Valvular calcification commonly occurs in elderly individuals, and is increasingly considered an important economic and health burden. However, no efficient drugs against valvular calcification are available. The present work aimed to examine emodin's suppressive effect on high-calcium-dependent valve calcification and explore the underpinning mechanisms.

Experiments were carried out in mice receiving vitamin D (Vit D) to induce valvular calcification.

Cell viability and apoptosis assays demonstrated celastrol suppressed proliferation and increased apoptosis in porcine aortic valve interstitial cells (PAVICs) at concentrations higher than 10μM. Emodin (5μM) attenuated the upregulation of osteogenic genes as well as calcium accumulation in PAVICs under high-calcium conditions. The elevations of calcium content in serum and valve, and calcium accumulation in valve and artery were suppressed by emodin in mice with valvular calcification after joint treatment with adenine and Vit D. In addition, p-AKT and p-FOXO1 were upregulated in PAVICs under high-calcium conditions, and this effect was reversed by emodin treatment. SC79, an AKT activator, reversed emodin's suppressive effects on increased calcium content, calcium deposition and osteogenic gene expression in PAVICs induced by calcific medium.

The short- and long-term regulatory mechanisms of NAC on the 6-OHDA hemiparkinsonian rat model were longitudinally investigated by performing positron emission tomography (PET) imaging using the specific dopamine transporter (DAT) radioligand [18F]FE-PE2I. The results demonstrate that after a unilateral dopamine insult NAC has a strong influence on the non-lesioned hemisphere by decreasing the levels of DAT in the striatum early after the lesion. We interpret this early and short-term decrease of DAT in the healthy striatum of NAC-treated animals as a beneficial compensatory effect induced by NAC.Cognitive impairment in Parkinson disease (PD) leads to substantial disability. Unlike external manifestations such as tremor, the decay of cognitive function is often an underlying process, and its neuroanatomic substrates are not yet fully elucidated. Knowledge regarding cognitive-related alterations in white matter (WM) pathways helps us understand the mechanisms of cognitive decline in patients with PD. Previous voxel-based analyses with Diffusion tensor imaging (DTI) metrics, such as fractional anisotropy (FA) and mean diffusivity (MD) have uncovered white matter differences in groupwise, but the conclusions were inconsistent. That was partially due to white matter fibers that are known to affect cognition, such as the corpus callosum (CC) and superior longitudinal fasciculus that cross in voxel, and are hard to interpreted by the abovementioned metrics. Furthermore, cognitive decay is a continuous process, it is difficult to reflect the continuous changes of white matter fibers between groups comparison. In the present work, we chose the constrained spherical deconvolution (CSD) and the fixel model, which avoided the effect of crossing fibers. To compare the white matter fiber in different cognitive stages of patients with PD, the results found that the CC, the cingulum bundle (CB), and the corticospinal tract (CST) showed the same trend in the decline of cognitive function, and this change may lead to the impairment of cognitive function. Our findings can help physicians determine the cognitive stage of PD from the perspective of white matter fiber and provide a reference for clinical trials and predictions.
Rs9296559 within CD2-associated protein (CD2AP) has been identified as a susceptibility locus for Alzheimer's disease (AD). Recent studies indicated that CD2AP functioned as a regulator of endocytic trafficking to modulate the β-amyloid (Aβ) generation in neurons. Moreover, knockdown of cindr, the Drosophila ortholog of CD2AP, enhanced tau-induced neurodegeneration, implying CD2AP also participated in tau pathology. However, the role of rs9296559 in regulating Aβ and tau metabolism in AD was still unclear.

Here, the associations of rs9296559 with CSF Aβ
, p-tau, and t-tau were performed using a linear regression model in a total of 543 cognitive normal (CN), mild cognitive impairment (MCI), and AD subjects from the Alzheimer's disease Neuroimaging Initiative (ADNI) cohort. The results were replicated in an independent cohort consisting of 198 Chinese subjects recruited from our hospital.

In the ADNI cohort, CC+TC genotypes significantly increased CSF t-tau and p-tau levels in MCI patients but did not alter CSF tau levels in AD. This association was also observed in the replication cohort. Moreover, there was no association between rs9296559 and CSF Aβ
level at different disease statuses in the two cohorts.

Our findings showed that rs9296559 was associated with higher CSF t-tau and p-tau levels in MCI, supporting that CD2AP modified AD risk by altering tau-related neurodegeneration in the early stage of the AD continuum. To the best of our knowledge, this is the first study to evaluate the association between CD2AP genotypes and AD CSF biomarkers.

We define the value of the Minimal Clinically Important Difference for Worsening (MCID-W) for Patient-Reported Outcomes Measurement Information System Physical Function short form 10-a (PROMIS-PF-10a) score for primary total joint arthroplasty (TJA) of the hip and knee and describe the risk factors for patients scoring worse than the MCID-W.

This retrospective study was performed using 3414 primary TJA patients. PROMIS-PF-10a scores were collected at the preoperatively and postoperatively, and patients were classified based on reaching Minimal Clinically Importance Difference for Improvement (MCID-I), MCID-W, or "no significant change" after TJA (scores betweex`n MCID-W and MCID-I). MCID-W and MCID-I values were determined by a distribution method. The association between numerous variables and scoring worse than the MCID-W of PROMIS-PF-10a was then evaluated through multiple logistic regression. A threshold for preoperative PROMIS-PF-10a score predicting decline past MCID-W was determined using the Youdesociated patient- and hospital-level risk factors for failure after TJA. Healthcare funding initiatives should be directed toward modifiable factors associated with clinically significant worse outcomes after TJA.This study is designed to investigate the roles of five key terminal electron acceptors (TEAs) O2, NO3-, Fe3+, SO42-, and CH2O, typically existing in the sludge on the degradation rates and pathways of three representative MPs polylactic acid (PLA), polyvinyl chloride (PVC), and polyhydroxyalkanoate (PHA). The results revealed that approximately 51.46 ∼ 52.70% of PHA was degraded within 43 days, despite PLA and PVC being degraded insignificantly. Different TEAs significantly affected the end-products of PHA. The production rate of acetate gradually decreased from 90.48, 42.67, 38.30, and 17.56 to 3.30% when the TEAs were tested with CH2O, O2, SO42-, NO3- and Fe3+, respectively. The main functional bacteria involved in the PHA degradation were hydrolysis bacteria Burkholderiaceae and homo-acetogenic bacteria Clostridiacea, which accounted for 0.83% and 18.91% of the microbes. Synthetic wastewater was used as substrate at stage 1. It changed into the digested FPW with dilution variation in order to increase the nitrogen and COD loading rates. This study showed that the process has the advantages in treating the real high ammonia concentration of digested wastewater containing organic compounds. The nitritation and anammox route was predominant in nitrogen removal, while COD oxidation and microbe proliferation played the main role in COD removal.Alkaline co-fermentation of primary sludge and external organic waste (OW) was studied to elucidate the influence of substrate ratios and long-term system robustness and microbial community dynamics using batch and semi-continuous reactors. Volatile fatty acid (VFA) production increased with increasing OW fraction in the substrate due to synergistic effects of co-degradation. VFA production at pH 10 increased up to 30,300 mgCOD/L (yield of 630 mg COD/gVSfed) but reduced over time to ≈10,000 mgCOD/L. JNK inhibitor II Lowering pH to 9 led to the restoration of VFA production with a maximum of 32,000 mg COD/L (676 mg COD/g VSfed) due to changes in microbial structure. VFA was composed mainly of acetic acid, but propionic acid increased at pH 9. The microbial community was dominated by Bacillaceae (34 ± 10%) and Proteinivoracales_uncultured (16 ± 11%) at pH 10, while Dysgonomonadaceae (52 ± 8%) was enriched at pH 9. The study demonstrated a zero-waste strategy that turns organic wastes into bio-based products.
