We undertook a comprehensive study of Tfh cells, along with B cells and antibody induction in children and adults with malaria. Activation and proliferation of circulating Tfh (cTfh) cell subsets was measured ex vivo and parasite-specific Tfh cell frequencies and functions studied with Activation Induced Marker (AIM) assays and intracellular cytokine staining.

Alterations in the anatomic and biomechanical properties of the ascending aorta (AAo) can give rise to various vascular pathologies. The aim of the current study is to gain additional insights in the biology of the AAo size and function.
We developed an AI based analysis pipeline for the segmentation of the AAo, and the extraction of AAO parameters. We then performed genome-wide association studies of AAo maximum area, AAo minimum area and AAo distensibility in up to 37,910 individuals from the UK Biobank. Variants that were significantly associated with AAo phenotypes were used as instrumental variables in Mendelian randomization analyses to investigate potential causal relationships with coronary artery disease, myocardial infarction, stroke and aneurysms.
Genome-wide association studies revealed a total of 107 SNPs in 78 loci. We annotated 101 candidate genes involved in various biological processes, including connective tissue development (THSD4 and COL6A3). Mendelian randomization analyses showed a causal association with aneurysm development, but not with other vascular diseases.

One common explanation for this lack of an effect is the facsimile accommodation hypothesis. This hypothesis states that people can "look past" the limitations of a reproduction, which obscures the effect. However, this hypothesis itself has never been tested. In the current paper, we therefore test the facsimile accommodation hypothesis. In Study 1 (N = 120) we found no evidence for the facsimile accommodation hypothesis there was no difference between the non-accommodation (instructed to evaluate the artwork as it is presented on the screen), the accommodation condition (instructed to evaluate the artwork as they imagine it would look in real life), and the control condition (no instructions). Though the control and accommodation condition did not differ which would be in line with the facsimile accommodation hypothesis, neither differed from the non-accommodation condition which they should have if the facsimile accommodation hypothesis was correct. We substantiate these findings by a pre-registered replication study (Study 2, N = 205), using Bayes Factors as well as equivalence testing to provide evidence for a null-effect, again finding no evidence for the facsimile accommodation hypothesis and even finding substantial evidence in favor of the null-hypothesis. In sum, we conclude that there is no evidence for the facsimile accommodation hypothesis and that we need to consider alternative explanations in order to understand the lack of empirical evidence for the genuineness effect.
To compare the outcomes of brolucizumab versus (vs) aflibercept in neovascular age-related macular degeneration (nAMD) patients with early residual fluid in the HAWK and HARRIER studies.
Post hoc analysis using pooled data from the Phase III studies HAWK (NCT02307682) and HARRIER (NCT02434328).
The early residual fluid cohort for this post hoc analysis consisted of patients treated with either brolucizumab 6 mg (N=730 patients) or aflibercept 2 mg (N=729 patients) and who had spectral domain optical coherence tomography (SD-OCT)-verified presence of intraretinal fluid (IRF) and/or subretinal fluid (SRF) at the Week 12 clinic visit.
After three initial monthly doses, brolucizumab-treated patients received injections every 12 (q12w) or every 8 (q8w) weeks, depending on nAMD disease activity, whereas aflibercept-treated patients received fixed q8w dosing.

In the AR HDFT-F technique reported by our group, the integration of AR into the microscope comes through the BrainLAB Curve navigation platform (BrainLAB AG, Munich Germany), Smartbrush software (BrainLAB AG), KINEVO 900 surgical microscope (Carl Zeiss, Oberkochen, Germany), and YELLOW 560 filter (Carl Zeiss).9 The microscope establishes a wired autodetection of the navigation platform, and the eyepiece functions as a "see-through display" of the AR images, which are overlapped onto the surgical field. To investigate the presence of flavoprotein fluorescence (FPF) at the optic nerve head (ONH) rim as a marker of mitochondrial dysfunction in primary open-angle glaucoma (POAG) and control eyes.
Retrospective cross-sectional study of patients recruited from the New York Eye and Ear Infirmary of Mount Sinai.
A total of 86 eyes (50 eyes of 30 patients with POAG and 36 eyes of 20 control participants) were enrolled. The presence of POAG was defined by circumpapillary retinal nerve fiber layer thickness (cpRNFLT) of less than the bottom fifth percentile of the normative database, glaucomatous ONH changes, and visual field defects on 24-2 tests.
Primary open-angle glaucoma and control eyes were imaged using the OcuMet Beacon. A 23°× 23° infrared scan was obtained, and an FPF scan was performed within a capture field spanning 13° in diameter. The ONH margins on the infrared image were identified by software algorithms. Then, FPF was measured within an elliptical annulus around the ONH rim, with the inner andgher FPF in POAG versus control eyes suggests the presence of mitochondrial dysfunction at the ONH rim in eyes with glaucomatous damage. The degree of FPF corresponds to disease severity, as measured by visual field and nerve fiber layer thickness metrics. Thus, FPF may represent a metabolic indicator of disease status that reveals the extent of injury in glaucoma.Toxoplasmosis is a cosmopolitan protozoan zoonosis caused by Toxoplasma gondii infamous for inducing severe clinical manifestations in humans. Although the disease affects at least one billion people worldwide, it is neglected in many countries including developed ones. In literature, the epidemiological data documenting the actual incidence of the disease in humans and domestic animals from Japan are limited and importantly many earlier papers on T. gondii infections were published in Japanese and a considerable part is not available online. Herein, we review the current summary about the epidemiological situation of T. gondii infection in Japan and the potential associated risk factors in humans and animals as well as the different T. gondii genotypes isolated in Japan. Several T. gondii isolates have been identified among cats (TgCatJpTy1/k-3, TgCatJpGi1/TaJ, TgCatJpObi1 and TgCatJpOk1-4) and goats (TgGoatJpOk1-13). This literature review underscores the need for a nationwide investigation of T. gondii infection in Japanese people and assessment of the socioeconomic impact of the disease burden. Furthermore, epidemiological studies in domestic and wild animals and estimation of degree of contamination of soil or water with T. gondii oocysts are needed, for a better understanding of the scope of this public health concern.

Historical treatment includes limiting dietary phenylalanine (Phe) consumption while supplementing with medical food; however, this treatment has been associated with complications, such as nutritional deficiencies and disruptions in the gut microbiota.
The study aim was to compare dietary and gut microbiome differences between adult patients on a traditional PKU diet with those receiving the enzyme substitution therapy Palynziq on a liberalized diet while controlling blood Phe levels to <600 μmol/L (to convert to mg/dL divide by 60.5).
