The injection volume is limited by an increase in intraocular pressure (IOP) and 50-100 µL are typically used for most intravitreally (IVT) applied commercial products. Direct measurement of IOP is difficult and has not been studied dependent on solution properties and injection rates. We used an instrumental set-up to study IOP ex vivo using healthy enucleated porcine eyes. IOP was determined as a function of injection volume for viscosities between 1 and 100 mPas, injection rates of 0.1, 1, and 1.5 mL/min, and needle length and diameter (27/30G and 0.5/0.75″) using Dextran solutions. IOP increased exponentially for injection volumes larger than 100 µL. We did not observe differences in IOP dependent on viscosity, injection rate, and needle diameter. However, variability increased significantly for injection volumes larger than 100 µL and, unexpectedly, declined with higher viscosities. We demonstrate that the exponential increase in IOP is not reflected by injection force measurements for typical configurations that are used for IVT application. The present findings may guide injection volumes for intravitreal injection and inform injection force considerations during technical drug product development.
To model and predict individual patient responses to radiation therapy.
We modeled tumor dynamics as logistic growth and the effect of radiation as a reduction in the tumor carrying capacity, motivated by the effect of radiation on the tumor microenvironment. The model was assessed on weekly tumor volume data collected for 2 independent cohorts of patients with head and neck cancer from the H. Lee Moffitt Cancer Center (MCC) and the MD Anderson Cancer Center (MDACC) who received 66 to 70 Gy in standard daily fractions or with accelerated fractionation. To predict response to radiation therapy for individual patients, we developed a new forecasting framework that combined the learned tumor growth rate and carrying capacity reduction fraction (δ) distribution with weekly measurements of tumor volume reduction for a given test patient to estimate δ, which was used to predict patient-specific outcomes.
The model fit data from MCC with high accuracy with patient-specific δ and a fixed tumor growth rate acrosents allowed for the accurate prediction of patient outcomes, which may inform treatment adaptation and personalization.
Radiation dermatitis is one of the most common acute toxicities induced by chemoradiation therapy (CRT) for head and neck cancer (HNC). The benefit of topical steroids in the management of radiation dermatitis is still unclear. This phase 3, multi-institutional, randomized, double-blind, placebo-controlled trial evaluated the efficacy and safety of topical steroids for radiation dermatitis inpatients with locally advanced HNC receiving CRT.
Eligible patients were scheduled to receive bilateral neck irradiation (≥66 Gy) with concurrent cisplatin (≥200 mg/m
) as definitive or postoperative CRT. These energy reserves are key for the development of decapods and their composition mainly depends on the type and quality of food available in their habitats. This study evaluated the lipid content and FA composition of three tissues (hepatopancreas, gills, and muscle) in two widely distributed, semi-terrestrial coastal crab species in Chile, Cyclograpsus cinereus from the upper intertidal and Hemigrapsus crenulatus from estuaries. This evaluation aimed to assess the physiological role of the bioenergetic reserves of these crabs, which tolerate fluctuating environmental conditions. Our results showed that both species had a higher lipid content in the hepatopancreas and a lower lipid content in its gills and muscle. All three of the evaluated tissues in C. cinereus showed high contents of saturated fatty acids (SFAs), and its hepatopancreas displayed the highest contents of monounsaturated (MUFAs) and polyunsaturated fatty acids (PUFAs).
