HF bulls was not affected by fertility status. In conclusion, while differences in field fertility among AI sires used in this study were not reflected in fertilization rate, differences in embryo quality were apparent as early as d 7. These differences likely contributed to the higher proportion of conceptuses surviving to d 15 in HF bulls.The objective of the study was to evaluate the effect of hygiene measures in automatic milking units on the transmission of 3 mastitis pathogens considered to be mainly or partly transmitted from cow to cow during milking events. Two studies were conducted as within-herd experimental trials in 2 Danish commercial dairy herds (A and B) with automatic milking systems. Interventions to enhance hygiene were implemented on the automatic milking units. The 2 studies evaluated separate interventions. In herd A, the hygiene interventions were manual wash with the Lely foam unit and adjustments on the brush-mediated teat cleaning procedure. In herd B, the hygiene intervention included automatic disinfection spray on the upper surface of the brush motor and daily change of brushes. Composite milk samples were collected longitudinally at 3- or 4-wk intervals from all lactating cows. Additional milk samples were taken from cows entering or leaving the study groups. Milk samples were analyzed with quantitative PCR. A hidden Markov model implemented within a Bayesian framework was used to estimate the transmission probability. For analysis, 701 samples from 156 cows were used for herd A, and 1,349 samples from 390 cows were used for herd B. In the intervention group in herd B, transmission of Streptococcus agalactiae was reduced to 19% (95% posterior credibility interval 0.00-64%) of the transmission in the control group, whereas transmission of Streptococcus dysgalactiae was reduced to 17% (95% posterior credibility interval 0.00-85%) of transmission in the control group. This suggests that automatic spray on the upper surface of the brush motor with disinfectant along with daily change of brushes collectively reduced transmission of Strep. agalactiae and Strep. dysgalactiae. Results on Staphylococcus aureus in herd B and results on manual foam cleaning and brush-mediated teat cleaning adjustments in herd A were inconclusive.Functional dyspepsia (FD) is a common disease of the digestive system and probiotics have been reported to be effective in the treatment of functional gastrointestinal diseases. The aim of this study was to determine the effect of the beverage containing Lactobacillus paracasei LC-37 (LC-37) and its ability to relieve symptoms of FD by a randomized clinical trial. Due to the mechanistic complexity underlying FD, intestinal microbiota and stool metabolomes were also evaluated. The results showed that FD was relieved in participants after treatment with the beverage containing LC-37 for 14 and 28 d. The clinical symptom scores were significantly decreased after these times (abdominal pain and belching were significantly decreased after 14 d and almost absent after 28 d of treatment). Probiotics, such as those containing the Lactobacillus, Lactococcus, and Weissella, significantly increased, and the abundance of harmful bacteria such as Lachnocliostridium significantly decreased. Furthermore, relevant beneficial intestinal metabolites such as pelargonic acid, benzoic acid, and short-chain fatty acids increased, and harmful intestinal metabolites such as hippuric acid decreased. Taken together, these findings suggested that the beverages containing LC-37 can increase the abundance of probiotics and decrease pathogenic bacteria, and thereby improve beneficial intestinal metabolites. Therefore, the beverages containing LC-37 may provide a natural alternative to combat FD.Extracellular vesicles (EV) are important for delivering biologically active substances to facilitate cell-to-cell communication. UAMC-3203 Milk-derived EV are widely known because of their potential for immune enhancement. However, procedures for isolating milk-derived EV have not been fully established. To obtain pure milk-derived EV and accurately reveal their function, such procedures must be established. The aim of the present study was to compare methods using commercially available kits for isolating milk-derived EV. Initially, we investigated procedures to remove casein, which is the major obstacle in determining milk-derived EV purity. We separated whey using centrifugation only, acetic acid precipitation, and EDTA precipitation. Then, we isolated milk-derived EV by ultracentrifugation, membrane affinity column, size exclusion chromatography (SEC), polymer-based isolation, or phosphatidylserine-affinity isolation. Using EV count per milligram of protein, which is a good indicator of purity, we determined that acetic acid precipitation was the best method for removing casein. Using nanoparticle tracking analysis, protein quantity analysis, and RNA quantity analysis, we comprehensively compared each isolation method for its purity and yield. We found that SEC-based qEV column (Izon Science) could collect purer milk-derived EV at higher quantities. Thus, a combination of acetic acid precipitation and qEV can effectively isolate high amounts of pure extracellular vesicles from bovine milk.The transport of young calves is a welfare concern. Young calves aged ≤3 mo are particularly vulnerable to compromised welfare, and are at a relatively high risk of morbidity and mortality compared with adult cattle. Calves face several potential challenges to welfare during and after transport, including food and water deprivation, disease, injury, and stress from handling, social mixing, and new environments. The key risk factors identified for poor calf welfare associated with transport include long transport and fasting durations, young age at transport, poor colostral immunity, timing within the calving season, lack of bedding in trucks, and high stocking density. Maximizing calf welfare thus requires a multifaceted approach, such as minimizing transport and fasting durations, transporting at an appropriate stocking density with comfortable bedding, only transporting calves that are healthy and fit, and optimizing pre-transport calf management. More research is needed to understand the effect of transport on the mental or affective state of calves.In the present study 114 lactic acid bacteria strains, isolated from raw mare milks from pastoral areas for ethnic minorities in northwest China, were screened for probiotic traits, and their characteristics were compared with those of Lactobacillus rhamnosus GG, a commercial strain. Among the 114 strains identified, the most common species was Pediococcus pentosaceus (n = 52), followed by Leuconostoc lactis (n = 35), Lactobacillus helveticus (n = 7), Lactobacillus plantarum (n = 6), Lactobacillus kefiri (n = 5), Lactobacillus curvatus (n = 4), Lactobacillus paracasei (n = 3), and Lactococcus garvieae (n = 3). Based on acid and bile salt tolerance, 15 strains were further selected. All selected strains were subjected to a series of in vitro tests to assess their technological properties, including cell surface hydrophobicity (13.6-56.2%), autoaggregation ability (9.26-38.30%), coaggregation ability, and heat and lysozyme survival rates (84.74-94.01% and 80.52-99.37%, respectively). In vitro antagonism showed that Lb. plantarum (M5-19, M8-60, M8-59) exhibited the most strong inhibitory activity against 7 tested pathogens. Moreover, antibiotic resistance and hemolytic activity were investigated for safety assessment. No strain exhibited hemolytic activity, and most of the strains were sensitive to a series of 14 antibiotics of clinical importance. Ultimately, the principal component analysis of all data obtained above showed that 2 Lb. plantarum strains (M8-59, M8-60) and Lb. paracasei M1-36 exhibited the best potential for their inclusion as adjunct functional cultures in local fermented dairy products.Genetic selection has been a very successful tool for the long-term improvement of livestock populations, and the rapid adoption of genomic selection over the last decade has doubled the rate of gain in some populations. Breeding programs seek to identify genetically superior parents of the next generation, typically as a function of an index that combines information about many economically important traits into a single number. In the United States, the data that drive this system are collected through the national dairy herd improvement program that began more than a century ago. The resulting information about animal performance, pedigree, and genotype is used to compute genomic evaluations for comparing and ranking animals for selection. However, the full expression of genetic potential requires that animals are placed in environments that can support such performance. The Agricultural Research Service of the US Department of Agriculture and the Council on Dairy Cattle Breeding collaborate to deliver staetailed data about the environment in which animals perform, including information about feeding, housing, milking systems, and infectious and parasitic load. However, many challenges accompany these new technologies, including a lack of standardization or validation, need for high-speed internet connections, increased computational requirements, and interpretations that are often not backed by direct observations of biological phenomena. This work will describe how US selection objectives are developed, as well as discuss opportunities and challenges associated with new technologies for measuring and recording animal performance.The objective of this study was to assess the effects of feeding negative dietary cation-anion difference (DCAD) dry cow diets on postpartum health. Cows from 4 commercial dairy farms in Ontario, Canada, were enrolled in a pen-level controlled trial from November 2017 to April 2019. Close-up pens (1 per farm), with cows 3 wk before expected calving, were randomly assigned to a negative DCAD [TRT; -108 mEq/kg of dry matter (DM); target urine pH 6.0-6.5] or a control diet (CON; +105 mEq/kg of DM with a placebo supplement). Each pen was fed TRT or CON for 3 mo (1 period) then switched to the other treatment for the next period, with 4 periods per farm. Urine pH was measured weekly until calving, and body condition score (BCS) was measured at enrollment and at 5 wk postpartum. Data from 15 experimental units [8 TRT and 7 CON, with 1,086 (TRT n = 681; CON n = 405) observational units (cows)] that received the assigned diet for >1 wk were included. The incidence of milk fever (MF), retained placenta (RP), metritis,.75) multiparous cows had reduced incidence of MF when fed TRT (TRT 2 ± 1%, vs. CON 13 ± 8%). We detected no treatment effects on RP, metritis, hyperketonemia, or PVD incidence. Cows fed TRT had lesser incidence of DA (1.7 ± 0.7% vs. 3.6 ± 1.6%) and tended to have lesser incidence of MAST compared with CON (1.8% ± 0.6% vs. 4.4 ± 1.4%). No treatment effect was detected on ≥1 disease events (TRT 38 ± 7%, vs. CON 42 ± 8%); however, multiparous cows on TRT were less likely to have ≥2 disease events than cows on CON (14 ± 4% vs. 23 ± 6%). Under commercial herd conditions, feeding prepartum diets with negative DCAD improved several measures of postpartum health.
