Although studies already exist in this regard, the molecular mechanisms by which CdSe/ZnS quantum dots exert their toxic effects are still unknown. For this reason, in this study, a comprehensive proteomic approach has been designed, applying the SILAC strategy to an in-vitro model (hepatic cells) and the super-SILAC alternative to an in-vivo model, specifically zebrafish larvae. This integral approach, together with additional bioanalytical assays, has made it possible for the identification of proteins, molecular mechanisms and, therefore, biological processes that are altered as a consequence of exposure to CdSe/ZnS quantum dots. It has been demonstrated, on the one hand, that these quantum dots induce hypoxia and ROS generation in hepatic cells, which leads to apoptosis, specifically through the TDP-43 pathway. On the other hand, it has been shown that exposure to CdSe/ZnS quantum dots has a high impact on developing organisms, inducing serious neural and developmental problems in the locomotor system.A hydroponic study was conducted to determine the effects of single and/or combined application of different doses (0, 5 and 10 μM L-1) of abscisic acid (ABA) and 6-benzylaminopurine (BAP) on cobalt (Co) accumulation, morpho-physiological and antioxidative defense attributes of tomato (Solanum lycopersicum L.) exposed to severe Co stress (400 μM L-1). The single Co treatment (T1), prominently decreased tomato growth, relative water contents, photosynthetic pigments (chlorophyll a and chlorophyll b), whereas enhanced oxidative stress and Co accumulation in shoot and root tissues. click here Nonetheless, the supplementation of ABA and 6-BAP via nutrient media significantly (P less then 0.05) enhanced plant biomass, root morphology and chlorophyll contents of tomato, compared to only Co treatment (T1). Moreover, the oxidative stress indicators such as malondialdehyde, proline and H2O2 contents were ameliorated through activation of enzymatic antioxidant activities i.e. link2 ascorbate peroxidase, superoxide dismutase, catalase, and peroxidase, in growth modulator treatments in comparison to T1. The Co uptake, translocation (TF) and bioaccumulation factor (BAF) by shoot and root tissues of tomato were significantly reduced under all the treatments than that of T1. The supply of 6-BAP alone or in combination with ABA at 10 μM L-1 application (T7) rate was found the most effective to reduce Co accumulation in the roots and shoots by 48.4% and 70.2% respectively than T1 treatment. It can be concluded that two plant growth modulators could improve the stress tolerance by inhibition of Co uptake in tomato plants.Microplastic contamination poses a great threat to the health of aquatic species, which may affect their reproduction and result in ecological consequences. There is a need to further elucidate the potential impact microplastics can impose on aquatic species. In this study, the effects of exposure to polystyrene microplastics on reproductive organs, and the underlying response mechanisms, were investigated using zebrafish Danio rerio. After 21 days of continuous waterborne exposure, no significant difference was observed at the lower concentration of 10 μg/L. At concentrations above 100 μg/L, significantly enhanced reactive oxygen species (ROS) level was found in both male and female liver and gonads. At the concentration of 1000 μg/L, significantly increased apoptosis levels were observed in male testes, triggering increased expression of p53-mediated apoptotic pathways; histological alteration in the form of a significant decrease in testis basement membrane thickness was also observed. This study demonstrated that exposure to microplastics can induce molecular responses and histological alterations in fish gonads, implying potential adverse impact on fish reproductive organs. This work provided new insights on the reproductive damage microplastics can cause in fish and have implications in fields of freshwater ecology and environmental toxicology.In this work, we developed an analytical approach using an ultrasound-assisted extraction (UAE) followed by continuous solid-phase extraction (SPE) and gas chromatography-mass spectrometry (GC-MS) detection in order to determine simultaneously 24 endocrine disrupting chemicals such as alkylphenols, organophosphorus pesticides, parabens, phenylphenols, triclosan and bisphenol A in vegetable and fruit samples. Different variables influencing UAE and SPE performance were optimized in order to maximize removal of the sample matrix and preconcentration of the analytes. The optimized extraction and GC-MS quantitation conditions provided acceptable sensitivity, selectivity, accuracy and precision. Limits of detection spanned the range 0.6-25 ng kg-1, recoveries were near-quantitative and relative standard deviations ranged from 4.5 to 7.6%. The proposed method was used to analyse 11 vegetable samples and 7 fruit samples purchased at various Spanish and Moroccan supermarkets. Most samples contained more than three of the analytes, at levels between 5.8 and 580 ng kg-1.In this study, polybrominated and mixed halogenated dibenzo-p-dioxins and furans (PBDD/Fs and PXDD/Fs), and dioxin-like polychlorinated biphenyls (dlPCBs) were quantified in 24 human milk samples of first-time lactating mothers from Greater Accra region in Ghana. The aims of the study were to determine the concentrations and toxic equivalent concentrations of PBDD/F, PXDD/F and dlPCBs in human milk, and to estimate an infant's daily intake. The samples were analysed for 12 dioxin-like PCBs, 7 congeners of 2,3,7,8-polybrominated dibenzo-p-dioxins and furans (PBDD/Fs), and 7 congeners of 2,3,7,8-mixed halogenated dioxins and furans (PXDD/Fs, where X = Br/Cl). The mean concentrations in human milk ranged from 0.15 to 212.9 pg/g lipid for dlPCB congeners (mean TEQ 1.67 pg WHO2005-TEQ/g lipid). Lesser concentrations for 2,3,7,8-PXDD/Fs (and PBDD/Fs congeners) ranged between less then 0.01-1.67 pg/g lipid, with a total mean tentative TEQ of 0.56 pg WHO2005-TEQ/g lipid. For an infant of average weight 7 kg, consuming an estimated volume of 600 mL human milk, the estimated average daily intake of dlPCBs in 21 human milk samples was 4.95 pg TEQ/kg bw/day; contributions from dlPCBs, PXDD/Fs and PBDD/Fs resulted in an average estimated daily intake of 6.56 pg TEQ/kg bw/day. The results obtained in this study, although lower than infant dietary intake estimates in human milk from industrialized countries, exceeded the recommended safety standards of 1 pg TEQ/kg bw/day and 1-4 pg TEQ/kg bw/day from the Agency for Toxic Substances and Disease Registry (ATSDR) and the World Health Organization (WHO), respectively.Subsurface storm flow of phosphorus (P), including particulate P, has been recently discussed as an important P transport path in contrast to typical surface runoff events. However, P speciation, and P concentration during storm events has not been extensively investigated; therefore, its contribution to the water quality is not clearly understood. In this study, the physicochemical properties of particulate P in tile water samples during a high flow event were investigated in Midwestern agricultural lands using wet chemical methods, 31P Nuclear Magnetic Resonance spectroscopy and P K-edge X-ray absorptions near edge structure spectroscopy. In slightly alkaline pH tile water, total P was ranging from ∼0.06 to 0.22 mg L-1, which is significantly greater than dissolved reactive P (DRP) (∼0.02-0.08 mg L-1). The tile water contains P enriched particulate matters (∼200-660 mg L-1). Total P in the colloidal fraction was from 1013 to 2270 mg kg-1. Phosphate and organic P species, especially monoesters, are sorbed in soil colloids like calcite, and iron oxides, and colloids are effective carriers of P in the subsurface transport process during storm events. The results of this study show that storm events can accelerate the subsurface transport of P with soil particles in addition to DRP.Despite the North American production ban of polychlorinated biphenyls (PCBs), PCBs are ubiquitous in the environment and in wildlife tissues. Chelydra serpentina serpentina (common snapping turtle) have been used as environmental indicators of PCB pollution upwards of 40 years given their high site fidelity and high trophic position. Despite their long use as indicators of PCB contamination, the effects of PCBs in reptiles remain largely unknown. In this study, we performed two experiments to assess i) bioaccumulation and ii) toxicity of PCBs to 1-month-old C. s. serpentina, to aid in interpretation of PCB burdens. Food pellets were spiked at an environmentally relevant concentration (0.45 μg/g) of the PCB mixture Aroclor 1254 to model hepatic bioaccumulation and depuration, through feeding, for 31 days and clean food for 50 days, respectively. No significant differences in PCB concentrations were observed in liver tissue over the course of the experiment, suggesting that juvenile turtles can likely metabolize low environmentally occurring concentrations of PCBs. Additionally, a dose-response experiment, performed to determine hepatic toxicity and bioaccumulation in juvenile C. s. serpentina, showed a 1.8-fold increase in hepatic expression of cyp1a when fed A1254-spiked pellets (12.7 μg/g; range 0-12.7 μg/g). This gene induction correlates with the significant increase of group 3 PCB congeners measured in the turtle liver, which are known to be metabolized by CYP1A. This study indicates that C. s. serpentina may be a good environmental indicator for PCBs, while more research is needed to assess the effects of body burdens in wild C. s. serpentina.This publication presents a comparison of the content of pollutants in groundwater samples taken at 117 measurement points in four regions of Poland during a drought period and in the reference period without drought. Based on the chemical analyses of water, an assessment of the health risk resulting from the use of underground water for consumption was carried out. The study aimed to determine whether drought affects the increase in health risk exposure of the population. link3 It was found that despite the occurrence of drought, the expected increase in the concentration of pollutants in water does not take place in all locations. This study found that in some cases the occurrence of drought did not cause an increase in the non-cancerogenic threat expressed by the hazard index. There were also no clear changes in excess lifetime cancer risk values except for selected measurement points. On the other hand, the statistical analysis of all data collected in the regions where the research was conducted showed a general trend of increasing environmental health risk caused by changes in groundwater pollution during drought.
