Deletion of phagocytic IRF5 resulted in significantly decreased IRF5 expression, attenuated pro-inflammatory and enhanced anti-inflammatory responses to HIE, and improved outcomes compared with IRF5fl/fl control pups. In vitro lentivirus transfection experiments revealed that overexpression of IRF5 in microglia amplified pro-inflammatory signals and exacerbated OGD-induced neuronal apoptosis and neurite fragmentation. IRF5 signaling mediates microglial pro-inflammatory activation and also affects anti-inflammatory responses. Phagocytic IRF5 signaling is detrimental in HIE and is a potential therapeutic target for post-ischemic inflammation.The present study explored the interrelations between a broad set of appraisal ratings and five physiological signals, including facial EMG, electrodermal activity, and heart rate variability, that were assessed in 157 participants watching 10 emotionally charged videos. A total of 134 features were extracted from the physiological data, and a benchmark comparing different kinds of machine learning algorithms was conducted to test how well the appraisal dimensions can be predicted from these features. For 13 out of 21 appraisals, a robust positive R2 was attained, indicating that the dimensions are actually related to the considered physiological channels. The highest R2 (.407) was reached for the appraisal dimension intrinsic pleasantness. Moreover, the comparison of linear and nonlinear algorithms and the inspection of the links between the appraisals and single physiological features using accumulated local effects plots indicates that the relationship between physiology and appraisals is nonlinear. By constructing different importance measures for the assessed physiological channels, we showed that for the 13 predictable appraisals, the five channels explained different amounts of variance and that only a few blocks incrementally explained variance beyond the other physiological channels.Implicit social-affective biases-reflected in a propensity to approach positive and avoid negative stimuli-have been documented in humans with paradigms, such as the Approach-Avoidance Task (AAT). However, the degree to which preemptively engaging cognitive control can help to down-regulate those behavioral tendencies remains poorly understood. While undergoing functional magnetic resonance imaging (fMRI), 24 healthy participants completed a cued version of the AAT, in which they responded to pictures of happy or angry faces by pulling a joystick toward themselves (approach) or pushing the joystick away (avoidance) based on the color of the stimulus frame. On some trials, they were cued to reverse the frame color/joystick action instructions. Before stimulus onset, a reverse cue was associated with deactivation of a visuo-spatial and motor planning network and subsequent slowing down in response to stimuli. During the stimulus phase, a reverse cue was associated with a) activation of cognitive control areas, including the right inferior frontal gyrus (IFG) and right inferior parietal lobule (IPL); and b) reduced right precentral gyrus activation when having to push (avoid) a happy face. Overall, these results suggest that proactively engaging cognitive control can help fine-tune behavioral and neural adjustment to emotionally incongruent behavioral conditions.In four experiments pupillary responses were used to examine attention allocation and encoding dynamics in free recall. In Experiment 1, pupillary responses increased (and then decreased) across serial position suggesting that attention was increasingly allocated to items during learning until working memory was overloaded. In Experiment 2, manipulating presentation duration resulted in larger and more sustained pupillary responses with increased presentation duration, suggesting that participants were likely engaging in more elaborative and attention-demanding processes. In Experiment 3a, manipulating list-length resulted in decreased pupillary responses across serial position suggesting that participants were prioritizing early list items and less attention was allocated to later items. In Experiment 3b, when list-length was known, pupillary responses in the long-list length condition tended to decrease across serial position whereas pupillary responses in the short list-length condition tended to increase and decrease across serial positon. These results suggest that participants flexibly allocate attention to items during encoding depending on the nature of the task and the types of processes that are engaged in. These results further suggest the potential of utilizing pupillary responses to track attention allocation during learning.Collective memory is shared by a group and is part of that group's identity. Memory for political leaders is a prototypical case of collective memory. The present study investigated collective memory for Swiss federal councilors in order to test the trajectory of collective memory across four different generations (i.e., Millennials, Generation X, Baby-Boomers, and Silents) in a collaborative government system. In contrast to a presidential system, Switzerland is governed by seven equal councilors who share power and responsibilities. Thus, the individual member of the government is less important, and the number of councilors is larger compared to a presidential system, which may influence collective memory. The results revealed a recency effect as well as a generation-specific reminiscence effect, but no primacy effect as reported for presidential systems. These results indicate that the contribution of semantic memory and autobiographic memory to the trajectory of collective memory vary across government systems. Specifically, for a collaborative government system, autobiographic memory has a stronger contribution to the trajectory of collective memory.Stem cells are characterized by self-renewal and by their ability to differentiate into cells of various organs. This review focuses on stem cell-based in vitro modeling of neurodevelopmental disorders, more specifically, the fundamentals and technical advancements in monolayer neuron and brain organoid cultures. Furthermore, we discuss the drawbacks of the conventional culture method and explore the advanced, cutting edge 3D organoid models for several neurodevelopmental diseases, including genetic diseases such as Down syndrome, Rett syndrome, and Miller-Dieker syndrome, as well as brain malformations like macrocephaly and microcephaly.
