Although recent studies establish the role of RIOK2 in ribosome maturation and cell cycle progression, its biological functions remain poorly elucidated, hindering the potential to explore RIOK2 as a therapeutic target. Here, we report the discovery of CQ211, the most potent and selective RIOK2 inhibitor reported so far. CQ211 displays a high binding affinity (Kd = 6.1 nM) and shows excellent selectivity to RIOK2 in both enzymatic and cellular studies. It also exhibits potent proliferation inhibition activity against multiple cancer cell lines and demonstrates promising in vivo efficacy in mouse xenograft models. The crystal structure of RIOK2-CQ211 sheds light on the molecular mechanism of inhibition and informs the subsequent optimization. To this end, we present five basic precepts that have guided historical and sociological studies of "science in action" science as a collective endeavor, as a social activity, as a set of practices, as a process that involves controversies, and as a formative process. By examining the topic in the light of these five points, we demonstrate how the history of this successful research tradition can lead us to broader reflections about the complex dynamics interweaving science and society.In order to realize broad excitation and narrow emission red light phosphor, a new vanadium phosphate Ba2BiV2PO11 was selected as a host for Eu3+. Monitored at 619 nm, a wide band from 240 to 400 nm could be observed and inferred to be composed of Eu3+-O2- and V5+-O2- charge transfer bands, which could make it match well with the UV chip and the blue chip along with the characteristic excitation of Eu3+ at 465 nm. Under 354 nm excitation, the sample could emit high color purity red light, and the thermal quenching integral intensity showed good thermal stability. The generation of charge transfer bands was investigated in detail combined with the luminescence properties and the structure of the matrix. Moreover, the as-prepared phosphor could improve the white light performance of blue chip-activated YAGCe3+ and n-UV chip-activated tricolor phosphors. All the results indicated the multiple application potential of Ba2BiV2PO11Eu3+ for white light-emitting diodes.
Dendritic cells (DCs) are specialized antigen-presenting cells that play a critical role in the immune response against human papillomavirus (HPV) infection, and represent a therapeutic target in cancer.

To identify and quantify DCs in tonsillar squamous cell carcinoma (TSCC) under the influence of HPV infection.

CD1a and CD83 antibodies were used to identify immature dendritic cells and mature dendritic cells by immunohistochemistry in 33 primary TSCC and 10 normal tonsils (NTs), respectively. For the TSCC samples, the number of DCs per area was evaluated in the intra- and peritumoral compartments. For the NTs, the quantification of DCs was evaluated in the intra- and peritonsillar compartments. HPV detection methods were determined according to the ASCO Clinical Practice Guidelines from the College of American Pathologists Guideline (2018).

There were fewer intratumoral CD1a+ DCs in the HPV-positive and HPV-negative TSCC groups than in the NT group (p<0.05). In the peritumoral compartment, there were fewer CD83+ DCs in the HPV-positive and HPV-negative TSCC groups than in the NT group (p<0.001). The quantification of DCs subtypes showed no statistical differences between HPV-positive and HPV-negative TSCC groups (p>0.137). Patients with HPV-positive TSCC had significantly better overall survival rate than those with HPV-negative TSCC (p=0.004).

Data on physicians' income are relevant for well-informed health policies, both due to their strategic role and the volume of resources that their activities represent to health systems. In Brazil, multiple sources of data measure the income of these professionals, each one with singularities that generate a complex and heterogeneous picture. This study explores the methodological aspects of different data sources, pointing to potentials and limitations to measuring the income of physicians.

We use the sources' documentation and data on the average monthly income in 2019, by gender and macro region, from four distinct surveys Continuous Pnad (National Household Sample Survey), RAIS (Annual Listing of Social Information), Medical Demographics, and IRPF (Personal Income Tax).

The results confirm the heterogeneity of definitions, variables, and methodologies. The data set can evidence phenomena such as the income difference between men and women. Regional inequalities are evident; however, the data interpretation is less assertive.
