is broken. A significantly higher butyric and propionic acid level was detected in broken cells at the end of the fermentation. Moreover, adapted bacterial communities were more efficient in fermenting bean cells where higher amounts of butyrate were produced in all colon regions independently of sample integrity. Bacterial communities of the distal colon appeared to be the most efficient in carbohydrate and protein fermentation as witnessed by the higher levels of gas, and short chain fatty acids. It was also found that cell integrity and adaptation to bean cells modulate the hierarchy of nutrient utilization, with non-starch polysaccharides preferred over starch and proteins by microbiota exposed to bean cells. Our results demonstrated that structural aspects of foods, such as cell integrity in plant tissues, may modulate nutrients utilization by gut microbiota.The natural structure of whole food plays an important role in the physiological impact of bioactive compounds present within the food, also known as the "matrix effect". However, most previous studies have compared only the long-term bioavailability of fish versus fish oil and have used commercial sources of fish oil. The present study aimed to investigate whether fish (salmon) matrix influences the transit of LCn-3PUFAs during in vitro digestion and affects bioavailability in healthy females. Meals containing intact salmon (intact structure), minced salmon (some structure) and defatted salmon + oil (no structure) with identical macronutrient compositions were developed. The geometry of the Pd monolayer was optimized on the (100) plane of γ-Al2O3 at fixed unit cell parameters defined by the oxide. Simulation of the deposition of a whole Pd monolayer in the flat Pd(100) form cut from the bulk led to the formation of a slightly distorted Pd(111) monolayer. The subsequent chemisorption or dissociation of CH4 or H2O on the Pd(111) layer resulted in a new hybrid Pd(100)/(111) structure containing alternating elements of (100) and (111) planes (the parallel bands of squares and triangles), which are similar for both CH4 and H2O reactions, and two isolated Pd mono-vacancies, respectively. The hybrid Pd(100)/(111) layer without chemisorbed species was found to be more stable than the initial distorted Pd(111) layer. The catalytic capabilities of these monolayer structures were investigated for the dissociation of methane and the water-gas shift reaction (WGSR) due to the lower predicted activation barriers for CH4, H2O, and O2 dissociation on the hybrid Pd(100)/(111) layer compared to that on the pure (bulk) Pd(100) surface. find more Moreover, the exothermic heats of these reactions were calculated to be moderate instead of endothermic heats on the Pd(100) or Pd(111) surfaces. The insertion of guest molecules and their conformation were inferred from Raman and FTIR spectra and from single-crystal X-ray diffraction and confronted with computational simulation. The magnetic and photomagnetic behaviors of the framework are significantly altered by the different guest molecules and different conformations. On the other hand, photoisomerization of the guest molecules becomes strongly hindered by the framework.
