Drug resistance and damage caused to the normal cells are the drawbacks which have limited the use of the existing effective anticancer drugs. Attainment of a steady and extended release by encapsulating dual drugs into biocompatible and biodegradable vehicles is the key to enable the use of these drugs for effective inhibition of cancer. In this study, carboxymethyl chitosan (CMCS), a proficient water-soluble derivative of chitosan has been synthesized using chemical route and used for the delivery of 5-Fluorouracil and doxorubicin individually as well as in combination. Carboxymethylation occuring at -NH2 and OH sites of chitosan, has been confirmed using FTIR. EDX and Fluorescence studies elucidate the encapsulation of 5-Fluorouracil and doxorubicin into CMCS. The capability of CMCS to release the drugs in a more sustained and prolonged manner is evident from the obtained release profiles. About 14.9 µg/ml is enough to cause 50% cell death by creating oxidative stress and effectuating DNA fragmentation. Amidst the existing reports, the uniqueness of this work lies in using this rare coalition of drugs for the suppression of breast cancer and in reducing the side effects of drugs by encapsulating them into CMCS, which is evidenced by the high hemocompatibilty of the samples.Deletion of dystrobrevin binding protein 1 has been linked to Hermansky-Pudlak syndrome type 7 (HPS-7), a rare disease characterized by oculocutaneous albinism and retinal dysfunction. We studied dysbindin-1 null mutant mice (Dys-/-) to shed light on retinal neurodevelopment defects in HPS-7. We analyzed the expression of a focused set of miRNAs in retina of wild type (WT), Dys+/- and Dys-/- mice. We also investigated the retinal function of these mice through electroretinography (ERG). We found that miR-101-3p, miR-137, miR-186-5p, miR-326, miR-382-5p and miR-876-5p were up-regulated in Dys-/-mice retina. Dys-/- mice showed significant increased b-wave in ERG, compared to WT mice. Bioinformatic analysis highlighted that dysregulated miRNAs target synaptic plasticity and dopaminergic signaling pathways, affecting retinal functions of Dys-/- mice. Overall, the data indicate potential mechanisms in retinal neurodevelopment of Dys-/- mice, which may have translational significance in HSP-7 patients, both in terms of diagnostic/prognostic biomarkers and novel pharmacological targets.This study examined associations between muscle strength and mental health among 2,652 elderly Koreans who participated in the 2015 and 2017 Korea National Health and Nutrition Examination Survey. We measured absolute handgrip strength and calculated handgrip strength relative to body mass index. Dynapenia criteria followed that of the Asian Working Group for Sarcopenia. Questionnaires were used to assess mental health indicators including suicidal ideation, depressed mood, and stress status. Among participants, 18.6%, 14.9%, and 6.7% reported stress, depressed mood, and suicidal ideation, respectively. Dynapenia prevalence was 25.1%. Adjusted odds ratios of stress, depressed mood, and suicidal ideation for men with dynapenia were 2.15, 2.30, and 2.11, respectively. Significant associations were absent among women. For men, handgrip strength and relative handgrip strength were inversely associated with risk of stress, depressed mood, and suicidal ideation. For women, handgrip strength and relative handgrip strength were inversely associated with the risk of stress, but not of depressed mood and suicidal ideation. In conclusion, all muscle-strength indices were associated with mental health risks among older men. In older women, low handgrip strength and relative handgrip strength were associated with increased stress. Muscle strength could be a clinical marker of poor mental health in older adults.The estrogen signaling pathway has been reported to modulate prostate cancer (PCa) progression through the activity of estrogen receptors α and β (ERα and ERβ). Given that selective estrogen receptor modulators (SERMs) are used to treat breast cancer, ERs have been proposed as attractive therapeutic targets in PCa. However, many inconsistencies regarding the expression of ERs and the efficacy of SERMs for PCa treatment exist, notably due to the use of ERβ antibodies lacking specificity and treatments with high SERM concentrations leading to off-target effects. To end this confusion, our objective was to study the impact of estrogenic and anti-estrogenic ligands in well-studied in vitro PCa models with appropriate controls, dosages, and ER subtype-specific antibodies. When using physiologically relevant concentrations of nine estrogenic/anti-estrogenic compounds, including five SERMs, we observed no significant modulation of PCa cell proliferation. Using RNA-seq and validated antibodies, we demonstrate that these PCa models do not express ERs. In contrast, RNA-seq from PCa samples from patients have detectable expression of ERα. Overall, our study reveals that commonly used PCa models are inappropriate to study ERs and indicate that usage of alternative models is essential to properly assess the roles of the estrogen signaling pathway in PCa.The ability to prevent blood loss in response to injury is a conserved function of all vertebrates. Complete deficiency of the central clotting enzyme prothrombin has never been observed in humans and is incompatible with postnatal life in mice, thus limiting the ability to study its role in vivo. Zebrafish are able to tolerate severe hemostatic deficiencies that are lethal in mammals. We have generated a targeted genetic deletion in the kringle 1 domain of zebrafish prothrombin. Homozygous mutant embryos develop normally into the mid-juvenile stage but demonstrate complete mortality by 2 months of age primarily due to internal hemorrhage. Mutants are unable to form occlusive venous and arterial thrombi in response to endothelial injury, a defect that was phenocopied using direct oral anticoagulants. Human prothrombin engineered with the equivalent mutation exhibits a severe reduction in secretion, thrombin generation, and fibrinogen cleavage. Together, these data demonstrate the conserved function of thrombin in zebrafish and provide insight into the role of kringle 1 in prothrombin maturation and activity. Understanding how zebrafish are able to develop normally and survive into early adulthood without thrombin activity will provide important insight into its pleiotropic functions as well as the management of patients with bleeding disorders.Ferritin protein is involved in biological tissues in the storage and management of iron - an essential micro-nutrient in the majority of living systems. While there are extensive studies on iron-loaded ferritin, its functionality in iron delivery is not completely clear. Here, for the first time, differential pulse voltammetry (DPV) has been successfully adapted to address the challenge of resolving a cascade of fast and co-occurring redox steps in enzymatic systems such as ferritin. Using DPV, comparative analysis of ferritins from two evolutionary-distant organisms has allowed us to propose a stepwise resolution for the complex mix of concurrent redox steps that is inherent to ferritins and to fine-tune the structure-function relationship of each redox step. Indeed, the cyclic conversion between Fe3+ and Fe2+ as well as the different oxidative steps of the various ferroxidase centers already known in ferritins were successfully discriminated, bringing new evidence that both the 3-fold and 4-fold channels can be functional in ferritin.Fibroblast-like transformation of retinal pigment epithelial (RPE) cells is a pathological feature of proliferative vitreoretinopathy (PVR) that may cause blindness. The effect of oxidative stress alone or together with transforming growth factor-beta 2 (TGF-β2) on epithelial-mesenchymal transformation (EMT) is not fully understood in RPE. TGF-β2 induced the upregulation EMT markers including α-smooth muscle actin (α-SMA), Snail, and Slug and downregulation of E-cadherin (E-cad) in ARPE-19 cells. Hydrogen peroxide (H2O2) not only upregulated α-SMA but also enhanced the effect of TGF-β2 on the expression of Snail and Slug. The CXCL family of cytokines could be the mediators of EMT induced by H2O2 and TGF-β2. H2O2 induced CXCL1, that upregulated α-SMA and fibronectin. Both SB225002, an inhibitor of CXCR2, and antioxidant N-acetylcysteine suppressed the TGF-β2-induced EMT in ARPE-19 cells. Taken together, the results suggest that oxidative stress enhanced TGF-β2-induced EMT through the possible autocrine effect of CXCL1 on CXCR2 in ARPE-19 cells.The massive use of fossil fuels since the industrial revolution has led to a rapid increase in the concentration of carbon dioxide (CO2) in the atmosphere. What effects elevated CO2 concentrations (ECO2) have on the defense mechanisms plants employ against insects remains poorly understood. This study showed that ECO2 of 750 ± 20 mmol/mol, increased the photosynthetic rate and biomass gain of tobacco and melon plants. However, while mass gain of Spodoptera litura, a nocturnal moth in the Noctuidae family, was higher when feeding on tobacco plants under ECO2, mass gain of Diaphania indica was reduced when feeding on melon plant at ECO2 compared to ambient CO2. Plants have many mechanisms to defend themselves against insects. Jasmonic acid (JA) is a crucial element of plant defense against lepidopteran insects. Our study showed that JA levels increased in tobacco plants under ECO2 but decreased in melon plants. 4-Methylumbelliferone It is speculated that ECO2 changes plant resistance to insects mainly by affecting the JA signaling pathway. Nutrient analysis suggested defensive metabolites rather than changes in the total nitrogen or protein content of the plants led to the changes in plant defense levels under ECO2. In summary, ECO2 affects the interaction between plants and insects. The results may provide a theoretical basis for studying the changes in crop resistance to pests under ECO2 and predicting the impact of ECO2 on future agro-ecosystems.We recently reported a sugar-induced bacterial release of 13-Docosenamide and its ability to quench fluorescein. This simple handle to monitor bacterial growth is readily applicable to develop a quicker antibiotic sensitivity testing method along with a low-cost field-use optical instrumentation. Conditions were standardized to perform this new procedure in the most preferred and CLSI-recommended microdilution format in 12-well strips. A simple and portable optoelectronic prototype was used to capture the image and read the fluorescence signal of the culture medium of the 12-well strips. This new Fluorescence Quenching Method along with the device enabled the choice of the right antibiotic within 8 h of sample collection from the patient. It was compliant to the Clinical Laboratory Standard Institute's quality control guidelines. Clinical assessment of the method using 440 urine samples from Urinary Tract Infection patients against 21 routinely used antibiotics showed a 94.3% match with the results of the Standard Disk Diffusion method. This new method saves the precious time taken for and the cost of antibiotic susceptibility testing for quicker and effective treatment with better compliance.
