ILCs accounted for 179/622 (28.8%) cases. In this article, we selected three rocks (limestone, shale, sandstone) and two coals (bituminous coal and anthracite coal) to form combined coal-rock, and analyze their dynamic mechanical properties by using the SHPB system. We find that the dynamic compressive strength and elastic modulus of combined rock-coal are lower than the average value of single rock and coal, while the ultimate strain and strain rate of combined coal-rock are higher than the average values of single rock and coal. Compressive strength and elastic modulus of the combined body increase gradually with increasing confining pressure, and the strain decreases accordingly. The dynamic stress-strain curve demonstrates an obvious double-peak at high strain rate (85.55 s-1 and above in the present work), while there is no obvious double-peak of the curve at low strain rate. Dynamic compressive strength of combined coal-rock body increases significantly with increasing confining pressure at low strain rate, but it increases more smoothly at higher strain rate. The elastic modulus also increases with increasing confining pressure, and it seems to be stable as confining pressure increases at low strain rate. The ultimate strain decreases gradually with increasing confining pressure but more gently compared with that at low strain rate. Besides, longitudinal fractures of combined coal-rock bodies include penetrating fractures, partially penetrating fractures, and interrupted fractures stopped at the coal-rock interface. The dynamic mechanical response of combined coal-rock is of guiding significance for maintaining the stability of the roadway and formulating the support measures for the roadway.We evaluated the effect of Acetyl-cholinesterase-inhibitors (AChEIs) on cognitive decline and overall survival in a large sample of older patients with late onset Alzheimer's disease (LOAD), vascular dementia (VD) or Lewy body disease (LBD) from a real world setting. Patients with dementia enrolled between 2005 and 2020 by the "Alzheimer's Disease Research Centers" were analysed; the mean follow-up period was 7.9 years. A 11 propensity score matching was performed generating a cohort of 1.572 patients (786 treated [AChEIs +] and 786 not treated [AChEIs-] with AChEIs. The MMSE score was almost stable during the first 6 years of follow up in AChEIs + and then declined, while in AChEIs- it progressively declined so that at the end of follow-up (13.6 years) the average decrease in MMSE was 10.8 points in AChEIs- compared with 5.4 points in AChEIs + (p  less then  0.001). This trend was driven by LOAD (Δ-MMSE-10.8 vs. -5.7 points; p  less then  0.001), although a similar effect was observed in VD (Δ-MMSE-11.6 vs. -8.8; p  less then  0.001). No effect on cognitive status was found in LBD. At multivariate Cox regression analysis (adjusted for age, gender, dependency level and depression) a strong association between AChEIs therapy and lower all-cause mortality was observed (H.R.0.59; 95%CI 0.53-0.66); this was confirmed also in analyses separately conducted in LOAD, VD and LBD. Among older people with dementia, treatment with AChEIs was associated with a slower cognitive decline and with reduced mortality, after a mean follow-up of almost eight years. Our data support the effectiveness of AChEIs in older patients affected by these types of dementia.The effects of organic fertilizers, based on Tithonia diversifolia, and in-organic fertilizers, based on hydroponics fertilizer, were evaluated on the performance, leaves, and fruit mineral concentrations of tomato (Solanum lycopersicum) and cucumber (Cucumis sativus) plants grown under soilless medium in a screen house. The treatments comprised six levels of liquid organic fertilizer (5, 15, 25, 35, 45, 55 mL), in-organic fertilizer, and a control. Both organic and in-organic fertilizers increased the growth, yield, leaf nutrient concentration, and the mineral contents of tomato and cucumber fruits in comparison with the control. In-organic fertilizer enhanced the performance and mineral concentrations of tomato and cucumber fruits in comparison with organic fertilizer. However, leaf analyses showed that all the essential elements for both tomato and cucumber crops were within the adequate ranges in the organic fertilizer treatments suggesting that this organic fertilizer can be used as an alternative to the expensive and scarce in-organic fertilizer. For organic fertilizer, the highest yield and mineral contents in fruits were attained at doses of 35 mL and 25 mL for tomato and cucumber plants, respectively. At these doses, the fruit weights of tomato and cucumber were increased by 137 and 198%, respectively, in comparison with the control. For a good yield of tomato and cucumber crop with a high fruit mineral content under the soilless medium of coco peat and rice husk, 35 mL and 25 mL of our tested organic fertilizer are recommended.Hematopoietic stem cells (HSCs) are crucial for the continuous production of blood cells during life. The transplantation of these cells is one of the most common treatments to cure patient suffering of blood diseases. However, the lack of suitable donors is a major limitation. One option to get HSCs matching perfectly a patient is cellular reprogramming. HSCs emerge from endothelial cells in blood vessels during embryogenesis through the endothelial to hematopoietic transition. Here, we used single-cell transcriptomics analysis to compare embryonic and post-natal endothelial cells to investigate the potential of adult vasculature to be reprogrammed in hematopoietic stem cells. Although transcriptional similarities have been found between embryonic and adult endothelial cells, we found some key differences in term of transcription factors expression. There is a deficit of expression of Runx1, Tal1, Lyl1 and Cbfb in adult endothelial cells compared to their embryonic counterparts. Using a combination of gene expression profiling and gene regulatory network analysis, we found that endothelial cells from the pancreas, brain, kidney and liver appear to be the most suitable targets for cellular reprogramming into HSCs. Overall, our work provides an important resource for the rational design of a reprogramming strategy for the generation of HSCs.PCSK9 is an effective target for lowering LDL-c. Previously, a camelid-human chimeric heavy chain antibody VHH-B11-Fc targeting human PCSK9 was designed. It had a potent hypolipidemic effect. However, the nanobody VHH-B11 interacts with PCSK9 at low affinity, while camelid VHH exhibits some immunogenicity. Moreover, the interacting epitope is yet to be identified, although VHH-B11 was shown to have distinct hPCSK9-binding epitopes for Evolocumab. This might impede the molecule's progress from bench to bedside. In the present study, we designed various configurations to improve the affinity of VHH-B11 with hPCSK9 ( less then  10 nM) that in turn enhanced the druggability of VHH-B11-Fc. Then, 17 amino acids were specifically mutated to increase the degree of humanization of the nanobody VHH-B11. Using phage display and sequencing technology, the linear epitope "STHGAGW" (amino acids 447-452) was identified in the hinge region of PCSK9 as the interacting site between VHH-B11-Fc and hPCSK9. Deciphering the interplay of these processes requires high-resolution, three-dimensional, quantitative imaging of chromatin structure in vitro. Herein, we introduce ChromSTEM, a method that utilizes high-angle annular dark-field imaging and tomography in scanning transmission electron microscopy combined with DNA-specific staining for electron microscopy. We utilized ChromSTEM for an in-depth quantification of 3D chromatin conformation with high spatial resolution and contrast, allowing for characterization of higher-order chromatin structure almost down to the level of the DNA base pair. Employing mass scaling analysis on ChromSTEM mass density tomograms, we observed that chromatin forms spatially well-defined higher-order domains, around 80 nm in radius. Within domains, chromatin exhibits a polymeric fractal-like behavior and a radially decreasing mass-density from the center to the periphery. Unlike other nanoimaging and analysis techniques, we demonstrate that our unique combination of this high-resolution imaging technique with polymer physics-based analysis enables us to (i) investigate the chromatin conformation within packing domains and (ii) quantify statistical descriptors of chromatin structure that are relevant to transcription.
