Nothing but the truth? Effects of acting on the credibility with the crosswise design.
Small molecules are immunochemically classified as hapten that lacking of at least two epitopes, usually using competitive format for establishing immunoassays. However, theoretically, noncompetitive immunoassay format is more sensitive and has a wider analytical range. In the present study, a novel hapten of halofuginone was synthesized and used to produce a monoclonal antibody (mAb). By analyzing the binding kinetics, we found that the affinity of analyte-enzyme to mAb was much greater than that of analyte, which could result in a low sensitivity of competitive assay format. Based on this, we established a novel noncompetitive immunoassay by using a replacement approach. The noncompetitive format has obvious advantages in sensitivity and analytical range, which promoted approximately 3.5- and 5-fold, respectively, compared to the competitive immunoassay. Ultimately, the newly designed noncompetitive immunoassay in this work will provide insights as well as alternative method to traditional small molecule competitive assays. This study evaluated the effects of treatments of squid pen chitooligosaccharide (COS) or epigallocatechin gallate (EGCG) or COS/EGCG mixture (11, w/w) at different concentrations (0, 200, and 400 mg/kg) on the discoloration and quality changes in yellowfin tuna slices stored at 4 °C for 12 days. Tuna slices added with 200 and 400 mg/kg of COS (C2 and C4, respectively) showed the lowest reduction in oxymyoglobin and a* value (redness) ascertained by the lower metmyoglobin formation than other samples. Additionally, C2 and C4 samples showed a lower total viable count and TBARS value than the remaining samples. EGCG alone and its mixture with COS exhibited lower efficacy in retaining the quality loss than COS alone. COS at both levels effectively reduced the metMb formation. It maintained the redness with sensory acceptability of slices up to 9 days, and C4 sample prolonged shelf-life for 12 days based on the microbiological limit.Pomelo peel as a by-product from pomelo consumption is rich in various nutrients and functional compounds, while most of the by-product is disposed as wastes. The utilization of pomelo peels could not only result in valued-added products/ingredients, but also reduce the environmental threats. By mainly reviewing the recent articles, pomelo peels could be directly used to produce candied pomelo peel, tea, jams, etc. Additionally, functional components (essential oils, pectin, polyphenols, etc.) could be extracted from pomelo peels and applied in food, pharmaceutical and chemical fields. The extraction methods exerted important influences on the composition, physicochemical properties, bioactivities and structures of the resultant fractions. Furthermore, pomelo peel was exploited to make adsorbents, bioethanol, etc. For the future investigations, the functionality- or bioactivity-oriented regimes to recovery valuable components from pomelo peel should be developed in an economic, effective and eco-friendly way and their applicability in large-scale production should be addressed. The aim of research was to determine polyphenols bioaccessibility and antioxidant properties of thermally-treated skimmed goat milk enriched with sunflower bee-collected pollen through in vitro digestion. HPLC analysis confirmed that pollen-enriched milk contained flavonols as the main phenolic fraction (80.7-76.2%) followed by phenolic acids (14.2-17.4%). CX-4945 Casein Kinase inhibitor Among individual compounds quercetin-3-O-glucoside (155.1-197.2 μg/L) and p-coumaric acid (29.5-30.7 μg/L) were the main quantified flavonols and phenolic acids, respectively. After digestion of milk/pollen sample, total polyphenols recovery was 30.71% with higher phenolic acids recovery (40.1%) compared to flavonols (28.3%) indicating strong interactions between caprine milk casein micelles and pollen polyphenols. Applied antioxidant assays (phosphomolybdenum, ABTS•+scavenging activity and ferrous-ion-chelating capacity) have confirmed complexity of prepared product- it had high ability to quench ABTS•+ radicals and to form chelating complexes with Fe2+ ions. Digestion provoked 20% reduction in total antioxidant capacity compared to the initial sample. TTSG milk/pollen powder could be good functional ingredient.Iron-based metal-organic framework, NH2-MIL-101(Fe), was doped with different dosages of cobalt phthalocyanine nanoparticles (CoPc) to synthesize a series of NH2-MIL-101(Fe)@CoPc nanocomposites. The NH2-MIL-101(Fe)@CoPc nanocomposites were then employed to construct novel impedimetric aptasensors for the detection of ochratoxin A (OTA). Combining the intrinsic advantages of NH2-MIL-101(Fe) (highly porous structure and excellently electrochemical activity) and CoPc (good physiochemical stability and strong bioaffinity), the NH2-MIL-101(Fe)@CoPc nanocomposites show promising properties, which are beneficial for immobilizing OTA-targeted aptamer strands. Amongst, the developed impedimetric aptasensor based on NH2-MIL-101(Fe)@CoPc61, prepared using the mass ratio of NH2-MIL-101(Fe)CoPc of 61, exhibits the best amplified electrochemical signal and highest sensitivity for detecting OTA. The detection limitation is 0.063 fg·mL-1 within the OTA concentration of 0.0001-100 pg·mL-1, accompanying with high selectivity, good reproducibility and stability, acceptable regenerability, and wide applicability in diverse real samples. Consequently, the proposed sensing strategy can be applied for detecting OTA to cope with food safety.Broadband acoustic resonance dissolution spectroscopy (BARDS) is a novel method that can be used for the analysis of food-based powders, which are mainly characterized by their composition and particle morphology. This study aimed to evaluate BARDS for the compositional analysis of food powders. The changes in the BARDS spectra due to the changes in composition and particle morphology of fifteen salt mixtures (constituting of NaCl, KCl, and MgCl2) in five particle size ranges were comprehensively studied. Moreover, different regression methods were utilized to estimate each mixture component content. The results revealed that the average time-frequency spectra of each mixture in a certain particle size class were highly distinct and allowed discrimination from others. The unique spectra of each salt mixture originated from the specific dissolution rate and degassing effect of each constitutive compound. Finally, the accurate prediction of each mixture component content confirmed the consistency and efficiency of the method.
