Several seed-borne viroids are currently listed as quarantine pathogens in many countries. Among them, columnea latent viroid (CLVd), pepper chat fruit viroid (PCFVd), potato spindle tuber viroid (PSTVd), tomato apical stunt viroid (TASVd), tomato chlorotic dwarf viroid (TCDVd), and tomato planta macho viroid (TPMVd) are of major concerns. The objective of this study was to design and test universal primers that could be used to detect six viroids in solanaceous plants using one-step RT-PCR and reverse transcription loop-mediated isothermal amplification (RT-LAMP). Results revealed that a pair of degenerate primers could be used in a one-step RT-PCR to amplify six pospiviroids from Solanaceae seeds and plants. Moreover, five primers were designed and used in RT-LAMP to amplify six pospiviroids. (Orchidaceae), a perennial plant, is a traditional Chinese herb (known as baiji) used to treat hemorrhage, scalding injuries, gastric ulcers, pulmonary diseases, and inflammation (Zu et al. 2019). In May 2019, foliar blight symptoms were observed on approximately 25% of B. striata (cv. Guiji No.1) plants in three plantations (∼4.5 hectares in total) in Ziyuan County, Guangxi Province, China. Initial symptoms were light brown, irregular, water-soaked spots on the plant leaves. Several spots often merged, forming large, irregular, lesions that extended onto the stem after a week and led to leaf abscission, and even plant death. To determine the causal agent, 5-mm squares cut from the margin of 6 infected leaves were surface disinfected in 1% sodium hypochlorite solution for 2 min, rinsed three times with sterile distilled water, plated on potato dextrose agar (PDA), and incubated at 28°C (12-h light-dark cycle) for 3 days. The emerging hyphal tip of a single mycelium was transe enclosed in transparent plastic bags and incubated in a greenhouse at 28°C for 14 days (12-h photoperiod). Three days post-inoculation, leaves exposed to the mycelial fragments had symptoms similar to those originally observed in the field. No symptoms were detected on control plants. Experiments were replicated three times with similar results. To fulfill Koch's postulates, R. solani AG-2-2 IIIB was re-isolated on PDA from symptomatic leaves and confirmed by sequencing, whereas no fungus was isolated from the control plants. To our knowledge, this is the first report of R. solani AG-2-2 IIIB causing foliar blight on B. striata in China, and these findings will be useful for further control strategies and research.Tall verbena (Verbena bonariensis) is widely used in landscape layout in sparse forests, botanical gardens and villa areas, and it is also a kind of Chinese herbal medicine. The flower and bud use as Chinese herbal medicine causing heat-clearing and detoxification. In August 2019, observed on tall verbena in Shangqiu, Henan Province, China. Symptoms white fungal growth observed on both sides of fresh and mature leaves of tall verbena grown on campus at Shangqiu Normal University. More than 60% plants were infected by this fungus and infected plants were observed to shed leaves early. One representative voucher specimen was deposited at the herbarium of Shangqiu Normal University (SQNU), Shangqiu, China, under the accession number of Vb01. Conidiophores of the fungus contained 5-6cells and measured 102.0-142.9μm (mean 126.2μm, n = 30) long, with foot-cells of conidiophores measured 36.5-46.9μm (mean 42.9μm, n = 30). Conidia produced in chains were variable in shape, ellipsoid to ovoid, and measured 24.8-30.7×1bena. Five non-inoculated seedlings were used as controls. The plants were maintained in a greenhouse at 26 to 28 ℃, and relative humidity of 60 to 65%. Inoculated leaves developed symptoms after nine days, whereas the control plants remained symptomless. The fungus presented on the inoculated plant was morphologically identical to that originally observed on diseased plants, fulfilling Koch's postulates. P. xanthii s. lat. has been reported on several hosts in China (Braun & Cook, 2012). This is the first report of P. xanthii s. lat. on Verbena bonariensis outside of Japan (Meboon, 2015).Alfalfa root rot caused by Fusarium tricinctum is one of the most important soil-borne diseases resulting in significant losses to alfalfa agriculture worldwide. Fungicides used in management of disease affect the environment and human health. In this study, a strain of Ochrobactrum intermedium (I-5), isolated from alfalfa rhizosphere soil, exhibited strong antifungal activity against a number of causative pathogens of alfalfa root rot, and showed the strongest antagonistic activity against F. tricinctum (the longest radius/shortest radius ratio of 3.09). When applied at 10%, a filtrate of I-5 liquid culture significantly reduced the spore production and germination and mycelial growth of F. tricinctum, and the inhibition rate was 76.67%, 78.93% and 55.77%, respectively. Furthermore, a filtrate and suspension of the strain, when applied at 10%, reduced alfalfa root rot by more than 73%. The strain clearly promoted the activities of invertase, urease, cellulose, and neutral phosphatase in alfalfa rhizosphere soil and significantly reduced the damage to rhizosphere soil quality attributable to alfalfa root rot.
