Here, we present an approach to characterize MAG-ganglioside interactions in real time, where MAG, GD1a, and GT1b contents are controlled and they are in their in vivo orientation within fluid lipid bilayers. Using a quartz crystal microbalance with dissipation monitoring (QCM-D) biosensor functionalized with a supported lipid bilayer (SLB) and MAG, we detect vesicular GD1a and GT1b binding and determine the interaction kinetics as a function of vesicular ganglioside content. MAG-bound vesicles are deformed similarly, regardless of the ganglioside or its mole fraction. We further demonstrate how MAG-ganglioside interactions can be disrupted by antiganglioside antibodies that override MAG-based neuron growth inhibition.The presence of methane and other hydrocarbons in domestic-use groundwater aquifers poses significant environmental and human health concerns. Isotopic measurements are often relied upon as indicators of groundwater aquifer contamination with methane. This study examined the relationships between microbiology and chemistry in groundwater wells located in the Denver-Julesburg Basin of Colorado, a rapidly urbanizing area with active oil and gas development. A primary goal was to determine if microbial data can reliably indicate the quantities and sources of groundwater methane. Comprehensive chemical and molecular analyses were performed on 39 groundwater well samples from five aquifers. Elevated methane concentrations were found in only one aquifer, and both isotopic and microbial data support a microbial origin. Microbial parameters had similar explanatory power as chemical parameters for predicting sample methane concentrations. Furthermore, a subset of samples with unique microbiology corresponded with unique chemical signatures that may be useful indicators of methane gas migration, potentially from nearby coal seams interacting with the aquifer. Microbial data may allow for more accurate determination of groundwater contamination and improved long-term water quality monitoring compared solely to isotopic and chemical data in areas with microbial methane.Emerging applications in spectroscopy-related bioanalytics demand for integrated devices with small geometric footprints and fast response times. While hollow core waveguides principally provide such conditions, currently used approaches include limitations such as long diffusion times, limited light-matter interaction, substantial implementation efforts, and difficult waveguide interfacing. Here, we introduce the concept of the optofluidic light cage that allows for fast and reliable integrated spectroscopy using a novel on-chip hollow core waveguide platform. The structure, implemented by 3D nanoprinting, consists of millimeter-long high-aspect-ratio strands surrounding a hollow core and includes the unique feature of open space between the strands, allowing analytes to sidewise enter the core region. Reliable, robust, and long-term stable light transmission via antiresonance guidance was observed while the light cages were immersed in an aqueous environment. The performance of the light cage related to absorption spectroscopy, refractive index sensitivity, and dye diffusion was experimentally determined, matching simulations and thus demonstrating the relevance of this approach with respect to chemistry and bioanalytics. The presented work features the optofluidic light cage as a novel on-chip sensing platform with unique properties, opening new avenues for highly integrated sensing devices with real-time responses. Application of this concept is not only limited to absorption spectroscopy but also includes Raman, photoluminescence, or fluorescence spectroscopy. Furthermore, more sophisticated applications are also conceivable in, e.g., nanoparticle tracking analysis or ultrafast nonlinear frequency conversion.The development of antimicrobial compounds is now regarded as an urgent problem. Antimicrobial peptides (AMPs) have great potential to become novel antimicrobial drugs. Feleucin-K3 is an α-helical cationic AMP isolated from the skin secretion of the Asian bombinid toad species Bombina orientalis and has antimicrobial activity. In our previous studies, amino acid scanning of Feleucin-K3 was performed to determine the key site affecting its activity. In this study, we investigated and synthesized a series of analogues that have either a natural or an unnatural hydrophobic amino acid substitution at the fourth amino acid residue of Feleucin-K3. Among these analogues, Feleucin-K59 (K59), which has an α-(4-pentenyl)-Ala substitution, was shown to have increased antimicrobial activity against both standard and drug-resistant strains of clinical common bacteria, improved stability, no hemolytic activity at antimicrobial concentrations, and no resistance. In addition, K59 has potent antibiofilm activity in vitro. More importantly, K59 showed better antimicrobial and antibiofilm activities against drug-resistant bacteria in in vivo experiments in mice than traditional antibiotics. In this preliminary study of the mechanism of action, we found that K59 could rapidly kill bacteria by a dual-action mechanism of disrupting the cell membrane and binding to intracellular DNA, thus making it difficult for bacteria to develop resistance.
