The present study describes a comprehensive investigation of the spectroscopic characteristics, stability and in vitro antioxidant and cytotoxic properties of the Flavin MonoNucleotide (FMN) and Flavin Adenine Dinucleotide (FAD) in Dextran70 (Dx70) and Dx70/phospatidylcholine (PC) biomimetic systems by means of the UV-Vis absorption, fluorescence spectroscopy, chemiluminescence and Neutral Red assay. The affinity of FMN, FAD and the precursor riboflavin (RF) to an unsaturated phospholipid bilayer model as well as the location of the probes within the lipid bilayer were assessed from united-atom molecular dynamics simulations carried out on an unsaturated phospholipid bilayer model system, and the theoretical and experimental characterization of the two probes within biomembranes was complemented with the light microscopy survey of the cell morphology of L929 fibroblast cells cultivated in the presence of various dosage of FAD/FMN. In lipid bilayers, FMN/FAD resulted in a noticeable improvement of the antioxidant activity (the scavenging of reactive oxygen species up to 40%) and a significant effect on cellular viability in the L929 fibroblast cells. The results are important in the oxidative stress process concerning the redox reactions of flavins in humans as well as in further studies on different systems belonging to the category of flavoenzymes/flavoproteins, required for cellular respiration.The study was conducted to (1) apply advanced synchrotron radiation-based technique-SR-FT/vIMS to detect chemical profiles that are related to protein and carbohydrate biopolymers, (2) quantify the relationship between spectral features and nutrient utilization and bioavailability of newly developed carinata and canola seed lines. The molecular spectral features of these seed lines were analyzed using SR-FT/vIMS with both univariate and multivariate spectral analysis techniques. The results showed that the inherent structural characteristics of new carinata and new canola seeds could be detected by SR-FT/vIMS. The univariate molecular spectral analysis showed differences in absorption intensities (peak heights and areas) of functional groups related to protein and carbohydrate molecular structures, while multivariate molecular spectral analysis without any spectral parameterization results showed similar protein and carbohydrate structure between new carinata and new canola seeds. Based on both, univariate and multivariate analysis, there were some differences between carinata seeds and canola seeds in protein and Carbohydrate (CHO) structure spectral characteristics, but these differences were not distinguishable in CLA and PCA plots regardless the color seed coat when using original spectral without spectral parameterization. However, sulfa drugs have raised worldwide concerns because of their adverse impact on human health. In this study, two sulfonamides, sulfametoxydiazine (SMD) and sulfamonomethoxine (SMM), were selected to explore the binding modes with human serum albumin (HSA). The spectroscopic approaches revealed that SMD or SMM could spontaneously enter into the binding site I of HSA through hydrogen bond interactions and van der Waals forces, and that SMD exhibited much stronger binding affinity toward HSA than SMM at different temperatures (p less then 0.01, n = 3). The binding constants for SMD-HSA and SMM-HSA were determined to be (8.297 ± 0.010) × 104 L·mol-1 and (1.178 ± 0.008) × 104 L·mol-1 at 298 K, respectively. The interaction of SMD or SMM to HSA induced microenvironmental and conformational changes in HSA, where SMD had a greater effect on the α-helix content of HSA. Results from molecular docking implied that the amino acid residues of HSA, such as Arg222, Ala291 and Leu238, played key roles in the sulfonamide-HSA binding process. Meanwhile, hydrogen bonds might be a key factor contributing to the binding affinity of sulfa drugs and HSA. Additionally, the combined use of SMD and SMM led to an obvious variation in Ka values of binary systems (p less then 0.01, n = 3). These findings might be helpful to understand the biological effects of sulfonamides in humans.Raman mapping microspectroscopy was used as an advantageous high spatial resolution method for detailed assessment of the structure of radiation-induced halos in bituminous coal (Upper Paleozoic) with numerous inclusions of uraninite and coffinite. The uranium content in inclusions in the samples studied ranged from 40 to 50 wt%. Raman structural parameters such as full width at half maximum, the positions of the D-band and G-band peaks and their area ratios were calculated, and these correlated well with vitrinite reflectance. Using linear profiles across the entire halos, changes in the degree of radiolytic alteration of coal matter caused by ionising radiation resulting from the decay of uranium and its daughter products, were described. Using micro-ATR-FTIR, oxidative radiolytic alteration of coal was identified in halos, with oxidisation to alcohols, ketones and carboxyl groups, which were then converted to COO- ions bound to the cations present, including UO22+. From our data, we conclude that the conversion of the original coal to a type of anthracite occurred during the process of CH and CC cleavage, dehydroaromatisation of naphthenic rings and oxidation resulting in the transformation of aliphatic structures into aromatic clusters. Generally, radiolytic alteration increased the structural organisation of coal. Although its vital importance, few treatments are yet available when a hepatic failure occurs, and hence, the use of stem cells has arisen as a possible solution for both human and veterinary medicines. Previous studies have shown the existence of hepatic progenitor cells in human fetuses that were positive for EpCAM and NCAM. There is limited evidence, however, further identification and characterization of these cells in other species. Considering the similarity between dogs and humans regarding physiology, and also the increasing importance of developing new treatments for both veterinary and translational medicine, this study attempted to identify hepatic progenitor cells in canine fetal liver. For that, livers from canine fetuses were collected, cells were isolated by enzymatic digestion and cultured. Cells were characterized regarding morphology and expression of EpCAM, NCAM, Nestin, and Thy-1/CD90 markers.
