We assessed the sex-specific differences in the molecular mechanisms of insulin resistance in muscle and adipose tissue, in a MS rat model induced by a high sucrose diet.
Male, female, and ovariectomized female Wistar rats were randomly distributed in control and high-sucrose diet (HSD) groups, supplemented for 24 weeks with 20% sucrose in the drinking water. At the end, we assessed parameters related to MS, analyzing the effects of the HSD on critical nodes of the insulin signaling pathway in muscle and adipose tissue.
At the end of the treatment, HSD groups of both sexes developed obesity, with a 15, 33 and 23% of body weight gain in male, female, and OVX groups respectively, compared with controls; mainly related to hypertrophy of peripancreatic and gonadal adipose tissue. They also developed hypertriglyceridemia, and liver steatosis, with the last being worse in the HSD females. Compared to the control groups, HSD rats had higher IL1B and TNFA levels and insulin resistance. HSD females were more intolerant to glucose than HSD males. Our observations suggest that insulin resistance mechanisms include an increase in phosphorylated AKT(S473) form in HSD male and female groups and a decrease in phosphorylated P70S6K1(T389) in the HSD male groups from peripancreatic adipose tissue. While in gonadal adipose tissue the phosphorylated form of AKT decreased in HSD females, but not in HSD males. Finally, HSD groups showed a reduction in p-AKT levels in gastrocnemius muscle.

A high-sucrose diet induces MS and insulin resistance with sex-associated differences and in a tissue-specific manner.Traffic accidents are a global concern due to the elevated mortality rates of both drivers and pedestrians. The World Health Organization declared 2011-2020 as the Decade of Action for Road Safety, endorsing initiatives to reduce traffic-related deaths. Yet, despite these incentives, fatal accidents still occur. Different studies have linked deficits in executive functions to risky driving attitudes and crashes. The present study focuses on demographic, cognitive and personality factors, related to the prefrontal cortex, that are characteristic of drivers prone to risky behavior behind the wheel. The penalty Points System was used to classify drivers as "safe", with no point loss over a two-year period, or "risky", with full point loss during the same interval. A neuropsychological assessment of prefrontal cognitive functions was carried out on each group to identify variables associated with safe and risky behavior. Neuropsychological indexes were obtained from a continuous performance task without cue (Simple Attention), a continuous performance task with cue (Conditioned Attention), the Tower of Hanoi test and the Neurologically-related Changes in Personality Inventory (NECHAPI). A Discriminant Analysis (DA) found that education level, reaction times in Simple and Conditioned Attention, learning errors in the Tower of Hanoi and vulnerability in the personality test, best predicted whether drivers were likely to be in the safe or risky group. Finally, a cross-validation analysis performed on the same sample correctly classified 87.5% of the drivers. These data suggest that prefrontal dysfunction contributes to risky behavior behind the wheel. The inclusion of cognitive programs to identify and train drivers with this propensity could reduce risky driving, and consequently, save lives on the road.There are thousands of disused and abandoned mining sites around the world with substantial accumulations of exposed mine spoil materials that pose a direct threat to their surrounding environment. Management of such sites, and neutralisation of the environmental threats they pose, is therefore extremely important and is an issue of global significance. Low cost management and remediation strategies need to be developed because many abandoned mine sites are in remote and/or economically challenged areas. One promising option is the incorporation of biochar into spoil materials, which has the potential to immobilise leachable toxic constituents and facilitate revegetation and thereby stabilisation of spoil heaps. This study investigated the capacity of readily available biochar materials made from wheat and rice waste products to immobilise and retain key metallic contaminants Pb and Zn from solution, and also investigated the utility of biochar application for remediating mine spoil heaps from different mine types in terms of facilitating establishment of vegetation coverage and minimising porewater element mobility within spoil heaps. The results demonstrated the high sorption capacity of the biochars (typically >97% of Pb or Zn in solution) and their ability to retain the metals despite an active desorption procedure (>93% of sorbed Pb retained and >75% of sorbed Zn). The remediation trial revealed that biochar application increased plant yield and decreased plant assimilation of many potentially toxic elements and also decreased spoil porewater concentrations of Al, Cd, Pb and Zn in most cases. In some spoil types investigated biochar addition also significantly decreased porewater concentrations of As (e.g. from ~30 mg/L to ~5 mg/L), demonstrating its potential utility for low cost environmental remediation across a range of mine spoil types.Proper identification of critical source areas (CSAs) is important for economic viability of any best management practices (BMPs) aimed at reducing sediment and phosphorus loads to receiving water bodies. Both continuous and event-based hydrologic and water quality models are widely used to identify and assess CSAs, however, their comparative assessment is lacking. In this study, we have used continuous Soil and Water Assessment Tool (SWAT) and event-based Agriculture Non-Point Source (AGNPS) pollution models to identify CSAs for sediment and phosphorus in a watershed in Ontario, Canada. Along with their original version, both models were re-conceptualized to incorporate saturation excess mechanism of runoff generation, which is also refereed as variable source area (VSA) integration. The models were set-up using high resolution spatial, crop- and land-management, and meteorological dataset; and calibrated with reasonable accuracy against streamflow, sediment and phosphorus concentration data at multiple locaNPS models. However, a more comprehensive analysis based on more detailed field observations is needed to further confirm the results.A challenge for current surface water restoration and management in China is acquiring the source information for complex pollution scenarios in order to develop effective control strategies. As an important part of dissolved organic matter, chromophoric dissolved organic matter (CDOM) contains unique chemical signals related to various pollution sources. Spectral methods such as fluorescence excitation-emission matrices coupled with parallel factor analysis enable rapid and low-cost CDOM characterization for source tracking. In this study, a typical small-sized river flowing through mixed land-use regions in southeastern China, the Lujiang River, was investigated to determine the responses of CDOM to spatiotemporal factors. The effects of land-use patterns were reflected by the fluorescent components of terrestrial and sewage substances. A high and stable proportion of terrestrial-like components (C1 + C2) in each sampling period (i.e., March 47.6 ± 5.7% and October 44.3 ± 2.7%) indicated a high input of non-point source (NPS) pollution from both agriculture and urban areas. In addition, the difference in solar radiation intensity induced by climate and air quality changes was also reflected by variability in the photodegradation product component (C3) of terrestrial precursors between October (24.8 ± 2.6%) and March (4.5 ± 2.0%), suggesting that terrestrial components could be a sensitive indicator for NPS pollutant monitoring. Increased sewage impact in downstream regions was reflected by a spike in the tryptophan-like component (C4); temporal variations in C4 (paired t-Test, p less then 0.005) also indicated that sewage substances were more prone to removal by microbial activity in warmer seasons. Cytidine The dynamics of C4 could serve as a good indicator of sewage disposal performance. The results of this study demonstrate that CDOM data have important practical applications for existing water restoration campaigns in southeastern China, as well as substantial potential for routine water quality monitoring.In light of current trends of land abandonment across Europe, the rewilding of abandoned land is increasingly discussed as a future land management option. Since many of the regions affected by rewilding developments will be located in vicinity to densely populated areas, it is imperative to better understand public's attitudes towards an expansion of wilderness. Prior research on wilderness attitudes has typically been descriptive and rarely explored the underlying processes that lead to differences in wilderness attitudes. Taking the Theory of Planned Behaviour as our framework of analysis, we examine public attitudes towards wilderness in the context of people's beliefs of wilderness, and to what extent these beliefs can explain differences in wilderness attitudes between social groups. Results of a questionnaire survey (n = 858) in the mountain region of South Tyrol in the Central Alps show that wilderness attitudes are significantly influenced by three dimensions of wilderness beliefs, corresponding to 1) respondents' general wilderness representations, 2) perceived extent of wilderness areas, and 3) expected consequences of wilderness expansion. In addition, significant differences in wilderness attitudes are found between age groups, user groups, and groups with varying degrees of interest in wildlife watching. Our results indicate that these differences are partly related to contradictory views of wilderness, suggesting that it is not only conflicting interests that may lead to differences in wilderness attitudes between social groups. We conclude that participatory processes are needed to consider the plurality of people's wilderness representations and their perception of risks and benefits for the design and implementation of socially-just rewilding initiatives.The harm from mercury pollution to human health and the environment has long been known. In recent years, the combination of industrial activities and long-term atmospheric transport has resulted in a sustained increase in mercury concentrations in soils. However, soil remediation and mercury-contaminated soil management in China are still in its infancy, and there is ample space for the development of related research. We systematically reviewed several pertinent topics and found that soil mercury pollution around mines and industrial soil in China is the most serious. The highest mercury content is found in the soil around the Tongren mercury mine in Guizhou Province and the thermometer factories. The average content of soil mercury is similar to that of atmospheric mercury emission in China. Mercury content in soil gradually decreases from the southeast to the northwest. In order to repair the mercury-contaminated soil, solidification and stabilization technology have been developed in China and applied in the engineering of restoration.
