Fragile X syndrome (FXS) is an X-linked genetic disorder caused by the absence of the fragile X mental retardation 1 protein. FXS is the most common inherited cause of intellectual disability and autism spectrum disorder (ASD). Approximately 60% of subjects with FXS present with ASD, and 2% to 4% of individuals diagnosed with ASD have FXS. Most individuals with ASD have a genetic disorder, so detailed molecular testing of individuals with ASD is medically indicated. Deletions of the protein patched homolog 1 antisense (PTCHD1-AS) gene have been associated with ASD. Here, we describe, for the first time, a boy with FXS because of a point mutation in the FMR1 gene and autism, and the latter comorbidity of ASD is likely because of a deletion of PTCHD1-AS. Thus, the observed phenotype of FXS with severe autism symptoms is likely caused by a double hit of genetic mutations.
The case is a 5-year-old boy with phenotypic characteristics of FXS. The psychological assessment based on parent report and the Autism Dilly all affected men, predominantly intellectual disability. However, there are other comorbidities present in a subset of patients, including ASD. We propose that the variable expressivity in FXS could be partially explained by the additive effect of a second genetic mutation that increases the individual susceptibility to the unique phenotypic findings, as is the case of the patient described here.
To compare the effectiveness of the Healthy Caregivers-Healthy Children (HC2) phase 1 (2011-2014) and 2 (2015-2018) child care center (CCC)-based obesity prevention intervention(s) on child dietary practices and body mass index percentile (PBMI) outcomes over 2 years. Phase 1 was implemented via a university-based research team, and phase 2 was delivered via a train-the-trainers approach (university-based research team trains preschool-based coaches, who in turn train CCC teachers to implement and disseminate HC2).
Phase 1 and 2 were both cluster randomized controlled trials of the HC2 obesity prevention intervention. Phase 1 was composed of 1224 children in 28 CCCs (12 intervention and 16 control). We aimed to identify the incidence, outcome, overall condition, and genetic features of patients affected with FA in Lebanon to optimize management, identify the most common genes, describe new mutations, and offer prenatal diagnosis and counseling to the affected families. Over a period of 17 years, 40 patients with FA were identified in 2 major diagnostic laboratories in Lebanon. Information was obtained on their clinical course and outcome from their primary physician. DNA was available in 20 patients and was studied for underlying mutations. FANCA seemed to be the most frequent genetic alteration and 2 novel mutations, one each in FANCA and FANCG, were identified. Nine patients developed various malignancies and died. However, long-haul travel is also associated with impaired neuromuscular function. An athlete's lower body neuromuscular status is frequently assessed and monitored using countermovement jump (CMJ). This study assessed the impact of westbound trans-meridian travel on changes in CMJ performance in highly trained aerobic athletes. Highly trained male rowers (n = 21, 23.7 ± 1.4 years, 1.91 ± 0.07 m, 86.9 ± 9.9 kg) undertook a westbound flight incurring 22 hours of flight time with 30 hours of total travel time across 9 time zones. Athletes completed a single set of 6 loaded CMJ repetitions before and after travel with performance measured using a digital optical encoder attached to a 20 kg barbell. Each CMJ repetition was assessed for mean concentric velocity, jump height (JH), eccentric displacement, JHdip ratio, mean power, and mean eccentric velocity, with comparisons made between mean set changes before and after travel. Changes were compared using a one tail paired t-test and characterized using Cohen's d (95% confidence interval) effect sizes. Significance was accepted at p ≤ 0.05. Small-to-moderate changes were observed in all variables following westbound travel. These changes may be because of retention of high training loads before travel resulting in the athletes being in a fatigued state, whereas travel time worked as a forced recovery period. Reductions in CMJ performance are likely following westbound travel, however, may be affected by training fatigue before travel that may confound CMJ performance as a monitoring metric.Lockie, RG, Moreno, MR, and Dawes, JJ. The standing broad jump (SBJ) also provides a measure of lower-body power, requires less equipment, and could be practical for training staff with limited resources. There has been no analysis of the relationships between the VJ and SBJ in law enforcement recruits to ensure they measure similar qualities. This research note investigated the relationships between the VJ and SBJ in recruits from one agency. The sample included 71 men and 23 women who completed the VJ and SBJ before academy. Independent samples t-tests compared the sexes in both jump tests. Pearson's correlations calculated relationships between the VJ and SBJ variables (VJ height, height relative to body mass, peak power and power-to-body mass ratio calculated from VJ height; SBJ distance and distance relative to body mass) in all recruits combined, men, and women.
