Sensitive and accurate determination of aflatoxin B1 (AFB1) is of great significance to food safety and human health as it is recognized as the most toxic mycotoxin and carcinogenic. Herein, we report a ratiometric luminescence aptasensor based on dual-emissive persistent luminescent nanoparticles (PLNP) for the accurate determination of trace AFB1 in complex food samples without autofluorescence and exogenous interference. Dual-emissive PLNP ZnGa2O4Cr0.0001 was prepared first and acted as the donor for energy transfer as well as the signal unit with phosphorescence at 714 and 508 nm (the detection and the reference signal, respectively). AFB1 aptamer was then bonded on the surface of PLNP to offer specific recognition ability. Aptamer complementary DNA modified with Cy5.5 was employed as the acceptor for energy transfer and the quenching group to eventually develop a turn-on ratiometric luminescence aptasensor. The developed ratiometric luminescence aptasensor combined the merits of long-lasting luminescence, in situ excitation and autofluorescence-free of PLNP, exogenous interference-free and self-calibration reading of ratiometric sensor, as well as the high selectivity of aptamer, holding great promise for accurate determination of trace AFB1 in complex matrix. The developed ratiometric aptasensor exhibited excellent linearity (0.05-70 ng mL-1), low limit of detection (0.016 ng mL-1), and good precision (2.3% relative standard deviation for 11 replicate determination of 1 ng mL-1 AFB1). The proposed ratiometric aptasensor was successfully applied for the determination of AFB1 in corn, wheat, peanut, millet, oats, and wheat kernels with recoveries of 95.1-106.5%.Metal ions stabilize protein-protein interactions and can modulate protein aggregation. Here, using liquid-based atomic force microscopy and molecular dynamics simulations, we study the concentration-dependent effect of Cu2+ ions on the aggregation pathway of α-synuclein (α-Syn) proteins, which play a key role in the pathology of Parkinson's disease. The full spectrum of α-Syn aggregates in the presence and absence of Cu2+ ions from monomers to mature fibrils was resolved and quantified at the gold-water interface. Raman spectroscopy confirmed the atomic force microscopy (AFM) findings on the heterogeneity in aggregated states of α-Syn. The formation of annular oligomers was exclusively detected upon incubating α-Syn with Cu2+ ions. Our findings emphasize the importance of targeting annular α-Syn protein oligomers for therapeutic intervention and their potential role as biomarkers for early detection and monitoring progression of neurodegeneration. Two series of Prussian blue analogs (PBA) were used as model compounds in order to disentangle the information contained in X-ray magnetic circular dichroism (XMCD) at the K-edges of transition metals. The number of 3d electrons on one site of the bimetallic cyanide polymer has been varied by associating to the [Fe(CN)6]3- or the [Cr(CN)6]3- precursors various divalent metal ions A2+ (Mn2+, Fe2+, Co2+, Ni2+, and Cu2+). The compounds were studied by X-ray diffraction and SQUID magnetometry, as well as by X-ray absorption spectroscopy and XMCD at the K-edges of the A2+ transition metal ion. The study shows that the 1s → 4p contribution to the A K-edge XMCD signal can be related to the electronic structure and the magnetic behavior of the probed A2+ ion the shape of the signal to the filling of the 3d orbitals, the sign of the signal to the direction of the magnetic moment with respect to the applied magnetic field, the intensity of the signal to the total spin number SA, and the area under curve to the Curie constant CA. The whole study hence demonstrates that PBAs are particularly well-adapted for understanding the information contained in the transition metals K-edge XMCD signals. It also offers new perspectives toward the full disentangling of the information contained in these signals and access to new insights into materials magnetic properties. The exact mechanism behind general anesthesia remains an open question in neuroscience. It has been proposed that anesthetics selectively prevent consciousness and memory via acting on microtubules (MTs). It is known that the magnetic field modulates MT organization. A recent study shows that a radical pair model can explain the isotope effect in xenon-induced anesthesia and predicts magnetic field effects on anesthetic potency. Further, reactive oxygen species are also implicated in MT stability and anesthesia. Based on a simple radical pair mechanism model and a simple mathematical model of MT organization, we show that magnetic fields can modulate spin dynamics of naturally occurring radical pairs in MT. We propose that the spin dynamics influence a rate in the reaction cycle, which translates into a change in the MT density. We can reproduce magnetic field effects on the MT concentration that have been observed. Our model also predicts additional effects at slightly higher fields. Our model further predicts that the effect of zinc on the MT density exhibits isotopic dependence. The findings of this work make a connection between microtubule-based and radical pair-based quantum theories of consciousness.A complete understanding of muscle mechanics allows for the creation of models that closely mimic human muscle function so they can be used to study human locomotion and evaluate surgical intervention. This includes knowledge of muscle-tendon parameters required for accurate prediction of muscle forces. However, few studies report experimental data obtained directly from whole human muscle due to the invasive nature of these experiments. This article presents an intraoperative, in vivo measurement protocol for whole muscle-tendon parameters that include muscle-tendon unit length, sarcomere length, passive tension, and active tension in response to external stimulation. HC-030031 mouse The advantage of this protocol is the ability to obtain these rare experimental data in situ in addition to muscle volume and weight since the gracilis is also completely removed from the leg. The entire protocol including the surgical steps for gracilis harvest takes ~ 3 h. Actual testing of the gracilis where experimental data is measured takes place within a 30-min window during surgery.Liver damage affects the prognosis of patients with erythropoietic protoporphyria (EPP). However, there is no radical cure for EPP patients with severe liver damage. This study aims to investigate the effectiveness of phlebotomy in patients with severe liver damage. We examined seven patients diagnosed with EPP and liver damage between 2010 and 2020. Of the 7 cases, phlebotomy was performed in 3 cases with severe hepatic disorder, and the improvement effect of hepatic disorder was observed in all cases. In addition, as an additional study, we also investigated the mechanism by which liver damage becomes more severe. Liver biopsy samples were stained with hematoxylin and eosin and immunohistochemistry was used to examine the expression of adenosine triphosphate-binding transporter G2 (ABCG2). Liver biopsies were performed in 3 of 7 patients with EPP. Of these three patients, ABCG2 expression was low in two patients, especially in the protoporphyrin (PP) deposition area. Two patients with reduced ABCG2 expression subsequently developed severe liver damage. However, the causal relationship between the decreased expression of ABCG2 and the exacerbation of liver damage has not been directly proved, and further investigation is required in the future. This study demonstrated the effectiveness of phlebotomy in EPP patients with severe liver damage. The East Greenland-Svalbard-Barents Sea (EGSB) bowhead whale stock (Balaena mysticetus) was hunted to near extinction and remains Endangered on the International Union of Conservation of Nature Red List. The intense, temporally extensive hunting pressure may have left the population vulnerable to other perturbations, such as environmental change. However, the lack of genomic baseline data renders it difficult to evaluate the impacts of various potential stressors on this stock. Twelve EGSB bowhead whales sampled in 2017/2018 were re-sequenced and mapped to a previously published draft genome. All individuals were unrelated and void of significant signs of inbreeding, with similar observed and expected homo- and heterozygosity levels. Despite the small population size, mean autosome-wide heterozygosity was 0.00102, which is higher than that of most mammals for which comparable estimates are calculated using the same parameters, and three times higher than a conspecific individual from the Eastern-Canada-West-Greenland bowhead whale stock. Demographic history analyses indicated a continual decrease of Ne from ca. 1.5 million to ca. 250,000 years ago, followed by a slight increase until ca. 100,000 years ago, followed by a rapid decrease in Ne between 50,000 and 10,000 years ago. These estimates are lower than previously suggested based on mitochondrial DNA, but suggested demographic patterns over time are similar. Many species of devastating insect pests have acquired a high degree of resistance to insecticides in the field during the last few decades. Spodoptera exigua, for example, is the most damaging pests of economic crops with a worldwide spread. In a present study, the comparative growth, reproduction, and detoxification enzyme activity were evaluated along with exposure to three insecticides at low lethal doses of lufenuron, indoxacarb, and spinosad as compared to the control. Results indicate that the larval developmental time was significantly extended on lufenuron (21.5 ± 29 days) followed by indoxacarb (20.28 ± 0.24 days) and spinosad (19.74 ± 0.23 days) as compared to that on the control (18.13 ± 0.13 days). Similarly, the lowest number of eggs of S. exigua females were recorded on lufenuron (328.75 ± 50.81 eggs) followed by spinosad (367 ± 36.4 eggs) and indoxacarb (411.58 ± 42.38 eggs) as compared to that on the control (560.2 ± 13.47). Interestingly, the lowest intrinsic rate of increase (r) (0.121 ± 0.009) and highest mean generation time (T) (36.2 ± 0.35 days) were observed when larvae were treated to a low lethal concentration (LC20) of lufenuron as compared to that of indoxacarb, spinosad, and control. In addition, considerably lower activity of all detoxification enzymes in larvae was recorded on lufenuron after control as compared to that on indoxacarb and spinosad. Our study serves as a reference and basis for the toxicity and low lethal evaluation of lufenuron, indoxacarb, and spinosad on life table parameters and enzymatic properties in S. exigua, which may contribute to identifying targets for effective control of S. exigua.Concentration of dissolved aromatic and halogenated non-methane volatile organic compounds (NMVOCs) was estimated in sewage flowing through the open drainage canal network of Kolkata megacity in India in dry (summer) and wet (post-monsoon) seasons at five locations. Seventeen aromatic and halogenated NMVOC species were studied by headspace solid-phase micro-extraction (HS-SPME) technique followed by gas chromatography-mass spectrometric (GC-MS) analysis. Distinct seasonal variations in the concentration of individual NMVOC species were observed, but spatial variation was negligible. Total dissolved NMVOC (TNMVOC) concentration was higher (16.64 µg l - 1) in summer over post-monsoon (12.70 µg l - 1). Chloroform and toluene were the most abundant species in both seasons. Principal component analysis indicated contribution from industrial sources (38.8% and 35.5%), solvent usage (35.9% and 35.5%), in situ formation through microbial pathways (22.2% and 11.5%) in dry and wet seasons, respectively.
