Despite intensive research, a known treatment for MS disease yet to be introduced. Thus, the development of novel and safe medications needs to be considered for the disease management. Application of mesenchymal stem cells (MSCs) as an emerging approach was recruited forthe treatment of MS. MSCs have several sources and they can be derived from the umbilical cord, adipose tissue, and bone marrow. Chemokines are low molecular weight proteins that their functional activities are achieved by binding to the cell surface G protein-coupled receptors (GPCRs). Chemokine and chemokine receptors are of the most important and effective molecules in MSC trafficking within the different tissues in hemostatic and non-hemostatic circumstances. Chemokine/chemokine receptor axes play a pivotal role in the recruitment and oriented trafficking of immune cells both towards and within the CNS and it appears that chemokine/chemokine receptor signaling may be the most important leading mechanisms in the pathogenesis of MS. In this article, we hypothesized that the chemokine/chemokine receptor axes network have crucial and efficacious impacts on behavior of the MSCs, nonetheless, the exact responsibility of these axes on the targeted tropism of MSCs to the CNS of MS patients yet remained to be fully elucidated. Therefore, we reviewed the ability of MSCs to migrate and home into the CNS of MS patients via expression of various chemokine receptors in response to chemokines expressed by cells of CNS tissue, to provide a great source of knowledge.
Utilization of complementary medicines (CMs) amongst children with eosinophilic esophagitis (EoE) in Australia is high. Carers' beliefs, perceptions and use of CM can influence the decision to use CM in children in their care. This study explores the factors influencing the use of CM for a child's EoE when the carer also uses CM.
Carers of children aged 0-18 years with EoE participated in a national cross-sectional online survey, conducted in Australia between September 2018 and February 2019. Data analysis included bivariate analysis, Cramer's V, backwards stepwise logistic regression and binomial logistic regression.
Of the 181 total survey responses, 165 (91.2%) respondents indicated they had utilized some form of CM for themselves. Children whose carer had used some form of CM for themselves were more likely to have used CM than children whose carer had not used CM (OR 4.6; p=0.001). Of the CM self-using carers, 125 (75.8%) had also chosen to utilize CM for their child's EoE. Use of CM in children w undertaken.
High CM self-use amongst carers is associated with their decision to also use CM for their child's EoE, a concern given the absence of evidence for CM's safety and efficacy in this population. CM use in this population warrants further attention. Effective conventional medicines for EoE are limited and utilization of CM amongst children with EoE in Australia is high. The 1% yearly population decline indicates that the cumulative threats are already beyond sustainable limits for the SLEB. However, a potential threefold increase in shipping traffic is expected within its critical habitat in the coming years resulting from proposed port-industrial projects in the Saguenay River. Current data indicate that SLEB typically use multiple sectors within their summer range, likely leading to differential VUN exposure among individuals. The degree of displacement and spatial mixing among habitats are not yet well understood but can be simulated under different assumptions about movement patterns at the individual and population levels. Here, we propose using an agent-based model (ABM) to explore the biases introduced when estimating exposure to stressors such as VUN, where individual-centric movement patterns and habitat use are derived from diffefic.In recent years, dense Alexandrium ostenfeldii blooms have been reported in different coastal areas. In this study, we report for the first time the occurrence of A. ostenfeldii blooms associated with the detection of paralytic shellfish toxins (PSTs) in the Peruvian scallop (Argopecten purpuratus) from Paracas Bay. Alexandrium ostenfeldii blooms occurred at the end of summer and early fall, after the increase of riverine input and under stratified conditions following a decrease in wind velocity. The highest abundances occurred during warm sea surface temperatures (18-27 °C). High PST concentrations that exceed the maximum permissible level (800 μg STX eq. kg-1) occurred even under low A. ostenfeldii abundances (20 × 103 cells l-1). Our results contribute to a better understanding of the dynamics of A. ostenfeldii in coastal systems influenced by riverine inputs and upwelling and can be used to improve monitoring programs and allow the implementation of mitigation measures along the Peruvian coast.The Japanese government approved a plan to discharge Fukushima Daiichi Nuclear Power Plant Accident contaminated water (FDNPPACW) into the Pacific Ocean. It immediately caused a new wave of global concern and anxiety. To assess this matter, this work briefly reviewed the dispersion of FDNPPA-derived radionuclides in the Pacific Ocean in the past and the resulting impacts on marine biota. Combining the drafted plan of discharging FDNPPACW and the public's concerns, 5 points, including (1) the detailed plan of discharging FDNPPACW, (2) the isotopes left in the advanced liquid processing system (ALPS)-treated water and their amounts, (3) the stability of the Kuroshio Extension, (4) the fates and transports of the main radionuclides (left in the ALPS-treated water) in North Pacific seawater, (5) and bioaccumulations and the ecological half-lives of the main radionuclides (left in the ALPS-treated water) in marine biota in the North Pacific, remain to be known to understand the impacts of discharging FDNPPACW into the Pacific Ocean.Trace metals in Laptev and East Siberian Sea sediments were analyzed to examine the element distribution pattern and to assess sediment contamination using contamination indices. The maximum Cr, Zn, Ni, Cu, and As concentrations occurred in the deeper parts of the East Siberian and Laptev Seas and the estuarine shelves of the Lena and Yana Rivers, and a high Cd content was observed in the sediment of the eastern part of the East Siberian Sea. The increased Cd concentration could be a result of the large number of phytoplankton supplied by the highly productive waters of the Bering Sea and the sorption capacity of Cd precipitation on Fe and Mn oxyhydroxides in seawater. The mean enrichment factors and geoaccumulation index values for Cr, Ni, Cu, Zn, Pb, and Cd showed no contamination by these metals at any sampling locations; however, slight As enrichment was found, possibly originating from natural sources.Various methods have been used to induce the neuronal differentiation of marrow mesenchymal stem cells (MSCs). However, the limited induction efficiency of cells in vitro has restricted their use. Therefore, identifying a simple and efficient treatment method is necessary. Dendrobium officinale is an important traditional Chinese medicine, and its main component, polysaccharides, has many pharmacological activities. However, the effects of D. officinale polysaccharide (DOP) on the neuronal differentiation of bone marrow mesenchymal stem cells (BMSCs) and treatment of ischaemic stroke remain unknown. We found that DOP promoted the neuronal differentiation of BMSCs by increasing the expression levels of neural markers, and the optimal concentration of DOP was 25 μg/mL. Additionally, the Notch signalling pathway was inhibited during the neuronal differentiation of BMSCs induced by DOP, and this effect was strengthened using an inhibitor of this pathway. The Wnt signalling pathway was activated during the differentiation of BMSCs, and inhibition of the Wnt signalling pathway downregulated the expression of neuronal genes. Furthermore, the transplantation of neuron-like cells induced by DOP improved neuronal recovery, as the brain infarct volume, neurologic severity scores and levels of inflammatory factors were all significantly reduced in vivo. In conclusion, DOP is an effective inducer of the neuronal differentiation of BMSCs and treatment option for ischaemic stroke.
Current standard of care imaging, cytology, or cystic fluid analysis cannot reliably differentiate malignant from benign pancreatic cystic neoplasms. This study sought to determine if the metabolic profile of cystic fluid could distinguish benign and malignant lesions, as well as mucinous and non-mucinous lesions.
Metabolic profiling by untargeted mass spectrometry and quantitative nuclear magnetic resonance was performed in 24 pancreatic cyst fluid from surgically resected samples with pathological diagnoses and clinicopathological correlation.
(Iso)-butyrylcarnitine distinguished malignant from benign pancreatic cysts, with a diagnostic accuracy of 89%. (Iso)-butyrylcarnitine was 28-fold more abundant in malignant cyst fluid compared with benign cyst fluid (P=.048). Furthermore, 5-oxoproline (P=.01) differentiated mucinous from non-mucinous cysts with a diagnostic accuracy of 90%, better than glucose (82% accuracy), a previously described metabolite that distinguishes mucinous from non-mucinous cysts. Combined analysis of glucose and 5-oxoproline did not improve the diagnostic accuracy. In comparison, standard of care cyst fluid carcinoembryonic antigen (CEA) and cytology had a diagnostic accuracy of 40% and 60% respectively for mucinous cysts. (Iso)-butyrylcarnitine and 5-oxoproline correlated with cyst fluid CEA levels (P<.0001 and P<.05 respectively). For diagnosing malignant pancreatic cysts, the diagnostic accuracies of cyst size > 3 cm, ≥ 1 high-risk features, cyst fluid CEA, and cytology are 38%, 75%, 80%, and 75%, respectively.
(Iso)-butyrylcarnitine has potential clinical application for accurately distinguishing malignant from benign pancreatic cysts, and 5-oxoproline for distinguishing mucinous from non-mucinous cysts.
