Breast cancer continues to be the most commonly diagnosed cancer among Canadian women, with as many as 25-60% of women suffering from chronic neuropathic pain (CNP) as a pervasive consequence of treatment. While pharmacological interventions have shown limited efficacy for the management of CNP to date, psychological interventions, such as mindfulness-based stress reduction (MBSR), may be a promising alterative for improving pain-related problems. The purpose of this study was to use brain imaging methods to investigate this potential.

Resting-state fMRI was used in female breast cancer survivors with CNP before and after an 8-week MBSR course (n = 13) and compared with a waitlist control group (n = 10).

Focusing on the default mode network, the most significant results show greater posterior cingulate connectivity with medial prefrontal regions post-MBSR intervention. Moreover, this change in connectivity correlated with reduced pain severity for the MBSR group.

These results provide empirical evidence of a change in the brain following MBSR intervention associated with changes in the subjective experience of pain.

Microbial infection stimulates neutrophil/macrophage/monocyte extracellular trap formation, which leads to the release of citrullinated histone H3 (CitH3) catalyzed by peptidylarginine deiminase (PAD) 2 and 4. Understanding these molecular mechanisms in the pathogenesis of septic shock will be an important next step for developing novel diagnostic and treatment modalities. We sought to determine the expression of CitH3 in patients with septic shock, and to correlate CitH3 levels with PAD2/PAD4 and clinically relevant outcomes.

Levels of CitH3 were measured in serum samples of 160 critically ill patients with septic and non-septic shock, and healthy volunteers. Analyses of clinical and laboratory characteristics of patients were conducted.

Levels of circulating CitH3 at enrollment were significantly increased in septic shock patients (n = 102) compared to patients hospitalized with non-infectious shock (NIC) (n = 32, p < 0.0001). The area under the curve (95% CI) for distinguishing septic shock from NIC using CitH3 was 0.76 (0.65-0.86). CitH3 was positively correlated with PAD2 and PAD4 concentrations and Sequential Organ Failure Assessment Scores [total score (r = 0.36, p < 0.0001)]. The serum levels of CitH3 at 24h (p < 0.01) and 48h (p < 0.05) were significantly higher in the septic patients that did not survive.

Mitral regurgitation (MR) is the second most common valve disease in industrialized countries. Despite its high prevalence, little is known about its impact on the flow dynamics in the left ventricle (LV). Because of the interdependence between valvular function and hemodynamics in the heart chambers, an exploration of the dynamics in the LV could lead to a diagnosis of MR. This in vitro study aimed to develop an advanced left heart simulator capable of reproducing several conditions of MR and to evaluate their impact on the LV flow dynamics in terms of flow structures and viscous energy dissipation (VED).

A simulator, previously developed to test mechanical and biological valves, was upgraded with an original anatomically-shaped mitral valve made from a hydrogel. The valve can be used in healthy or pathological configurations. The nature and severity of the disease was controlled by applying specific strain to the chordae. In this study, in addition to a healthy condition, two different severities of MR were investigated moderate MR and severe MR. Planar time-resolved particle image velocimetry measurements were performed in order to evaluate the velocity field in the LV and the VED induced by each condition.

Our results showed that MR led to flow disturbances in the LV that were characterized by an increase in mitral inflow velocity and by elevated values of VED. Interestingly VED increased in proportion to the severity of MR and with a dissipation predominating during systole.

Considering these results, the introduction of new parameters based on LV VED could provide crucial information regarding the coupling between the mitral valve and the LV and allow for a better stratification of patients with MR.

The endothelial glycocalyx (GCX) plays a critical role in the health of the vascular system. Degradation of the GCX has been implicated in the onset of diseases like atherosclerosis and cancer because it disrupts endothelial cell (EC) function that is meant to protect from atherosclerosis and cancer. Examples of such EC function include interendothelial cell communication via gap junctions and receptor-mediated interactions between endothelial and tumor cells. This review focuses on GCX-dependent regulation of these intercellular interactions in healthy and diseased states. The ultimate goal is to build new knowledge that can be applied to developing GCX regeneration strategies that can control intercellular interaction in order to combat the progression of diseases such as atherosclerosis and cancer.

In vitro and in vivo studies were conducted to determine the baseline expression of GCX in physiologically relevant conditions. Chemical and mechanical GCX degradation approaches were employed to degrade the GCX. The impact of intact versus degraded GCX on intercellular interactions was assessed using cytochemistry, histochemistry, a Lucifer yellow dye transfer assay, and confocal, intravital, and scanning electron microscopy techniques.

Relevant to atherosclerosis, we found that GCX stability determines the expression and functionality of Cx43 in gap junction-mediated EC-to-EC communication. Relevant to cancer metastasis, we found that destabilizing the GCX through either disturbed flow-induced or enzyme induced GCX degradation results in increased E-selectin receptor-mediated EC-tumor cell interactions.

Our findings lay a foundation for future endothelial GCX-targeted therapy, to control intercellular interactions and limit the progression of atherosclerosis and cancer.

In this research, we used an electrophysiological index of working memory (contralateral delay activity) to passively measure working memory activity during visual search. Participants were asked to indicate whether a novel target was present or absent in a lateralized search array with three visual set sizes (2, 4, or 6). If rejected distractors are maintained in working memory during search, working memory activity should increase with the number of distractors that need to be evaluated. Therefore, we predicted the amplitude of the contralateral delay activity would be larger for target-absent trials and would increase with visual set size until WM capacity was reached. In Experiment 2, we found partial evidence in support of distractor maintenance during search for stimuli with high target/distractor similarity. Events presented in static blurry grayscale displays have been shown to be recognized only when and after presented with optic flow. In this study, we investigate the effects of optic flow and color on identifying blurry events by studying the identification accuracy and eye-movement patterns. Three types of color displays were tested grayscale, original colors, or rearranged colors (where the RGB values of the original colors were adjusted). In each color condition, participants identified 12 blurry events in five experimental phases. In the first two phases, static blurry images were presented alone or sequentially with a motion mask between consecutive frames, and identification was poor. In Phase 3, where optic flow was added, identification was comparably good. In Phases 4 and 5, motion was removed, but identification remained good. Thus, optic flow improved event identification during and after its presentation. Color also improved performance, where participants were consistently better at identifying color displays than grayscale or rearranged color displays. Importantly, the effects of optic flow and color were additive. Finally, in both motion and postmotion phases, a significant portion of eye fixations fell in strong optic flow areas, suggesting that participants continued to look where flow was available even after it stopped. We infer that optic flow specified depth structure in the blurry image structure and yielded an improvement in identification from static blurry images.It is well established that ensemble coding is regulated by physical similarity and variance in a set of stimuli. For example, observers are more accurate at judging the mean size of objects in a set if the overall size variance in the set is small. However, sometimes similarity among set members can be purely subjective. For example, faces from another race tend to look more similar than faces from one's own race. Very little is known about whether such subjective similarity also regulates ensemble coding in the same manner as objective similarity. To investigate this question, we had British and Chinese participants view sets of four faces that were of either own-race or other-race, own-gender or other-gender. After viewing each set the task was to judge whether a test face was presented in the set. Our results showed that, as demonstrated in prior research, participants often mistook a morphed set average to be a member of the set. Critically, this tendency to average a face set was not stronger for other-race faces.
