It has been proposed that at stalled replication forks, monoubiquitinated-FANCD2 serves to recruit DNA repair proteins that contain ubiquitin-binding motifs. Here we have reconstituted the FA pathway in vitro to study functional consequences of FANCIFANCD2 monoubiquitination. We report that monoubiquitination does not promote any specific exogenous proteinprotein interactions, but instead stabilizes FANCIFANCD2 heterodimers on dsDNA. This clamping requires monoubiquitination of only the FANCD2 subunit. We further show that purified monoubiquitinated FANCIFANCD2 forms filament-like arrays on long dsDNA using electron microscopy. Our results reveal how monoubiquitinated FANCIFANCD2, defective in many cancer types and all cases of FA, is activated upon DNA binding. © 2020, Tan et al.The hippocampus supports memory encoding and retrieval, which may occur at distinct phases of the theta cycle. These processes dynamically interact over rapid timescales, especially when sensory information conflicts with memory. The ability to link hippocampal dynamics to memory-guided behaviors has been limited by experiments that lack the temporal resolution to segregate encoding and retrieval. Here, we simultaneously tracked eye movements and hippocampal field potentials while neurosurgical patients performed a spatial memory task. Phase-locking at the peak of theta preceded fixations to retrieved locations, indicating that the hippocampus coordinates memory-guided eye movements. In contrast, phase-locking at the trough of theta followed fixations to novel object-locations and predicted intact memory of the original location. Theta-gamma phase amplitude coupling increased during fixations to conflicting visual content, but predicted memory updating. Hippocampal theta thus supports learning through two interleaved processes strengthening encoding of novel information and guiding exploration based on prior experience. © 2020, Kragel et al.A prevalent model is that sharp-wave ripples (SWR) arise 'spontaneously' in CA3 and propagate recent memory traces outward to the neocortex to facilitate memory consolidation there. Using voltage and extracellular glutamate transient recording over widespread regions of mice dorsal neocortex in relation to CA1 multiunit activity (MUA) and SWR, we find that the largest SWR-related modulation occurs in retrosplenial cortex; however, contrary to the unidirectional hypothesis, neocortical activation exhibited a continuum of activation timings relative to SWRs, varying from leading to lagging. Peptides with the right properties, for example Antimicrobial peptides (AMPs), can disrupt this protective barrier by formation of leaky pores. Unfortunately, matching peptide properties with their ability to selectively form pores in bacterial membranes remains elusive. In particular, the proline/glycine kink in helical peptides was reported to both increase and decrease antimicrobial activity. We used computer simulations and fluorescence experiments to show that a kink in helices affects the formation of membrane pores by stabilizing toroidal pores but disrupting barrel-stave pores. The position of the proline/glycine kink in the sequence further controls the specific structure of toroidal pore. Moreover, we demonstrate that two helical peptides can form a kink-like connection with similar behavior as one long helical peptide with a kink. Abstract L’escargot Cornu aspersum, originaire des côtes méditerranéennes d’Afrique du Nord, est largement répandu sur la plupart des continents et souvent invasif dans ses aires d’introduction récente. Cette espèce peut contribuer à l’expansion géographique de parasites comme démontré pour Brachylaima spp. Ces trématodes cosmopolites peuvent représenter une menace pour la santé comme en Australie où Brachylaima cribbi parasite l’espèce humaine. Dans cette étude, nous démontrons pour la première fois la présence de Brachylaima spp. dans deux populations françaises de C. aspersum, à Thorigné-Fouillard (Ille-et-Vilaine) et Arçais (Deux-Sèvres), avec une prévalence totale de 10.4 % (Thorigné-Fouillard) et 73.3 % (Arçais), et une intensité de métacercaires en moyenne trois fois plus importante à Thorigné-Fouillard (37) qu’à Arçais (11). Cornu aspersum peut jouer le rôle de premier et de second hôte intermédiaire, comme démontré à Arçais. La morphométrie des métacercaires, en particulier la grande taille corporelle d’environ 2 mm de long, suggèrent qu’il ne s’agit pas d’espèces de Brachylaima déjà décrites chez C. aspersum (B. cribbi en Australie et B. aspersae, B. llobregatensis et B. mascomai en Europe). L’analyse moléculaire, basée sur 28S et COI, suggère la présence de deux espèces dans notre étude, dont l’une est probablement Brachylaima mesostoma, un parasite intestinal d’oiseaux passeriformes décrit en Europe Centrale. Nous soulignons le besoin de recherche future pour identifier les espèces de Brachylaima présentes en France et mesurer le risque associé à la consommation d’escargots terrestres prélevés en milieu naturel.
