Construction sites exhibit unique hazardous characteristics. Hence, investigating the causes of fatal and nonfatal accidents is extremely important to promote safety on construction sites. Literature shows a risk perception (RP) scale for construction workers; to expand the existing research, this study aims to identify risky scenarios for assessing the RP of Thai construction workers, with the goal of reducing workplace incidents.

The scale development process consists of four phases. Item development was also performed, followed by factor analysis. link2 Reliability and validity assessments were finalized in the process. A survey of 500 construction workers in Thailand was conducted to investigate risky scenarios. Through the process, a total of 17 items remained in the final RP scale, and the reliability and validity of these items were confirmed.

The RP scale was affirmed to have four dimensions (probability, severity, worry, and unsafe). These dimensions of workers' RP were negatively correlated with the workers' risk-taking behaviors. This study further showed that participants with a high level of affective RP were more likely to have a high level of cognitive RP. link3 Hence, those with a high level of RP tended to be involved in fewer risk-taking behaviors. Practical Applications The final version of the scale was reliable and valid in determining the RP and risk-taking behaviors of construction workers in Thailand.

The objective of this study was to determine the reciprocal relationship between safety professionals perceived organizational support (POS) and perceived safety climate. Safety professionals are most effective when they perceive support from management and employees and they also attribute most of their success to support from the organization. Their work directly improves safety climate, and organizations with a high safety climate show a higher value for the safety professional. The causal direction of this relationship is, however, unclear.

Using a sample of 162 safety professionals, we conducted a cross-lagged panel study over one year to examine whether safety professionals' POS improves their perceived safety climate and/or whether safety climate also increases POS over time. Data were collected at two points and, after testing for measurement invariance, a cross-lagged SEM was conducted to analyze the reciprocal relationship.

Our findings show that safety professionals' POS was positively relateionals feel supported, this study points out the kind of support that contributes to improved organizational safety. Support for safety professionals may come in classical forms such as approval, pay, job enrichment, and information on or influence over organizational policies. There are numerous examples of bioactive compounds containing carbonyl groups including modified proteins with oxidation of side chain of amino acid residues to aldehyde/ketone groups which are frequently considered as markers of oxidative stress. The carbonyl unit can be also distinguished as a substructure in many illegal drugs including anabolic steroids as well as cations derivatives. Based on chemoselective formation of oximes by solid phase immobilized hydroxylamine derivatives we proposed the protocol for derivatization and selective detection of carbonylated compounds in human serum albumin hydrolysate as a complex peptide mixture and of testosterone in urine samples. This allowed for the removal of the matrix and the qualitative and quantitative analysis of the derivatized analyte by LC-MS/MS (or LC-MRM). Herein we report the preparation and chemical characterization of a novel, ChemMatrix - based resin functionalized with aminooxyacetic acid (AOA). The hydroxylamine moiety in this resin is combined with a peptide linker (GRG) containing an arginine residue to enhance the ionization efficiency. Application of an isotopically labeled carbonylated peptide ((H-Leu-Val-Thr(O)-Asp-Leu-Thr-Lys [13C6,15N2]-OH and testosterone-d3 allowed us to carry out quantitative analyses of detected compounds. Our method is general and may be applied for analysis of carbonylated compounds in biological samples. Our method based on application of functionalized resin allowed to quantify the level of free testosterone in small sample (0.5 mL) of urine, while the non-derivatized testosterone from urine sample was not detected during direct LC-MRM analysis. The level of melatonin in human milk might be closely related to infant development and the building up of their circadian rhythms. The large population investigation on this topic would provide insights for the prevention and treatment of diseases related to early development and circadian rhythms. However, it has not been well studied. Trace level endogenous melatonin and difficulties in sample collection are among the challenges limiting the progress. High throughput analytical method with high specificity and sensitivity to determine the endogenous melatonin concentration is highly desired. A newly developed easily operated and high-throughput sensitive on-line enrichment liquid chromatography-tandem mass spectrometry (LC-MS/MS) method would be reported in this paper. Melatonin-d3 (MEL-d3) was used as a surrogate standard for the calibration curve and melatonin-d4 (MEL-d4) was used as an internal standard. Sample preparation was simply performed in 96-well plate by protein precipitation using acetonitrileiquid handling system and was more suitable for large population cohort studies on trace level endogenous melatonin determination.Monitoring of cysteine (Cys) is of significant importance for studying Cys-involved biological functions and clinically diagnosing Cys-related diseases. Recently, few fluorescent probes with two different reacting sites were reported to be capable of sensing different concentration ranges of Cys with distinct fluorescence signals, particularly suiting for bioimaging. However, due to relative sophisticated synthesis and moderate selectivity, the applications of these probes were still severely restricted. In this work, we proposed a novel probe design strategy by utilizing two same reacting groups, instead of two different reacting groups, to simplify the synthesis route and minimize the interference from competing species. Same reacting groups in a probe with different steric hindrances could exhibit different reactivities to Cys. This probe showed distinguishable fluorescence peak wavelengths towards low and high concentration ranges of Cys, giving green and blue emissions, respectively. Moreover, this probe was successfully applied for monitoring of Cys concentration in living cells. We believe this work provided a simpler strategy for dual-site fluorescent probes to sense difference concentration ranges of Cys, which may inspire more probe design in future.Herein, we report a novel hairpin structure-mediated diagnostic method for the simple and rapid colorimetric detection of miRNA through the sensing of pyrophosphate. When the hairpin structure of the template DNA (h-Probe) was hybridized with the primer, the DNA primer extension mediated by nPfu special enzyme was blocked. However, this h-Probe was extended using nPfu special enzyme, upon the structural change of the template DNA, from a hairpin to a linear structure, in the presence of the target miRNA. The miRNA-hybridized template DNA sequence was cleaved by a duplex-specific nuclease (DSN), which cleaved the DNA from the RNA-DNA hybrid, thereby allowing the target miRNA to be recycled. Primer extension using nPfu special enzyme produced pyrophosphate when nucleotide triphosphate was incorporated into the DNA; this pyrophosphate was sensed in terms of a color change, from pink to colorless, when using pp Probe, a probe developed previously by our group. This novel system for the colorimetric detection of target miRNA operated with high sensitivity (LOD = 132 aM) and selectivity, with the whole detection process requiring only 30 min. Furthermore, this system could also detect miRNA fluorimetrically with similar sensitivity (LOD = 105 aM), highlighting the dual-sensing properties of pp Probe. This unique, extremely simple, and rapid system for the detection of miRNA through a highly sensitive color change would presumably be useful in applications requiring point-of-care detection. The extraordinary genome-editing tool CRISPR/Cas12a has also been utilized as a powerful sensing technology owing to its highly-specificity and isothermal signal amplification. Nevertheless, the widespread application of Cas12a-based sensing methods in nucleic acid detection is limited by the targeting range and high undesired background. Herein, we established a universal Cas12a-based nucleic acid sensing strategy by using proximity extension and transcription-unleashed self-suppling of crRNA. The target was recognized and bound to a pair of adjacent probes, and then triggered the proximity-induced primer extension and transcription amplification to produce numerous crRNAs. The amplified abundant crRNAs assembled with Cas12a and dsDNA activators containing PAM to form a ternary complex, which trans-cleaved ssDNA-FQ reporters continuously to generate a strong fluorescent signal. Thus, the cascade enzymatic amplification was performed and subsequently applied for detecting target DNA down to 41.7 amol with a low nonspecific background. The application of this strategy in RNA detection has also been demonstrated, and it is expected to provide a universal and sensitive sensing platform for molecular diagnosis applications. The modulation of the ion-fluxes across a polymeric membrane is important for designing attractive methodologies. As an alternative to the commonly used dynamic electrochemistry approaches, light can be used as an external stimulus and provides a very convenient way to manipulate ions release and/or extraction into a polymeric membrane. Herein, we designed a solid-contact polymeric membrane ion-selective sensor that exhibits dynamic response by light irradiation at 375 nm. The electrode membrane contains a light-sensitive lipophilic salt (bis(4-tert-butylphenyl)iodonium tetrakis[3,5-bis(trifluoromethyl)phenyl]borate (R+-R-, BTDT-TFPB) instead of traditional ion exchanger. Under light illumination, the decomposition of the lipophilic cation makes the membrane with ion-exchange properties. The solid-contact ion-selective electrodes based on potentiometry and constant potential coulometry have been explored for direct ion sensing. Copper was selected as a mode analyte and can be determined at micromole levels. The proposed dynamic ion sensors show promise for on-demand ion sensing.
