The bioplatform involves sandwich-type immunoassays using specific antibodies for β-conglycinin and glycinin, which are the main allergenic soy proteins, and carboxylic acid-modified magnetic microbeads. Amperometric detection at -0.20 V (vs. an Ag pseudo-reference electrode) was performed using single or dual screen-printed carbon electrodes and the H2O2/hydroquinone (HQ) system. The measured variation in the cathodic current was directly proportional to the concentration of target allergenic proteins. The developed bioplatforms exhibit a good selectivity and sensitivity providing limits of detection (LOD) values of 0.03 and 0.02 ng mL-1 for β-conglycinin and glycinin, respectively. The determination of both proteins can be carried out in only 1.5 h. The electrochemical bioplatforms allow their accurate determinations (with results statistically comparable to those provided by ELISA methodologies) in raw cookie dough and baked cookies enriched with soy flour. The results obtained confirm, in a pioneering way with electrochemical biosensors, the possibility of discriminating samples incurred with as little as 0.0005 ppm of a food allergen in model cookie extracts.Fluorescence microscopy is an extremely powerful technique that allows to distinguish multiple labels based on their emission color or other properties, such as their photobleaching and fluorescence recovery kinetics. These kinetics are ideally assumed to be mono-exponential in nature, where the time constants intrinsic to each fluorophore can be used to quantify their presence in the sample. However, these time constants also depend on the specifics of the illumination and sample conditions, meaning that identifying the different contributions in a mixture using a single-channel detection may not be straightforward. In this work, we propose a factor analysis approach called Slicing to identify the different contributions in a multiplexed fluorescence microscopy image exploiting a single measurement channel. With Slicing, a two-way dataset is rearranged into a three-way dataset, which allows the application of a trilinear decomposition model to derive individual profiles for all the model components. We demonstrate this method on bleaching - recovery fluorescence microscopy imaging data of U2OS cells, allowing us to determine the spatial distribution of the dyes and their associated characteristic relaxation traces, without relying on a parametric fitting. By requiring little a priori knowledge and efficiently handling perturbation factors, our method represents a general approach for the recovery of multiple mono-exponential profiles from single-channel microscopy data.Post-printing treatment is a promising means of enhancing the performance of devices manufactured using conventional three-dimensional printing (3DP) technologies. In this study we developed a post-printing solution foaming process-involving respective treatment with formic acid (60%, v/v) and sodium bicarbonate (5%, w/v) solutions to generate CO2 as a foaming agent-to increase the surface roughness and porosity of the polyamide 6 (PA6) monolithic packing in a multimaterial fused deposition modeling 3D-printed solid phase extraction (SPE) column, thereby enhancing the extraction of Mn, Co, Ni, Cu, Zn, Cd, and Pb ions from complicated real samples prior to their determination using inductively coupled plasma mass spectrometry. After optimizing the column fabrication process, the solution foaming treatment process, the extraction conditions, and the automatic analytical system, the 3D-printed SPE column incorporating the solution foaming-treated PA6 monolithic packing extracted these metal ions with 18.9- to 42.0-fold enhancements, relative to those of the as-printed column, with absolute extraction efficiencies all greater than 94.3% and method detection limits ranging from 0.2 to 7.7 ng L-1. We verified the reliability and applicability of this method through analyses of the tested metal ions in several reference materials (CASS-4, SLEW-3, 1643f, and 2670a) and spike analyses of seawater, river water, ground water, and urine samples. We conclude that post-printing treatment can dramatically improve the performance of 3D-printed analytical devices.As an important post-translational modification in response to oxidative and nitrosative stress, protein tyrosine nitration is deeply involved in many physiological and pathological processes. Identifying tyrosine nitration in proteins is challenging due to its low abundance.Consequently, pre-separation and enrichment of tyrosine-nitrated peptides (TNPs) are necessary before submitting them to mass spectrometry analysis. However, the most popularly used anti-nitrotyrosine antibody pull-down method showed limitations like sequence preference and unspecific binding. Therefore, developing novel affinity purification materials for TNPs is of significance. In the present study, we screened the phage-displayed 12-mer randomized peptide library for affinity binding peptide of the synthetic standard TNP (sTNP, sequence H2N-GGGGY*GGG-COOH) and identified a peptide named NT-1 (H2N-TLWPFDLWLKTR-COOH) as a promising candidate. NT-1 at extremely low concentration (3 nM) in solutions could be efficiently captured by immobilized sTNP as determined by pull-down and subsequent MALDI-TOF MS analysis. Surface plasmon resonance (SPR) measurement confirmed that NT-1 possesseed a good selectivity, showing more than 100-fold higher binding affinity with TNP than its non-nitrated counterpart. Moreover, NT-1 could efficiently capture various types of TNPs in solutions even in the presence of 1000-fold excessive amount of trypsinized BSA fragments. Most importantly, NT-1 showed superiority to commercially used nitrotyrosine antibody as the former captured more TNPs, with less sequence preference. In summary, our study provided NT-1 as a novel affinity binding ligand for TNPs and should be useful in developing an alternative enrichment strategy for TNPs.Medicalization is increasingly recognized as a bidirectional process, with patients and their families as agents. Through reporting and comparing results of two independently conducted qualitative studies, this paper aims to analyze how parents embrace and resist the medicalization of autism in their everyday lives and in healthcare contexts. Both studies involved participant-observation with services that targeted autistic people and interviews with parents, professionals, and autistic people. Results show that parents of autistic people both embrace and resist medicalization. While parents (sometimes ambivalently) accept the responsibilization inherent in their engagement with interventions (a sort of "therapeutization" of life) and reject lay expertise by deferring to experts' knowledge, they also resist the application of medical labels, language and practices in various ways in their everyday lives. We find that giving birth in a private hospital leads to a 4 percentage point increase in the probability of having an unplanned caesarean birth. Over our study period, this equates to an additional 3241 caesarean births.
This systematic review evaluates and summarizes the effects of pelvic floor muscle training on urinary incontinence and sexual function in people with multiple sclerosis.

PubMed, Scopus, PEDro, CINAHL, Web of Science, and ULAKBIM databases with keywords multiple sclerosis, urinary incontinence, sexual function, and pelvic floor muscle training were screened for randomized controlled studies and clinical trials from the beginning until July 2020 (updated in August 2021).

Two authors independently made the study selection. Turkish and English publications were taken into consideration. The risk of bias for the included studies was assessed using the Revised Cochrane risk-of-bias tool for randomized trials. The Tool for the assEssment of Study qualiTy and reporting in EXercise (TESTEXT) was used to assess the methodological quality of studies. Data extraction was performed by two researchers independently.

However, it should be noted that all included studies had some concerns or a high risk of bias in at least one domain of risk of bias assessment, and most of the studies did not have high quality.Several putative neurorestorative therapies in multiple sclerosis (MS) have recently failed; this includes high-dose biotin, bexarotene, a retinoic acid receptor gamma agonist, and opicinumab (anti-LINGO-1). Are these failures biological or due to poor trial design? We argue that the failure to include exercise in these trials and selecting participants without the capacity for repair may explain these disappointing results. We propose the need for mapping the biological mechanisms of recovery within trials, understanding the critical window when remyelination/repair occurs in terms of targeting interventions at the right time and selecting subjects who are capable of repair. We also make the case for testing combinations that include other pro-repair interventions such as exercise, Nrf2 inducers and possibly neurostimulation. The MS community can't afford for any more treatments to fail because of poor trial design and ignoring biology.
