A greater molecular understanding of the bacterial targets of antibiotics has the potential to reveal novel therapeutic strategies or identify agents against antibiotic-resistant pathogens.The high-strength-to-weight ratio and corrosion resistance properties of glass-fiber-reinforced polymer (GFRP) composites makes them potentially well-suited for application in ship structures, bridges and off-shore oil platforms. These structures are often formed by stiffened plates and are subjected to axial load and out-of-plane load. Cutouts and openings are provided in the plates for access and maintenance. The main objective of this study was to examine the buckling behavior of GFRP-stiffened composite plates with square cutouts under a combination of axial and out-of-plane load up to failure. Four blade-stiffened composite plates without a cutout and four with square cutout were fabricated with stiffeners as a continuous layup of the flange plate using glass fiber and epoxy resin. The initial geometric imperfections were measured, and plate imperfections (Δx), stiffener imperfections (Δsy) and overall imperfections (Δsx) were calculated from the measurements. All fabricated-stiffened composite plates were tested up to failure. The finite element model was developed in ANSYS software and validated with the experimental results. It was observed that GFRP-stiffened composite plates failed by stiffener compression/stiffener tension mode of failure. The presence of out-of-plane loads and cutouts reduced the axial load carrying capacity of the stiffened composite plates.Cleft lip and palate are common congenital pathologies that affect the human population worldwide. The formation of cleft lip is associated with multiple genes and their coded proteins, which regulate the development of craniofacial region, but the exact role of these factors is not always clear. CFSE in vivo The use of morphological studies for evaluation of human cleft-affected tissue has been limited because of insufficiency of available pathological material. The aim of this study was to detect and compare the immunohistochemical expression of cleft candidate gene coded proteins (DLX4, MSX2, HOXB3, SHH, PAX7, SOX3, WNT3A, and FOXE1) in the non-syndromic unilateral cleft lip patient tissue and control group tissue. A semiquantitative counting method was used to evaluate the tissue in biotin-streptavidin-stained slides. Statistically significant differences between the patient and control groups were found for the number of immunoreactive structures for SHH (p = 0.019) and FOXE1 (p = 0.011) in the connective tissue and SOX3 (p = 0.012) in the epithelium. Multiple statistically significant very strong and strong correlations were found between the immunoreactives in cleft-affected tissue. These significant differences and various correlations indicate that multiple morphopathogenetic pathways are possibly involved in unilateral cleft lip pathogenesis. Therefore, we further discuss these possible interactions.This study examined the longitudinal association between changes in sugar-sweetened and/or caffeinated beverage consumption and smoking/vaping behaviour among Canadian adolescents. Using longitudinal data from the COMPASS study (2015/16 to 2017/18), four models were developed to investigate whether beverage consumption explained variability in smoking and vaping behaviour in adolescence (1) smoking initiation, (2) vaping initiation, (3) current smoking status, and (4) current vaping status. Models were adjusted for demographic factors. Multinomial logit models were used for model 1, 2, and 3. A binary logistic regression model was used for model 4. An association between change in frequency of beverage consumption and smoking/vaping behaviour was identified in all models. A one-day increase in beverage consumption was associated with smoking initiation (OR = 1.38, 95% CI 1.25, 1.51), vaping initiation (OR = 1.23, 95% CI 1.14, 1.32), identifying as a current smoker (OR = 1.17, 95% CI 1.01, 1.35), and currently vaping (OR = 1.08, 95% CI 1.04, 1.11). Change in high-energy drink consumption was the best predictor of smoking behaviours and vaping initiation but not current vaping status. Given the health consequences of smoking and vaping and their association with high-energy drink and coffee consumption, policy initiatives to prevent smoking/vaping initiation, and to limit youth access to these beverages, warrant consideration.Non-small cell lung cancer (NSCLC) is a lethal non-immunogenic malignancy and proto-oncogene ROS-1 tyrosine kinase is one of its clinically relevant oncogenic markers. The ROS-1 inhibitor, crizotinib, demonstrated resistance due to the Gly2032Arg mutation. To curtail this resistance, researchers developed lorlatinib against the mutated kinase. In the present study, a receptor-ligand pharmacophore model exploiting the key features of lorlatinib binding with ROS-1 was exploited to identify inhibitors against the wild-type (WT) and the mutant (MT) kinase domain. The developed model was utilized to virtually screen the TimTec flavonoids database and the retrieved drug-like hits were subjected for docking with the WT and MT ROS-1 kinase. A total of 10 flavonoids displayed higher docking scores than lorlatinib. Subsequent molecular dynamics simulations of the acquired flavonoids with WT and MT ROS-1 revealed no steric clashes with the Arg2032 (MT ROS-1). The binding free energy calculations computed via molecular mechanics/Poisson-Boltzmann surface area (MM/PBSA) demonstrated one flavonoid (Hit) with better energy than lorlatinib in binding with WT and MT ROS-1. The Hit compound was observed to bind in the ROS-1 selectivity pocket comprised of residues from the β-3 sheet and DFG-motif. The identified Hit from this investigation could act as a potent WT and MT ROS-1 inhibitor.The aim of this descriptive study was to investigate the impact of body image, insight, and mental health confidence on medication adherence among young adult women with mental disorders. Data collection occurred from August to September 2018. The study participants were 147 young adult women aged 19 to 45 with mental disorders who were psychiatry outpatients either getting treatment in general hospitals located in Seoul and Gyeonggi Province or receiving rehabilitation therapy through local mental health welfare centers in Korea, and agreed to participate in the study. The measurement tools used were the Body Image Scale; the Scale to Assess Unawareness of Mental Disorder, Korean short-form version; the Korean version of the Mental Health Confidence Scale; and the Korean version of the Medication Compliance Scale. The collected data were analyzed using descriptive statistics, t-test, analysis of variance, Pearson correlation analysis, and multiple regression analysis in SPSS/WIN 25.0 (IBM Corp., Armonk, NY, USA).
