Here we illustrate the outcomes of an extensive lead-generation campaign for identification of Mer inhibitors, focusing on the results from concurrent, orthogonal high-throughput screening approaches. Data mining, HT (high-throughput), and DECL (DNA-encoded chemical library) screens offered means to evaluate large numbers of compounds. We discuss campaign strategy and screening outcomes, and exemplify series resulting from prioritization of hits that were identified. Concurrent execution of HT and DECL screening successfully yielded a large number of potent, selective, and novel starting points, covering a range of selectivity profiles across the TAM family members and modes of kinase binding, and offered excellent start points for lead development.The exact cause of atherosclerosis is not known, and therefore, the current treatment options are limited. The activation of endothelial cells by oxidized low-density lipoprotein (ox-LDL) plays a key role in the initiation and progression of atherosclerosis. Phoenixin-20 is one of the newly identified neuropeptides with pleiotropic effects in the regulation of reproduction and other biological functions. G-protein receptor-coupled 173 (GPR173) is the putative receptor of Phoenixin-20. In the present study, we show that endothelial GPR173 is repressed upon ox-LDL stimulation in human aortic endothelial cells (HAECs). We further elaborate on the hypothesis that GPR173 could be involved in the pathogenesis of atherosclerosis through a series of experiments. Our results indicate that ox-LDL remarkably triggers the increase of ROS, NOX-4, pro-inflammatory cytokines IL-1β, IL-8, and MCP-1 expression, as well as adhesion molecules ICAM-1 and VCAM-1 release. However, the agonism of GPR173 using Phoenixin-20 significantly ameliorates all of these harmful effects from ox-LDL by suppressing the NF-κB pathway. Furthermore, we show that agonism of GPR173 by Phoenixin-20 prevents the attachment of monocytes THP-1 to endothelial cells, which is an important therapeutic approach to preventing atherogenesis. In conclusion, our study demonstrates that GPR173 agonism by Phoenixin-20 plays a protective role against ox-LDL-induced endothelial dysfunction, implying that Phoenixin-20 may have therapeutic implications in atherosclerosis.Tactile sensor arrays have attracted considerable attention for their use in diverse applications, such as advanced robotics and interactive human-machine interfaces. However, conventional tactile sensor arrays suffer from electrical crosstalk caused by current leakages between the tactile cells. The approaches that have been proposed thus far to overcome this issue require complex rectifier circuits or a serial fabrication process. This article reports a flexible tactile sensor array fabricated through a batch process using a mesh. A carbon nanotube-polydimethylsiloxane composite is used to form an array of sensing cells in the mesh through a simple "dip-coating" process and is cured into a concave shape. The contact area between the electrode and the composite changes significantly under pressure, resulting in an excellent sensitivity (5.61 kPa-1) over a wide range of pressure up to 600 kPa. The mesh separates the composite into the arranged sensing cells to prevent the electrical connection between adjacent cells and simultaneously connects each cell mechanically. Additionally, the sensor shows superior durability compared with previously reported tactile sensors because the mesh acts as a support beam. Furthermore, the tactile sensor array is successfully utilized as a Braille reader via information processing based on machine learning.A series of hole transporting materials (HTMs) with fused tetraphenylethylene cores (9,9'-bifluorenylidene and dibenzo[g,p]chrysene) as well as different substitution positions of arylamine side arms has been designed and synthesized. A reference HTM with a non-fused tetraphenylethylene core is also prepared for a comparative study. It is noted that fused tetraphenylethylene molecules show a bathochromic spectral shift, electronegative character, and lower reorganization energies than the non-fused ones. Furthermore, the molecules with side arms located on the meta-position on the tetraphenylethylene core in terms of a double bond exhibit a deeper highest occupied molecular orbital level than those of the para-position-based ones whether tetraphenylethylene is fused or not. Moreover, the reorganization energies of fused meta-position-based HTMs are lower than those of para-position-based HTMs. Fused tetraphenylethylene HTMs own a better hole-extraction capability than the non-fused ones. When used in perovskite solar cells, all devices with fused tetraphenylethylene HTMs display better performance than those of the non-fused ones. The HTMs based on dibenzo[g,p]chrysene exhibit better performance than those of bifluorenylidene. Moreover, the devices with HTMs with side arms located on the meta-position on the tetraphenylethylene core display higher power conversion efficiency than those of the para-position-based ones. Eprenetapopt cell line The results give some new insight and reference to develop ideal HTMs for perovskite solar cells.Mn-based oxides exhibit outstanding low-temperature activity for the selective catalytic reduction of NO x with NH3 (NH3-SCR) compared with other catalysts. However, the underlying principle responsible for the excellent low-temperature activity is not yet clear. Here, the atomic-level mechanism and activity-limiting factor in the NH3-SCR process over Mn-, Fe-, and Ce-based oxide catalysts are elucidated by a combination of first-principles calculations and experimental measurements. We found that the superior oxidative dehydrogenation performance toward NH3 of Mn-based catalysts reduces the energy barriers for the activation of NH3 and the formation of the key intermediate NH2NO, which is the rate-determining step in NH3-SCR over these oxide catalysts. The findings of this study advance the understanding of the working principle of Mn-based SCR catalysts and provide a fundamental basis for the development of future generation SCR catalysts with excellent low-temperature activity.
