All rights reserved.INTRODUCTION The VICTORY AF Study was designed to evaluate the risk of procedure and/or device related strokes in patients with PersAF on warfarin undergoing ablation with a Phased RF System. METHODS The VICTORY AF trial was a prospective, multicenter, single arm, investigational study. PersAF patients on VKA without major structural heart disease or history of stroke/TIA undergoing Phased RF ablation for AF were included. The primary outcome was the incidence of procedure and/or device related stroke within 30 days of the ablation by a board certified neurologist's assessment. The secondary outcomes were acute procedural success, 6 month effectiveness (defined as the reduction in AF/AFL episodes lasting ≥10 minutes by 48-hour Holter 6-months post-ablation) and number of patients with pulmonary vein stenosis. RESULTS A total of 129 (108 PersAF, 21 long-standing PersAF) patients were treated (mean age 60.6±7.7; 79.8% male, 54.3% CHA2Ds2-VASc score ≥2). Two non-disabling strokes were reported (1.6%); one prior to discharge and the second diagnosed at the 30-day visit. Due to slow enrollment the study was terminated prior to reaching the 95% one-sided upper confidence boundary for stroke incidence. Acute procedural success was 93.8%, and at 6 months, 72.8% of patients demonstrated ≥90% reduction in AF burden, 78.9% were off all AADs. There were no patients with PV stenosis of >70%. CONCLUSIONS VICTORY AF demonstrated a 1.6% incidence of stroke in PersAF undergoing ablation with a phased RF system which did not meet statistical confidence due to poor enrollment. The secondary outcomes suggest comparable efficacy to phased RF in the TTOP trial. Rigorous clinical evaluation of the stroke risk of new AF ablation technologies as well as restriction to VK-antagonist anti-coagulation appear to be unachievable goals in a clinical multi-center IDE trial of AF ablation in the current era. All rights reserved.Ivermectin (IVM) is one of the most widely used antiparasitic drugs worldwide and has become the drug of choice for anthelmintic and tick treatment in beef cattle production. It is known that pharmacokinetic parameters are fundamental to the rational use of a drug and food safety and these parameters are influenced by different factors. The aim of this study was to evaluate the pharmacokinetic profile of IVM in Bos indicus, Bos taurus, and crossbreed cattle (B. indicus × B. taurus) kept under same field conditions and the possible impacts of sex and IVM formulation (1% and 3.15%). It was observed that IVM concentration was significantly affected by breed. The plasma concentrations of IVM, AUC, Cmax , and t1/2β were significantly higher in B. indicus compared to B. taurus. Crossbreed animals showed an intermediate profile between European and Indian cattle. No alteration in pharmacokinetics parameters was detected when comparing different gender. Concerning the pharmacokinetic data of IVM formulation, it was verified that Tmax , AUC, and t1/2β were higher in 3.15% IVM animals than those from 1% IVM formulation. The existing methods for evaluation of maturation either focus on the conversion of modified precursors or detect the mature miRNA. While the former is not applicable to native pre-miRNA the latter approach underestimates maturation when both non-matured and matured miRNA molecules are subject to cleavage. We present a set of two orthogonally labeled FIT PNA probes that distinguishes between cleaved pre-miRNA and the mature miRNA duplex. The probes allow monitoring Dicer-mediated miR21 maturation and assaying Ago2-mediated unwinding of the miR21 duplex. A two-channel fluorescence readout enables measurements in real-time without the need for specialized instrumentation or further enzyme mediated amplification. KGaA, Weinheim.BACKGROUND Recent studies suggest that vitamin D deficiency is associated with intestinal dysfunctions, but the underlying mechanism remains unclear. This study aimed to investigate whether maternal vitamin D deficiency increases intestinal permeability in offspring and its related mechanism. METHODS Timed-pregnant mice were fed with either a standard chow diet (SC) or a vitamin D-deprived chow diet (VD-) 6 weeks prior to breeding and kept on the same diet until the end of gestation. All offspring were fed an SC for 3 weeks after weaning and then observed for effects associated with maternal vitamin D deficiency. RESULTS Maternal vitamin D deficiency increased intestinal permeability in offspring, which corresponded with the decreased expression of the tight junction protein claudin-1. Maternal vitamin D deficiency also repressed the messenger RNA expression of wingless/integrated family member 3a (Wnt3a) and the protein expression of nuclear β-catenin. The decreased Wnt3a gene expression in male was concurrent with the changes in histone H4 acetylation at either promoter or coding regions.
