Excitingly, despite the fact that NanoLuc/fluorofurimazine emits mostly blue light, we prove that cells trapped in mice lungs vasculature could be visualised via the NanoLuc/fluorofurimazine pair and compare the results to the AkaLuc/AkaLumine system. Therefore, among the tested analogues, fluorofurimazine enables higher substrate loading and improved optical imaging sensitivity in small animals, upgrading the use of NanoLuc derived bioluminescent systems for deep tissue imaging.
Glioblastoma multiforme (GBM) is the most frequent, lethal and aggressive tumour of the central nervous system in adults. The discovery of novel anti-GBM agents based on the isocitrate dehydrogenase (IDH) mutant phenotypes and classifications have attracted comprehensive attention.
Diterpenoids are a class of naturally occurring 20-carbon isoprenoid compounds, and have previously been shown to possess high cytotoxicity for a variety of human tumours in many scientific reports. In the present study, 31 cassane diterpenoids of four types, namely, butanolide lactone cassane diterpenoids (I) (1-10), tricyclic cassane diterpenoids (II) (11-15), polyoxybutanolide lactone cassane diterpenoids (III) (16-23), and fused furan ring cassane diterpenoids (IV) (24-31), were tested for their anti-glioblastoma activity and mechanism underlying based on IDH1 mutant phenotypes of primary GBM cell cultures and human oligodendroglioma (HOG) cell lines.
We confirmed that tricyclic-type (II) and compound 13 (Caesalpin A, CSAg GLS.
Therefore, the present results demonstrated that compared with other diterpenoids, tricyclic-type diterpenoids could be a targeted drug candidate for the treatment of secondary IDH1 mutant type glioblastoma through negatively regulating GLS.Smart nano-micro platforms have been extensively applied for diverse biomedical applications, mostly focusing on cancer therapy. In comparison with conventional nanotechnology, the smart nano-micro matrix can exhibit specific response to exogenous or endogenous triggers, and thus can achieve multiple functions e.g. site-specific drug delivery, bio-imaging and detection of bio-molecules. These intriguing techniques have expanded into ophthalmology in recent years, yet few works have been summarized in this field. In this work, we provide the state-of-the-art of diverse nano-micro platforms based on both the conventional materials (e.g. natural or synthetic polymers, lipid nanomaterials, metal and metal oxide nanoparticles) and emerging nanomaterials (e.g. up-conversion nanoparticles, quantum dots and carbon materials) in ophthalmology, with some smart nano/micro platformers highlighted. The common ocular diseases studied in the field of nano-micro systems are firstly introduced, and their therapeutic method and the related drawback in clinic treatment are presented. The recent progress of different materials for diverse ocular applications is then demonstrated, with the representative nano- and micro-systems highlighted in detail. At last, an in-depth discussion on the clinical translation challenges faced in this field and the future direction are provided. This review would allow the researchers to design more smart nanomedicines in a more rational manner for specific ophthalmology applications. The behavior of arsenic (As) in paddy soil is of great interest because rice is an efficient As accumulator, which can result in potential health risks to humans. find more In this study, we investigated the distribution and translocation of As speciation within the soil-rice system in the presence of Fenton solution (100 μM H2O2 and 100 μM Fe2+). The results showed that the iron plaque concentrations were enhanced either by the addition of Fenton solution or under flooded condition. In addition, total As, As5+, and As3+ concentrations were lower in rice tissues treated with Fenton solution than those without Fenton solution treatments. Therefore the addition of Fenton solution can reduce As speciation accumulation in rice tissue. This study revealed the function of Fenton solution on the fate of As speciation distribution within soil-rice system and the link between iron plaque and As accumulation in rice tissue under flooded and non-flooded conditions.Organophosphorus pesticides are the most used pesticides in the United States. Most organophosphorus pesticides are composed of a phosphate (or phosphorothioate or phosphorodithioate) moiety and a variable organic group. Organophosphorus pesticides are scrutinized by regulatory bodies and agencies because of their toxicity or suspected carcinogenicity. Upon exposure, organophosphorus pesticides and their metabolites eliminate in urine; these urinary biomarkers are useful to evaluate human exposure. We developed a method using stable isotope dilution, ion chromatography tandem mass spectrometry for quantification in urine of 6 O,O-dialkylphosphates, metabolites of organophosphorus insecticides, and glyphosate, the most used herbicide in the United States. With simple and minimal sample preparation, the analytical method is selective and sensitive (limits of detection are 0.2-0.8 μg/L), accurate (>85%) and precise (relative standard deviation less then 20%), depending on the analyte. To assess the suitability of the method in real exposure scenarios, we analyzed samples collected anonymously from subjects with suspected exposure to pesticides (n = 40) or who had been on an organic diet (n = 50). We detected glyphosate in 80% of subjects reporting an organic diet and in 78% of those with suspected glyphosate exposure; concentrations ranged from less then 0.2 to 28.6 μg/L. Median concentrations were 0.39 μg/L for the organic diet group and 0.40 μg/L for individuals with suspected exposure. Interestingly, interquartile ranges were considerably higher among those reporting pesticide exposure (0.63 μg/L) than those consuming organic diets (0.42 μg/L). These data suggest that the method meets typical validation benchmark values and is sensitive to investigate background exposures in the general population.Sea turtle populations foraging in coastal areas adjacent to human activity can be exposed to numerous chemical contaminants for long periods of time. For trace elements, well-developed, sensitive and inexpensive analytical techniques remain the most effective method for assessing exposure in sea turtles. However, there are many thousands more organic contaminants present in sea turtles, often at low levels as complex mixtures. Recently developed species-specific in vitro bioassays provide an effective means to identify the presence, and effect of, organic chemicals in sea turtles. This study used a combination of chemical analysis and effects-based bioassays to provide complementary information on chemical exposure and effects for three green turtle foraging populations (Chelonia mydas) in southern Queensland, Australia. Blood was collected from foraging sub-adult green turtles captured in Moreton Bay, Hervey Bay, and Port Curtis. Twenty-six trace elements were measured in whole blood using ICP-MS. Organic contaminants in turtle blood were extracted via QuEChERS and applied to primary green turtle skin fibroblast cell in vitro assays for two toxicity endpoints; cytotoxicity and oxidative stress.
