Herein, we investigated the community composition and phylogeny of rare (relative abundance 1%) bacteria in dryland grassland soils on the Tibetan Plateau. Results revealed similar biogeographical patterns of rare and abundant bacteria at both compositional and phylogenetic levels, but rare subcommunity was more heavily influenced by stochasticity (72%) than the abundant (57%). The compositional variation of rare bacteria was less explained by environmental factors (41%) than that of the abundant (80%), while the phylogeny of rare bacteria (36%) was more explained than that of the abundant (29%). The phylogeny of rare bacteria was equally explained by local factors (soil and vegetation) and geospatial distance (11.5% and 11.9% respectively), while that of the abundant was more explained by geospatial distance (22.1%) than local factors (11.3%). Furthermore, a substantially tighter connection between the community phylogeny and composition was observed in rare (R2 = 0.65) than in abundant bacteria (R2 = 0.08). Our study provides novel insights into the assembly processes and biographical patterns of rare and abundant bacteria in dryland soils. © 2020 Society for Applied Microbiology and John Wiley & Sons Ltd.BACKGROUND Precise timing of pesticide applications, as recommended by decision support systems, can ensure crop protection, while maintaining efficient use of pesticides. Yet, farmers often deviate from recommended timing strategies. Here, we assess and explain farmers' choices to follow or not follow recommendations for the timing of fungicide applications against potato late blight in Switzerland. RESULT Based on daily fungicide application records as well as regional application recommendations and disease pressure we found that 36% of applications take place earlier than recommended. Using regression analysis, we identified the exposure to economic risks of infection, susceptibility of the planted potato varieties to late blight infections, as well as yearly differences in disease occurrence as the most important determinants of farmers' application decision. CONCLUSION Our results indicate that decisions to not follow application recommendations and apply early are linked to available information and uncertainty with respect to disease predictions. Based on our results, we make recommendations on how to account for farmers' uncertainty with regard to the timing of pesticide applications in the design of pesticide policies and agricultural decision support systems. These include the use of new technologies and data, mandatory reporting and tailor made taxes and insurance solutions. Although the focus of this article is on late blight in Switzerland, our analysis can easily be extended to other countries and important plant diseases like powdery mildew in grapevines or Fusarium head blight in winter wheat. All rights reserved.Most of the oil in low temperature, non-uplifted reservoirs is biodegraded due to millions of years of microbial activity, including via methanogenesis from crude oil. To evaluate stimulating additional methanogenesis in already heavily biodegraded oil reservoirs, oil sands samples were amended with nutrients and electron acceptors, but oil sands bitumen was the only organic substrate. Methane production was monitored for over 3000 days. Methanogenesis was observed in duplicate microcosms that were unamended, amended with sulfate, or that were initially oxic, but methanogenesis was not observed in nitrate-amended controls. The highest rate of methane production was 0.15 μmol CH4 g-1 oil d-1 , orders of magnitude lower than other reports of methanogenesis from lighter crude oils. Methanogenic Archaea and several potential syntrophic bacterial partners were detected following the incubations. GC-MS and FTICR-MS revealed no significant bitumen alteration for any specific compound or compound class, suggesting that the very slow methanogenesis observed was coupled to bitumen biodegradation in an unspecific manner. After 3000 days, methanogenic communities were amended with benzoate resulting in methanogenesis rates that were 110-fold greater. This suggests that oil-to-methane conversion is limited by the recalcitrant nature of oil sands bitumen, not the microbial communities resident in heavy oil reservoirs. All rights reserved.Canine melanomas and mast cell tumours (MCTs) frequently metastasize to lymph nodes, worsening prognosis compared with dogs without metastasis. Sentinel lymph node (SLN) evaluation is more specific than evaluation of the lymph node closest to the tumour, which may not be the draining lymph node. Computed tomography lymphangiography (CTL) allows for SLN identification and one study of canine mammary tumours found that CTL was able to assist in determination of the metastatic status of inguinal SLNs prior to extirpation and histopathology. The objective of the present study was to evaluate CTL for use in determining metastasis to the SLN in dogs with a pre-operative diagnosis of melanoma or MCT in various locations by correlating CTL findings with histopathology. The hypothesis was that CTL would not be able to determine the metastatic status of lymph nodes, based on author experience. Dogs were prospectively enrolled and underwent CTL and subsequent SLN extirpation. Histopathology results for the primary tumour, SLN, and additional extirpated lymph nodes were recorded.
