The enhanced HO• and O2-• formation on the elevated voltage MAOed coatings caused serious break of the cell walls and plasma membranes of S. aureus. In parallel, the enhanced Zn2+ the coatings. (3) The enhanced Zn2+ release and ROS formation on the coatings killed S. aureus by inducing serious break of their cell walls and plasma membranes. This effect in combination of un-attenuated osteoblast proliferation endowed the moderate Zn doped coatings with enhanced osseointegration in S. aureus-infected rat tibias. INTRODUCTION Peri-miniscrew inflammation is one of the causes of orthodontic miniscrew failure. OBJECTIVE The aim of this study was to correlate the quantification throughout autofluorescence images and PCR microbiologic count of biofilm retained around orthodontic miniscrew and the presence of Porphyromonas gingivalis. MATERIALS AND METHODS Forty miniscrews used for orthodontic treatment were evaluated during orthodontic treatment, collected from patients and divided into two groups health and inflamed miniscrews. To be considered inflamed, the samples should present loss of stability checked by periotest®, clinical presence of mucositis, red aspect of the gum or bleeding around the miniscrew. Immediately after removal of the miniscrews, they were photographed using a macro 100 lens and a Pentax camera coupled to a fluorescent equipment - Qscan (AioBio - Korea) with 405 nm excitation wavelength and a blue band filter. A microbiologic sample was collected with a sterile microbrush scrubbed around the miniscrew tread. Fluorescent images were analyzed with ImageJ software to quantify fluorescent intensity and fluorescent area and microbiological samples were submitted to UCF count for total contamination and q-PCR assay to quantify Porphyromonas gingivalis. RESULTS The results showed a good correlation between UFC count and fluorescent intensity and PCR/fluorescent area. The health miniscrews presented less fluorescent intensity and lower UFC count when compared to inflamed miniscrews. q-PCR analysis showed a higher number of P. gingivalis contamination around inflamed miniscrews. CONCLUSION Quantification of biofilm retained by miniscrew by images of auto-fluorescence is a simple and reliable method with great potential for clinical use to monitory inflammation around miniscrew and risk of loss. V.The combination of multiple sessions of Photodynamic therapy (PDT) and surgery have been used to treat extensive superficial lesion of squamous cell carcinoma in situ. Methyl aminolevulinate (MAL) mediated PDT was applied to reduce the tumoral area and a small surgical removal was performed to complete elimination of the lesion. The reduction of the tumor area avoided the need for a skin graft application as well as possible postoperative complications, offering a more favorable cosmetic outcome. Two-years of follow-up showed no recurrence. The case is interesting and demonstrate potentialities for the combined use of PDT and surgery. V.Photodynamic Therapy (PDT) is a challenging treatment method in which a target region is irradiated with a light source having an appropriate wavelength. Due to the increment of antibiotic resistant bacteria, PDT has also become a salient method for infection treatments. Ipatasertib The light source, photosensitizer and oxygen are the crucial components of PDT. Besides, the amount and the location of singlet oxygen gives information about the effectiveness of PDT. The quantitative evolution of singlet oxygen is a gold standard for the real time monitoring of the treatment efficiency during PDT. In the proposed study, the effect of radiation modes on PDT is investigated with singlet oxygen explicit dosimetry (SOED) and singlet oxygen luminescence dosimetry (SOLD) methods. For this purpose, super pulse and pulse radiation modes are applied for antimicrobial PDT (AmPDT). Five in vitro experiments were carried out to investigate the effect of radiation mode. According to the achieved results, super pulse mode provides 3-10% more singlet oxygen concentration and 2-5% more bacteria (Staphylococcus Aureus) death (necrosis and apoptosis) than pulse mode. Furthermore, radiation mode effect on instantaneous and cumulative singlet oxygen concentration is considered in the experiments. It is demonstrated that the singlet oxygen concertation measured by SOED and SOLD methods are coherent. Thus, the SOED method can be used for real-time singlet oxygen measurements during PDT. V.BACKGROUND To evaluate retinal and choriocapillaris (CC) vessel density in macular region in patients affected by adult-onset foveomacular vitelliform dystrophy (AOFVD) using optical coherence tomography angiography (OCTA) METHODS A total forty-four right eyes of 44 AOFVD patients (20 females, 24 males, mean age 69.17 ± 11.57 years) divided in 3 stages (vitelliform, pseudohypopyon and vitelliruptive) and 60 normal right eyes of 60 controls (20 females, 40 males, mean age 66.04 ± 6.40 years) were included in this prospective study. We evaluated the vessel density of superficial capillary plexus (SCP), deep capillary plexus (DCP) and CC in different macular areas (whole image, parafovea and fovea). We also analyzed the subfoveal choroidal thickness (SFCT) with Enhanced Depth Image (EDI)-OCT. RESULTS The vessel density of SCP and of DCP did not differ between patients and controls in all macular sectors.
