We study the propagation of in-plane elastic waves in a soft thin strip, a specific geometrical and mechanical hybrid framework which we expect to exhibit a Dirac-like cone. We separate the low frequencies guided modes (typically 100 Hz for a 1-cm-wide strip) and obtain experimentally the full dispersion diagram. Dirac cones are evidenced together with other remarkable wave phenomena such as negative wave velocity or pseudo-zero group velocity (ZGV). Our measurements are convincingly supported by a model (and numerical simulation) for both Neumann and Dirichlet boundary conditions. Finally, we perform one-way chiral selection by carefully setting the source position and polarization. Therefore, we show that soft materials support atypical wave-based phenomena, which is all of the more interesting as they make most of the biological tissues.Amorphous solids remain outside of the classification and systematic discovery of new topological materials, partially due to the lack of realistic models that are analytically tractable. Here we introduce the topological Weaire-Thorpe class of models, which are defined on amorphous lattices with fixed coordination number, a realistic feature of covalently bonded amorphous solids. Their short-range properties allow us to analytically predict spectral gaps. Their symmetry under permutation of orbitals allows us to analytically compute topological phase diagrams, which determine quantized observables like circular dichroism, by introducing symmetry indicators in amorphous systems. These models and our procedures to define invariants are generalizable to higher coordination number and dimensions, opening a route toward a complete classification of amorphous topological states in real space using quasilocal properties.BsYetJ is a bacterial homolog of transmembrane BAX inhibitor-1 motif-containing 6 (TMBIM6) membrane protein that plays a key role in the control of calcium homeostasis. However, the BsYetJ (or TMBIM6) structure embedded in a lipid bilayer is uncharacterized, let alone the molecular mechanism of the calcium transport activity. Herein, we report structures of BsYetJ in lipid nanodiscs identified by double electron-electron resonance spectroscopy. Our results reveal that BsYetJ in lipid nanodiscs is structurally different from those crystallized in detergents. We show that BsYetJ conformation is pH-sensitive in apo state (lacking calcium), whereas in a calcium-containing solution it is stuck in an intermediate, inert to pH changes. Only when the transmembrane calcium gradient is established can the calcium-release activity of holo-BsYetJ occur and be mediated by pH-dependent conformational changes, suggesting a dual gating mechanism. Conformational substates involved in the process and a key residue D171 relevant to the gating of calcium are identified. Our study suggests that BsYetJ/TMBIM6 is a pH-dependent, voltage-gated calcium channel.Polycomb repressive complex 2 (PRC2) installs and spreads repressive histone methylation marks on eukaryotic chromosomes. Because of the key roles that PRC2 plays in development and disease, how this epigenetic machinery interacts with DNA and nucleosomes is of major interest. Nonetheless, the mechanism by which PRC2 engages with native-like chromatin remains incompletely understood. In this work, we employ single-molecule force spectroscopy and molecular dynamics simulations to dissect the behavior of PRC2 on polynucleosome arrays. Our results reveal an unexpectedly diverse repertoire of PRC2 binding configurations on chromatin. Besides reproducing known binding modes in which PRC2 interacts with bare DNA, mononucleosomes, and adjacent nucleosome pairs, our data also provide direct evidence that PRC2 can bridge pairs of distal nucleosomes. In particular, the "1-3" bridging mode, in which PRC2 engages two nucleosomes separated by one spacer nucleosome, is a preferred low-energy configuration. Moreover, we show that the distribution and stability of different PRC2-chromatin interaction modes are modulated by accessory subunits, oncogenic histone mutations, and the methylation state of chromatin. Overall, these findings have implications for the mechanism by which PRC2 spreads histone modifications and compacts chromatin. Recent advances in deep learning allow neural networks to quickly compute these motions; however, they lack the precision required to produce kinematically and dynamically feasible motions. While infeasible, the network-computed motions approximate the optimized results. The proposed method warm starts the optimization process by using the approximate motions as a starting point from which the optimizing motion planner refines to an optimized and feasible motion with few iterations. In experiments, the proposed deep learning-based warm-started optimizing motion planner reduces compute and motion time when compared to a sampling-based asymptotically optimal motion planner and an optimizing motion planner. When applied to grasp-optimized motion planning, the results suggest that deep learning can reduce the computation time by two orders of magnitude (300×), from 29 s to 80 ms, making it practical for e-commerce warehouse picking.Millirobots that can adapt to unstructured environments, operate in confined spaces, and interact with a diverse range of objects would be desirable for exploration and biomedical applications. The continued development of millirobots, however, requires simple and scalable fabrication techniques. Here, we propose a minimalist approach to construct millirobots by coating inanimate objects with a composited agglutinate magnetic spray. Our approach enables a variety of one-dimensional (1D), 2D, or 3D objects to be covered with a thin magnetically drivable film (~100 to 250 micrometers in thickness). The film is thin enough to preserve the original size, morphology, and structure of the objects while providing actuation of up to hundreds of times its own weight. Under the actuation of a magnetic field, our millirobots are able to demonstrate a range of locomotive abilities crawling, walking, and rolling. Moreover, we can reprogram and disintegrate the magnetic film on our millirobots on demand. We leverage these abilities to demonstrate biomedical applications, including catheter navigation and drug delivery.
