CONCLUSION Quercetin can mitigate LPS-induced injury in HOKs by downregulating miR-22, thereby activating PI3K/AKT and JAK1/STAT3 cascades.Objective To evaluate the effect of individual lower urinary tract symptoms (LUTS) and LUTS-specific bother on daily/leisure activities, work productivity and treatment behaviors and satisfaction in a Brazilian population reporting symptoms of the overactive bladder (OAB) syndrome.Methods Secondary analysis of Brazil LUTS study data, including individuals ≥40 years old with a possible diagnosis of OAB, based on a score of ≥8 on the OAB-V8 questionnaire. Participants used a 5-point Likert scale to rate occurrence of LUTS during the previous month. Regression models were constructed to analyze association of symptom frequency and bother, controlled for demographics, comorbid conditions, habits and body mass index to outcomes related to people's lives and treatment patterns.Results This analysis included 5,184 individuals (53% female), 24.4% of whom received a possible diagnosis of OAB. There was a greater likelihood of OAB symptoms in men reporting depression/anxiety (2.0 times), diabetes (1.84 times), or constipation (1.88 times) and women reporting depression/anxiety (2.6 times), constipation (1.7 times), and being overweight (1.44 times) or obese (1.92 times). Symptoms of all categories, including voiding, storage, and post-micturition, were associated with a negative impact on individuals´ lives, quality of life and treatment-related outcomes. Treatment seeking for OAB was low among men and women overall (35.1% and 43.6%, respectively), with highest rates among individuals in the 60-69 age group.Conclusions LUTS of all categories impacted all domains studied. These results highlight the importance of comprehensive LUTS assessment in OAB patients, including voiding, storage and post-micturition symptoms.The effectiveness of a treatment on a count outcome can be assessed using a negative binomial regression, where treatment effects are defined as the difference between the expected outcome under treatment and under control. Recruiting enough participants is perhaps the most significant procedure that determines the success of medical research, and high participation rate brings about many benefits. Therefore, acquiring enough participants is important for medical researchers. To understand how to improve participation rate, we need to clarify factors affecting the public's attitude toward medical research. Through this review, we aim to examine which factors affect the attitude of human participants toward medical research.Methods The relevant data were searched by using the keywords "Public," "Participants," "Medical" and "Research" in PubMed (MEDLINE), International Pharmaceutical Abstracts, Web of Science, Science Direct, CINAHL Plus, EMBASE, and China Knowledge Resource Integrated databases. A manual search was done to acquire peer-reviewed articles and reports about participation in medical research.Results Sixty-three studies were identified for inclusion after full text screening. The included studies were of variable quality. Some factors affecting people's attitude toward participating in medical research have been identified and discussed in our review.Conclusion This review demonstrated that willingness of participants to take part in medical research was influenced by a variety of factors. These factors may be used to predict the public's willingness to take part in medical research and may potentially be used in developing strategies aimed at improving participation rate.The presence of 5-HT7r's in both human and rat cardiovascular and immune tissues and their contribution to inflammatory conditions prompted us to hypothesize that these receptors contribute in acute myocardial infarction (MI) with underlying chronic endothelial dysfunction. We investigated the role of 5-HT7 receptors on heart tissue that damaged by isoproterenol (ISO)-induced MI in rats with high-fat diet (HFD). In vitro and in vivo effects of 5-HT7r agonist (LP44) and antagonist (SB269970) have been investigated on the H9C2 cell line and rats, respectively. For in vivo analyses, rats were fed with HFD for 8 weeks and after this period ISO-induced MI model has been applied to rat. To investigate the role of 5-HT7r's, two different doses of LP44 and SB269970 were evaluated and compared with standard hypolipidemic agent, atorvastatin. In vitro studies showed that LP44 has protective and proliferative effects on rat cardiomyocytes. Also in in vivo studies stimulating 5-HT7r's by LP44 improved blood lipid profile (decreased total cholesterol, low-density lipoprotein-C, and triglyceride, increased high-density lipoprotein), decreased cardiac damage markers (creatine kinase and troponin-I), and corrected inflammatory status (tumor necrosis factor-α, interleukin-6). Our results showed significant improvement in LP44 administered rats in terms of histopathologic analyses. In damaged tissues, 5-HT7 mRNA expression increased and agonist administration decreased this elevation significantly. We determined for the first time that 5-HT7r's are overexpressed in ISO-induced MI of rats with underlying HFD-induced endothelial dysfunction. Restoration of this overexpression by LP44, a 5-HT7r agonist, ameliorated heart tissue in physiopathologic, enzymatic, and molecular level, showing the cardiac role of these receptors and suggesting them as future potential therapeutic targets.
