While drug-loaded microparticles (MPs) can serve as drug reservoirs for sustained drug release and therapeutic effects, needle clogging by MPs poses a challenge for ocular drug delivery via injection. Two polymers commonly used in ophthalmic procedures-hyaluronic acid (HA) and methylcellulose (MC)-have been tested for their applicability for ocular injections. HA and MC were physically blended with sunitinib malate (SUN)-loaded PLGA MPs for subconjunctival (SCT) injection into rat eyes. The HA and MC viscous solutions facilitated injection through fine-gauged needles due to their shear-thinning properties as shown by rheological characterizations. The diffusion barrier presented by HA and MC reduced burst drug release and extended overall release from MPs. The significant level of MP retention in the conjunctiva tissue post-operation confirmed the minimal leakage of MPs following injection. The safety of HA and MC for ocular applications was demonstrated histologically.Surface water eutrophication due to excessive nutrients has become a major environmental problem around the world in the past few decades. Among these nutrients, nitrogen and phosphorus are two of the most important harmful cyanobacterial bloom (HCB) drivers. A reliable prediction of these parameters, therefore, is necessary for the management of rivers, lakes, and reservoirs. The aim of this study is to test the suitability of the powerful machine learning (ML) algorithm, random forest (RF), to provide information on water quality parameters for the Tri An Reservoir (TAR). Three species of nitrogen and phosphorus, including nitrite (N-NO2-), nitrate (N-NO3-), and phosphate (P-PO43-), were empirically estimated using the field observation dataset (2009-2014) of six surrogates of total suspended solids (TSS), total dissolved solids (TDS), turbidity, electrical conductivity (EC), chemical oxygen demand (COD), and biochemical oxygen demand (BOD5). Field data measurement showed that water quality in the TAR was eutrophic with an up-trend of N-NO3- and P-PO43- during the study period. The RF regression model was reliable for N-NO2-, N-NO3-, and P-PO43- prediction with a high R2 of 0.812-0.844 for the training phase (2009-2012) and 0.888-0.903 for the validation phase (2013-2014). The results of land use and land cover change (LUCC) revealed that deforestation and shifting agriculture in the upper region of the basin were the major factors increasing nutrient loading in the TAR. Among the meteorological parameters, rainfall pattern was found to be one of the most influential factors in eutrophication, followed by average sunshine hour. Our results are expected to provide an advanced assessment tool for predicting nutrient loading and for giving an early warning of HCB in the TAR.Alternative promoter and alternative splicing are two important mechanisms of gene regulation and protein diversity in different physiological contexts of eukaryotes, especially in stem cells and developmental stages. Pou5f1 gene which codes the stemness marker OCT4, utilizes alternative splicing and promoter mechanisms, which result in generation of multiple spliced variants and subsequently multiple protein isoforms. By far, nine variants of OCT4 (OCT4A, OCT4B, OCT4B1, OCT4B2, OCT4B3, OCT4B4, OCT4C, OCT4C1, and OCT4D) have been introduced. It has been well established that OCT4A plays essential roles in early developmental stages as well as maintenance of stemness in embryonic stem cells (ESCs). However, the roles and functions of other variants and isoforms of OCT4 in biological systems are less appreciated. In this study, we report a new OCT4 variant, designated as OCT4B5. RT-PCR assay on different human cell lines including pluripotent, normal and cancer cells showed that OCT4B5 is expressed at variable level in different cell lines. By semi-quantifying of OCT4B5 expression in pluripotent and differentiated states of NT2 cell lines, we reveal that this variant of OCT4 is highly expressed in undifferentiated state and its expression is down-regulated upon differentiation. Compared to OCT4A which is sharply down-regulated in retinoic acid induced differentiation of NT2 cell line, the expression of OCT4B5 remains at low level in differentiated state. Overall, this study emphasizes the complexity of OCT4 gene expression and regulation in different states of stem cells and physiological contexts. Graphical Abstract.Heavy metal (HM) contamination in agricultural soils has been a significant health concern worldwide due to their persistent and non-biodegradable nature and biomagnification to higher trophic levels. The present study was conducted to assess Cd and Pb concentrations in soil samples collected from potato-growing areas in Thall, Dir-Kohistan, and to determine their associated health and ecological risks. The contamination factor calculated for Cd (0 to 1.74) and Pb (0 to 0.91) showed their moderate to low contamination in the study area. selleck products Geo-accumulation indices of Cd and Pb were less than 1, indicating moderate soil pollution of these HM in the study area. However, the ecological risk factor (Eri) of Cd was greater than 40 for only three soil samples, indicating the moderate potential of ecological risks of respective soils. The principal component analysis (PCA) and Pearson correlation suggested that the contamination in different soils was lithogenic followed by anthropogenic activities. The hazard quotient (HQ) in children and adults was found in the following order dermal > ingestion > inhalation. Moreover, the values of HQ through various exposure routes were higher in children compared to adults, which showed that adults were at a lower level of risk associated with HM contamination. The results of the present study can serve as baseline data for government agencies related to environmental protection, which could devise policies to minimize Cd and Pb contamination in the agricultural soils.
A previously acquired fear response often spreads to perceptually or conceptually close stimuli or contexts. This process, known as fear generalization, facilitates the avoidance of danger, and dysregulations in this process play an important role in anxiety disorders. Oxytocin (OT) has been shown to modulate fear learning, yet effects on fear generalization remain unknown.

We employed a randomized, placebo-controlled, double-blind, between-subject design during which healthy male participants received either intranasal OT or placebo (PLC) following fear acquisition and before fear generalization with concomitant acquisition of skin conductance responses (SCRs). Twenty-four to 72h before the fear learning and immediately after the fear generalization task, participants additionally complete a discrimination threshold task.

Relative to PLC, OT significantly reduced perceived risk and SCRs towards the CS+ and GS1 (the generalization stimulus that is most similar to CS+) during fear generalization, whereas the discrimination threshold was not affected.
