Gut microbiota and microbial factors regulate the pathogenesis of nonalcoholic fatty liver disease (NAFLD) in patients with obesity and metabolic abnormalities, but little is known about their roles in nonobese NAFLD. Expansion of Escherichia is associated with NAFLD pathogenesis. We aimed to investigate the pathogenic role of Escherichia fergusonii and its products in the development of nonobese NAFLD.

We characterized the intestinal microbiome signature in a cohort of NAFLD patients and healthy controls by 16S ribosomal RNA sequencing. The role of E fergusonii was estimated in rats after 16 weeks of administration, and features of NAFLD were assessed. E fergusonii-derived microRNA-sized, small RNAs (msRNAs) were analyzed by deep sequencing.

We detected an expansion of Escherichia_Shigella in NAFLD patients compared with healthy controls, and its increase was associated with disease severity independent of obesity. E fergusonii, a member of the genus Escherichia, induced the development of nonobese NAFLD characterized by hepatic steatosis and hepatocyte ballooning in rats without obesity. It disturbed host lipid metabolism by inhibiting hepatic lipid β-oxidation and promoting de novo lipogenesis. We also showed that E fergusonii caused the development of hepatic inflammation and fibrosis in a sizable fraction of animals at an advanced stage of NAFLD. Mechanistically, E fergusonii-derived msRNA 23487 down-regulated host hepatic peroxisome proliferator-activated receptor α expression, which could contribute to lipid accumulation in the liver.

These results suggest that E fergusonii promotes the pathogenesis of steatohepatitis and fibrosis in nonobese rats by secreting msRNA 23487, and it might be a potential biomarker for predicting steatohepatitis in nonobese NAFLD.

In contrast, in scenarios where there is no clearly defined point of entry (gradual onset), and the potential hazard gradually transitions to an immediate hazard, the onset point is more ambiguous. In this study, a reasonable hazard onset was proposed for measuring RTs of motorcycle riders to gradual-onset hazards, following which the RTs to abrupt- and gradual-onset hazards were determined and compared. A study on motorcycle rider RTs was conducted in Singapore in which a sample of young male adults (licensed riders with more than one year's effective riding experience, and unlicensed subjects) were equipped with eye tracking glasses and were presented with two pairs of abrupt-/ gradual-onset hazard scenarios on a simple motorcycle simulator. It is, therefore, essential to better understand distraction prevalence at railway crossings to support the use of countermeasures to improve safety outcomes. We conducted field observations at a railway crossing in Brisbane, Australia and its adjacent road intersection to gauge the prevalence of distracted pedestrians. Overall, 585 pedestrians were observed and video recorded during the daytime. The video recordings were coded to estimate the prevalence of distraction behaviour that road users engaged in, factors that affected these proportions, and dynamic changes in behaviour. Compliance with signals was also analysed. Moreover, there is a reverse U-shaped relationship between motorcycle ownership to passenger car ownership ratio (MPC) and the MDC ratio, while a U-shaped relationship exists between relative growth in higher mobility roads as compared to higher accessibility roads (MPA) and the MDC ratio. In the present study, we determined Sigmar1's subcellular localization, transmembrane topology, and function using complementary microscopy, biochemical, and functional assays in cardiomyocytes. Quantum dots in transmission electron microscopy showed Sigmar1 labeled quantum dots on the mitochondrial membranes, lysosomes, and sarcoplasmic reticulum-mitochondrial interface. Subcellular fractionation of heart cell lysates confirmed Sigmar1's localization in purified mitochondria fraction and lysosome fraction. Immunocytochemistry confirmed Sigmar1 colocalization with mitochondrial proteins in isolated adult mouse cardiomyocytes.
