Nowadays, real-world data can be used to improve currently available dosing guidelines and to support regulatory approval of drugs for use in neonates by overcoming practical and ethical hurdles. This prospective, open-label, investigator-initiated study of azlocillin in neonates with EOS was conducted using an adaptive two-step design. First, a maturational pharmacokinetic-pharmacodynamic model of azlocillin was developed, using an empirical dosing regimen combined with opportunistic samples resulting from waste material. However, few studies have integrated multimodal imaging to simultaneously investigate relationships among structure, function, and cognition. We aimed to clarify how specifically DLPFC structure and function contribute to working memory in healthy older adults. In total, 138 participants aged 65-88 underwent 3 T neuroimaging and were divided into higher and lower groups based on a median split of in-scanner n-back task performance. Three a priori spherical DLPFC regions of interest (ROIs) were used to quantify blood-oxygen-level-dependent (BOLD) signal and FreeSurfer-derived surface area, cortical thickness, and white matter volume. Binary logistic regressions adjusting for age, sex, education, and scanner type revealed that greater left and right DLPFC BOLD signal predicted the probability of higher performing group membership (P values.05). Importantly, this suggests BOLD signal and surface area may independently contribute to working memory performance in healthy older adults.Aptamer-drug conjugates (ApDCs) are promising anticancer therapeutics with cancer cell specificity. However, versatile in vivo applications of ApDCs are hampered by their limited serum stability and inability to reach the tumour upon systemic administration. Here, we describe DNA nanoparticles of ApDCs as a platform for tumour-targeted systemic delivery of ApDCs. DNA nanoparticles of approximately 75 nm size were fabricated by self-assembly of a polymerised floxuridine (FUdR)-incorporated AS1411 aptamer produced via rolling circle amplification. The DNA nanoparticles of ApDCs showed highly efficient cancer cell uptake, enhanced serum stability, and tumour-targeted accumulation. These properties could be successfully utilised for tumour-specific apoptotic damage by ApDCs, leading to significant suppression of tumour growth without considerable systemic toxicity. Molecular analysis revealed that the enhanced anticancer potency was due to the synergic effect induced by the simultaneous activation of p53 by AS1411 and the inhibition of thymidylate synthase by FUdR, respectively, both of which were generated from the DNA nanoparticles. We therefore expect that the DNA nanoparticles of ApDCs can be a promising platform for tumour-targeted delivery of various nucleoside-incorporated ApDCs to treat cancer.A highly sensitive in situ method to detect bacterial pathogens is of utmost importance in preventing the outbreak of foodborne diseases. In this study, a simple method enabling the detection of a single bacterial cell in a sample was developed based on magnetic capture particles (CPs), and europium-fluorescent labeling particles (LPs) functionalized with antibodies. After mixing the sample with the particles in a sample tube, the sample tube was connected to an assay chip, where the CP-bacteria-LP complex was transported from the sample chamber to a detection chamber using a simple assay device. The number of bacteria was quantitatively determined by measuring the fluorescence emitted from the detection chamber. This assay method enabled the detection of a single cell of Vibrio parahaemolyticus from 0.1 mL pure broth culture samples within 30 min. A simple enrichment method that can be performed using only the vibrating action of the assay device without any additional instruments was also developed for the analysis of food samples. By analyzing the enriched sample using the assay method, we could detect V. parahaemolyticus quantitatively with a detection limit of 1 colony forming unit from oyster samples within 130 min. Due to simplicity of this methodology and the instrumentation involved, and its capability of rapid single-cell detection, it may be considered as an in situ method for the determination of food safety.Consumers expect perceptual constancy between multiple bites of the same food. In this study, we investigated how sweetness, creaminess, expected fullness and liking of chocolate coated rice waffles can be modified by bite-to-bite variation in chocolate thickness. 3D inkjet printing was used to accurately deposit the chocolate layers varying in thickness (0.8, 1.6 and 3.2 mm) onto rice waffles. In the first study, single bites of rice waffles with a homogeneous chocolate coating were evaluated. With increasing thickness of chocolate coating, the sweetness, creaminess, and expected fullness increased significantly. In the second study, we evaluated seven chocolate coated rice waffles containing a constant total chocolate amount but different chocolate thicknesses between three sequential bites. The order of chocolate thickness between bites had significant, but small effects on sweetness, expected fullness and liking. Interestingly, rice waffles with a homogeneous chocolate coating were preferred over rice waffles with an inhomogeneous chocolate coating.
