Geoffroea decorticans (chañar) is commonly used for culinary and medicinal purposes in rural communities. The aim of this work was to chemically characterize three Geoffroea decorticans extracts and determine their capacity to modulate the wnt/β-catenin pathway. This signaling pathway plays a key role in embryonic development but its overactivation leads to cancer cell growth. Phytochemical analysis of extracts showed presence of major classes of phytochemicals. Gas chromatography-mass spectrometry results revealed the presence of acids, esters and furanic compounds. Using Xenopus embryos as in vivo model organisms, we found that the extracts modulated dorso-ventral axis formation and rescued hyperdorsalized phenotypes produced by LiCl treatment. In agreement with these findings, Geoffroea decorticans extracts decreased β-catenin levels and suppressed the expression of wnt target genes such as xnr3 and chordin, thus demonstrating an inhibitory regulation of the wnt/β-catenin signaling pathway. All these results support a new role for Geoffroea decorticans fruit derivatives with possible anti-carcinogenic actions.Extracellular vesicles (EVs) are lipid bilayer particles that are released by various cells and provide a real-time snapshot of the state of these cells in tissue in a noninvasive manner. EVs contain components, including mRNA, miRNAs, proteins, and metabolites. Therefore, EVs hold promise for the discovery of liquid biopsy-based biomarkers for disease diagnosis. In the present study, metabolome analysis of urine EVs in rats with kidney injury caused by cisplatin and puromycin aminonucleoside was performed using liquid chromatography/mass spectrometry to identify candidate biomarkers that reflect the type and extent of injury in drug-induced nephrotoxicity. A total of 396 metabolites were detected in urine EVs, of which 65 were identified as potential biomarkers in urine EVs of drug-induced nephrotoxicity. Pathway analysis revealed that these metabolites may reflect changes occurring within damaged cells during kidney injury, suggesting that metabolomics of urine EVs could be a useful informative tool.Studies have demonstrated that stochastic configuration networks (SCNs) have good potential for rapid data modeling because of their sufficient adequate learning power, which is theoretically guaranteed. Empirical studies have verified that the learner models produced by SCNs can usually achieve favorable test performance in practice but more in-depth theoretical analysis of their generalization power would be useful for constructing SCN-based ensemble models with enhanced generalization capacities. In particular, given a collection of independently developed SCN-based learner models, it is useful to select certain base learners that can potentially obtain preferable test results rather than considering all of the base models together, before simply taking their average in order to build an effective ensemble model. In this study, we propose a novel framework for building SCN ensembles by exploring key factors that might potentially affect the generalization performance of the base model. Under a mild assumption, we provide a comprehensive theoretical framework for examining a learner model's generalization error, as well as formulating a novel indicator that contains measurement information for the training errors, output weights, and a hidden layer output matrix, which can be used by our proposed algorithm to find a subset of appropriate base models from a pool of randomized learner models. A toy example of one-dimensional function approximation, a case study for developing a predictive model for forecasting student learning performance, and two large-scale data sets were used in our experiments. The experimental results indicate that our proposed method has some remarkable advantages for building ensemble models.Studies have reported that emotional facial expression recognition (EFER) may be altered in individuals with depression. This study examined EFER in adolescent girls with and without depression and further examined associations between relevant clinical features of depression and EFER. Fifty adolescent girls aged 12 to 19 years old meeting criteria for depression or subthreshold levels of symptomatology and 55 adolescent girls with no psychiatric diagnosis completed EFER tasks. Reaction time and accuracy for recognising expressions at high and low intensities, and sensitivity in recognising happiness, sadness, anger and fear were assessed. Data were analysed using linear mixed models. Adolescents with depression were marginally faster than those in the comparison group to recognize sadness, although this trend disappeared once covarying for age and antidepressant use. Amongst adolescents with depression, clinical features were associated with poorer EFER performance. In contrast, anxiety symptoms were linked to better accuracy and heightened sensitivity towards happiness. A better understanding of EFER in adolescent girls with and without depression, and how clinical features might be associated with altered patterns of EFER could help to explain clinical heterogeneity observed in such studies of adolescents with depression. Knowledge of socio-cognitive alterations associated with depression will help to better develop and tailor interventions.Although effective in major depressive disorder (MDD), repetitive transcranial magnetic stimulation (rTMS) is costly and complex, limiting accessibility. To address this, we tested the feasibility of novel rTMS techniques with cost-saving opportunities, such as an open-room setting, large non-focal parabolic coils, and custom-built coil arms. We employed a low-frequency (LF) 1 Hz stimulation protocol (360 pulses per session), delivered on the most affordable FDA-approved device. MDD participants received an initial accelerated rTMS course (arTMS) of 6 sessions/day over 5 days (30 total), followed by a tapering course of daily sessions (up to 25) to decrease the odds of relapse. The self-reported Beck Depression Inventory II (BDI-II) was used to measure severity of depression. Forty-eight (48) patients completed the arTMS course. No serious adverse events occurred, and all patients reported manageable pain levels. Response and remission rates were 35.4% and 27.1% on the BDI-II, respectively, at the end of the tapering course. Repeated measures ANOVA showed significant changes of BDI-II scores over time. Even though our protocol will require further improvements, some of the concepts we introduced here could help guide the design of future trials aiming at increasing accessibility to rTMS.Synthesis of the bacterial cell envelope requires a regulated partitioning of resources from central metabolism. Varying SMF intensities (5-30 mT) were applied to the co-digestion digesters containing the optimum FA concentration (1.5 wt.%), which were selected from co-digestion systems with a varying FA (0-2 wt.%). All experimental groups exhibit the greater COD removal rates (51.56-64.19%) and cumulative biogas yields (604.14-671.64 mL/g VS) than reference group (37.77% and 433.19 mL/g VS). The digester with optimum FA concentration (1.5 wt.%) under 5 mT shows the highest biogas yield (671.64 mL/g VS), and exhibits superior digestate stability (45.4%) and fertility (7.01%) for fertilizer utilization. A powerful strategy for understanding the underlying mechanism of the SMF and FA accelerant in an enhanced AcoD system is proposed. This work documents an innovative technique for an enhanced AcoD system using the SMF coupled with FA accelerant.The aim of this work was to study the cellulose fibers deconstruction by twin-screw extrusion with in situ enzymatic hydrolysis via bioextrusion, for cellulose nanofibrils (CNF) production. Cellulose pulp was extruded with an optimized screw profile, with or without (reference) the addition of an enzymatic solution. An increase of crystallinity index from 67.0% to 73.7% and decrease of DP from 1003 to 419 were observed with bioextrusion. Direct activity measurements of the enzyme confirmed its activity during the process (sugar content increasing from 0.07 ± 0.004 to 2.38 ± 0.003 mg/mL) and after the process (specific activities around 0.20 CMCU/mL). Enzymes were not deactivated during bioextrusion and could be recycled. CNF properties were higher with bioextrusion compared to reference (respective quality indices of 55.5 ± 2.7 and 39.8 ± 2.8), with a lower energy consumption. This proof of concept could be optimized for the industrial production of highly concentrated CNF.Magnetic porous biochars (MCHCl, MCHAc) with nanostructure on surfaces were prepared from penicillin fermentation dregs by pyrolysis with K2FeO4 activation and used in penicillin adsorption. MCHCl and MCHAc had high BET surface areas of 672 and 735 m2/g, respectively; mainly be attributed to the activation of K2FeO4 as well as acid pickling. Saturation magnetizations of MCHCl and MCHAc were 75.29 and 42.45 emu/g, respectively; the magnetism was mainly derived from the Fe3O4 and Fe3C in magnetic biochars. MCHCl had nano sticks of ~ 80 nm and MCHAc had petal-like slice of ~ 30 nm on surfaces. The maximum adsorption capacities of penicillin on MCHCl and MCHAc were 196 and 322 mg/g at 308 K, respectively. The adsorptions of penicillin on MCHCl and MCHAc were consistent with pseudo primary kinetics and the Langmuir adsorption isotherm model, and thermodynamic analysis indicated that the adsorption mechanism included physical and chemical adsorption.Angiogenesis is a vital process during the regeneration of bone tissue. The aim of this study was to investigate angiogenesis at the fracture site as well as at distal locations from obesity-induced type 2 diabetic mice that were treated with bone morphogenetic protein-2 (BMP-2, local administration at the time of surgery) to heal a femoral critical sized defect (CSD) or saline as a control. Mice were fed a high fat diet (HFD) to induce a type 2 diabetic-like phenotype while low fat diet (LFD) animals served as controls. Endothelial cells (ECs) were isolated from the lungs (LECs) and bone marrow (BMECs) 3 weeks post-surgery, and the fractured femurs were also examined. Our studies demonstrate that local administration of BMP-2 at the fracture site in a CSD model results in complete bone healing within 3 weeks for all HFD mice and 66.7% of LFD mice, whereas those treated with saline remain unhealed. At the fracture site, vessel parameters and adipocyte numbers were significantly increased in BMP-2 treated femurs, irrespective of diet.
