During the first wave of the COVID-19 pandemic, it was established that otorhinolaryngologists were at a high risk of contracting the infection due to examinations of the upper airways. The aim of this study was to evaluate the impact of the COVID-19 pandemic on mental health among healthcare workers (HCWs) in primary ear-nose-throat (ENT) practices.

This was a cross-sectional questionnaire study among HCWs assessing symptoms of anxiety (Generalized Anxiety Disorder, ten-item scale) and depression (Patients Health Questionnaire, nine-item scale). The survey targeted otolaryngologists and staff in primary private practices in the Capital Region of Denmark during the COVID-19 lock-down in May 2020.

For 30% of the participants, signs were observed of depressive symptoms and 13% had signs of anxiety symptoms. Seventy percent felt sufficiently protected by their available personal protective equipment. Fifty-two percent worried about becoming infected in relation to their work and 56% feared infecting their families.

HCWs in Danish primary ENT practices did not display extraordinary stress reactions during the first wave of the pandemic. Noticeably, participants were concerned about acquiring the infection or transmitting it to their household.

The technique involves performing a skin-sparing mastectomy using Wise pattern incisions and creating a breast mound from the residual lower mastectomy flap. In this case report, we present a 58-year old woman with breast cancer who was not eligible for traditional breast reconstruction, and instead an uncomplicated Goldilocks mastectomy was performed.This review covers congenital hair shaft anomalies, which are conditions affecting hair shaft morphology. Sometimes suspected with the naked eye, often in need of microscopic examination to properly diagnose, these conditions could lead to the discovery of a complex genetic syndrome. Further knowledge is needed in order to establish a diagnosis, approach treatment alternatives and shed light on prognoses, which benefits patients. After transformation, the recombinant MVF-ErbB3II protein was expressed in E. coli BL21 (DE3) and purified using nickel ion affinity chromatography. link2 Subsequently, the purified MVF-ErbB3II protein was used as antigen to immunize rats subcutaneously for induction of anti-MVF-ErbB3IIpcAb. The titer of anti-MVF-ErbB3II pcAb was analyzed by ELISA. The ErbB3 specificity and targeting ability of pcAb were evaluated by Western blotting, immunoprecipitation (IP) and flow cytometry (FCM). Results SDS-PAGE confirmed that MVF-ErbB3II protein was successfully expressed and purified. ELISA showed that the titer of pcAb was 1 024 000. Western blotting, IP and FCM assays showed that the anti-MVF-ErbB3II pcAb not only had good antigen specificity against purified MVF-ErbB3II and native ErbB3 but targeted the ErbB3 dimerization interface. Conclusion The prokaryotic expression and purification of MVF-ErbB3II is successfully achieved, rat anti-MVF-ErbB3II pcAb is prepared and characterized successfully.Objective To investigate the changes of key factors such as placental villi and lymph vessels in chronic venous disease (CVD) during pregnancy. Methods According to the CEAP classification criteria, tissues of CVD patients were collected, and was divided into control group and CVD group. Real-time quantitative PCR was used to detect mRNA level of CD31, D2-4, fms-related tyrosine kinase 1 (FLT-1), placental growth factor (PlGF) and hypoxia-inducible factor-1 (HIF-1α) in placental tissues. Immohistochemical staining was performed to measure the expression level of CD31, D2-40, FLT-1, PlGF, HIF-1α, B-cell lymphoma 2 (Bcl2), Bcl2-related X protein (BAX), caspase 3 and caspase-9. The change in average number of syncytiotrophoblast cell nodes in placental villi was observed by optical microscope and transmission electron microscope. Periodic acid Schiff (PAS) staining was used to detect the PAS-positive substances in placental villi. Results Compared with the control group, the average numbers of placenta villi, sy villi of patients with CVD, coupled with an accelerated apoptosis of villi cells. Meanwhile, CVD patients show an impaired function of placental villi and obstructed gas exchange between mother and infant.Objective To find out indicators for rapid identification of Gram-positive (G+) and Gram-negative (G-) bacteria through transcriptome sequencing of peripheral blood mononuclear cells (PBMCs) and serum liquid-phase chip technology in early sepsis. Methods 35 eligible cases out of 182 sepsis patients in the emergency intensive care unit (EICU) were selected for retrospective analysis. They were divided into G+ group (12 cases) and G- group (23 cases) based on their blood culture results. General characteristics like patients' age, gender, sequential organ failure assessment (SOFA) scores, etc. and other laboratory indexes such as blood routine, IL-6, CRP, procalcitonin(PCT)of these two groups were analyzed. PBMCs were isolated through single density gradient centrifugation. Total RNA was extracted for transcriptome sequencing to find out differential genes. Serum liquid-phase chip technology was performed to detect serum granulocyte colony stimulating factor (G-CSF), granulocyte-macrophage colony stimulating fa applicable in distinguishing G- and G+ bacteria. Conclusion IL-6, IL-1β and IL-10 levels can be used as indicators for early identification of sepsis induced by G+ or G- bacteria.Objective To explore the effect of glucose-6-phsophatase, catalytic subunit (G6PC) on the proliferation, migration and invasion of cervical cancer HeLa cells and the possible molecular mechanism. Methods RNA interfering (RNAi) was used to knockdown the expression of G6PC in HeLa cells, and the silencing effect of protein was confirmed by Western blotting. MTT assay and plate clony formation assay were performed to detect the effect of G6PC knockdown on the proliferation of HeLa cells; scratch healing assay and TranswellTM chamber assay were applied to observe the effect of G6PC knockdown on the invasion and migration abilities of HeLa cells; the tube-formation assay was used to detect the effect of G6PC knockdown on the angiogenesis ability of HeLa cells; the expression levels of epithelial-mesenchymal transition (EMT)-related proteins and AKT/mTOR signaling pathway-related proteins were determined by Western blotting. Results The expression of G6PC was effectively silenced by RNAi technology. G6PC knockdown obviously inhibited the proliferation, migration and angiogenesis of HeLa cells. link3 Meanwhile, G6PC knockdown suppressed the EMT process, the phosphorylation of AKT and mTOR proteins. Conclusion G6PC knockdown can effectively inhibit the proliferation, migration, angiogenesis and EMT process of HeLa cells, which is related to the blocked AKT/mTOR signaling pathway.Objective To investigate the inhibitory effect of betaine (BET) on the proliferation of C4-2B prostate cancer cells and its possible mechanism. Methods C4-2B cells were treated with 0, 100, 200, 400, 600, 800 mmol/L BET. CCK-8 assay was used to assess the cell proliferation, plate cloning formation assay to detect clone formation ability and flow cytometry to evaluate cell apoptosis, and the cell morphological alteration was observed by microscopy. The protein expression of BAX, Bcl2, cleaved caspase 3 (c-caspase-3), phosphatidylinositol 3-kinase (PI3K), protein kinase B (AKT), and NF-κB p65 were detected by Western blotting, and the changes of BAX, Bcl2, c-caspase-3, and NF-κB p65 proteins were further verified after the cells were treated with NF-κB pathway inhibitor BAY11-7082. Results BET inhibited the proliferation of C4-2B cells in a dose-dependent manner. The 50% inhibitory concentration (IC50) was 422.7 mmol/L after the cells were treated with BET for 48 hours. Compared with the control group (0 mmol/L BET treatment), the proliferation of C4-2B cells was inhibited along with morphological changes, decreased clone formation ability and increased apoptosis rate in 200, 300, 400 mmol/L BET treated groups.
