The main objective of the present work was to evaluate the effects of Cd stress in T. arvense using a physiological approach. Four-week-old plants were exposed to increasing concentrations of Cd (0, 10, 50 and 100 μM Cd(NO3)2) for 14 days. Plants were harvested at days 0, 3, 7 and 14 to assess the following parameters shoot and root length and fresh weight; osmolality and conductivity in leaves; chlorophyll a and b contents and photosynthetic efficiency in leaves (young and expanded); water content, Cd accumulation and nutrient imbalances in leaves (young and expanded) and roots. Thlaspi arvense presented a reduction of 27% in the length of roots exposed to 100 μM Cd, and in plants exposed to 50 and 100 μM Cd, a reduction in the fresh weight of both shoots (53 and 58%, respectively) and roots (58 and 63%, respectively) was observed. In general, in plants exposed to the highest concentrations of Cd, a decrease in leaves osmolality and membrane permeability and in water content of both roots and leaves were observed, as well as several nutrient imbalances. As for the photosynthetic parameters, young and expanded leaves responded differently to Cd stress. Exposed young leaves presented no alterations in photosynthetic efficiency or chlorophyll contents, while expanded leaves of 100 μM Cd-treated plants showed a reduction of up to 67% in chlorophyll contents and a concomitant reduction of 30% in photosynthetic efficiency. Overall, Cd-induced senescence in the non-accumulator plant T. arvense exposed to high concentrations of Cd, which was measured as a decrease in several physiological and biochemical parameters and nutrient imbalances. These Cd-induced alterations resulted in lower plant growth, which might have further implications on plant performance at the population level.
Hypoxia-activated prodrugs (HAPs) have the potential for eliminating chemo- and radiation-resistant hypoxic tumour cells, but their activity is often compromised by limited penetration into hypoxic zones. Nitrochloromethylbenzindoline (nitroCBI) HAPs are reduced in hypoxic cells to highly cytotoxic DNA minor groove alkylating aminoCBI metabolites. Avacopan solubility dmso In this study, we investigate whether a lead nitroCBI, SN30548, generates a significant bystander effect through the diffusion of its aminoCBI metabolite and whether this compensates forany diffusion limitations of the prodrug in tumour tissue.
Metabolism and uptake of the nitroCBI in oxic and anoxic cells, and diffusion through multicellular layer cultures, was characterised by LC-MS/MS. To quantify bystander effects, clonogenic cell killing of HCT116 cells was assessed in multicellular spheroid co-cultures comprising cells transfected with cytochrome P450 oxidoreductase (POR) or E. coli nitroreductase NfsA. Spatially-resolved pharmacokinetic/pharmacodynamic (PK/PD) models, parameterised by the above measurements, were developed for spheroids and tumours using agent-based and Green's function modelling, respectively.
NitroCBI was reduced to aminoCBI by POR under anoxia and by NfsA under oxia, and was the only significant cytotoxic metabolite in both cases. In spheroid co-cultures comprising 30% NfsA-expressing cells, non-metabolising cells were as sensitive as the NfsA cells, demonstrating a marked bystander effect. Agent-based PK/PD models provided good prediction of cytotoxicity in spheroids, while use of the same parameters in a Green's function model for a tumour microregion demonstrated that local diffusion of aminoCBI overcomes the penetration limitation of the prodrug.

It has been reported that the cold-induced vasodilation (CIVD) response can be trained using either regular local cold stimulation or exercise training. The present study investigated whether repeated exposure to environmental stressors, known to improve aerobic performance (heat and/or hypoxia), could also provide benefit to the CIVD response.
Forty male participants undertook three 10-day acclimation protocols including daily exercise training heat acclimation (HeA; daily exercise training at an ambient temperature, T
= 35°C), combined heat and hypoxic acclimation (HeA/HypA; daily exercise training at T
= 35°C, while confined to a simulated altitude of ~ 4000m) and exercise training in normoxic thermoneutral conditions (NorEx; no environmental stressors). To observe potential effects of the local acclimation on the CIVD response, participants additionally immersed their hand in warm water (35°C) daily during the HeA/HypA and NorEx. Before and after the acclimation protocols, participants completed hnot contribute to any changes in the CIVD response.Transgenic mice overexpressing human high molecular weight fibroblast growth factor 2 (HMWFGF2) isoforms in osteoblast and odontoblast lineages (HMWTg) exhibit decreased dentin and alveolar bone mineralization, enlarged pulp chamber, and increased fibroblast growth factor 23 (FGF23). We examined if the alveolar bone and dentin mineralization defects in HMWTg mice resulted from increased FGF23 expression and whether an FGF23 neutralizing antibody could rescue the hypomineralization phenotype. HMWTg and VectorTg control mice were given subcutaneous injections of FGF23 neutralizing antibody twice/week starting at postnatal day 21 for 6 weeks. Since Calcitriol (1,25D) have direct effects in promoting bone mineralization, we also determined if 1,25D protects against the defective dentin and alveolar bone mineralization. Therefore, HMWTg mice were given subcutaneous injections of 1,25D daily or concomitantly with FGF23 neutralizing antibody for 6 weeks. Our results showed that HMWTg mice displayed thickened predentin, alveolar bone hypomineralization, and enlarged pulp chambers.
