The interaction between histones and DNA is important for eukaryotic gene expression. A loose interaction caused, for example, by the neutralization of a positive charge on the histone surface by acetylation, induces a less compact chromatin structure, resulting in feasible accessibility of RNA polymerase and increased gene expression. In contrast, the formation of a tight chromatin structure due to the deacetylation of histone lysine residues on the surface by histone deacetylases enforces the interaction between the histones and DNA, which minimizes the chance of RNA polymerases contacting DNA, resulting in decreased gene expression. Therefore, the balance of the acetylation of histones mediated by histone acetylases (HATs) and histone deacetylases (HDACs) is an issue of transcription that has long been studied in relation to posttranslational modification. In this review, current knowledge of HDACs is briefly described with an emphasis on recent progress in research on HDACs, especially on class IIa HDACs.Light and nutrient are important factors for vegetable production in plant factory or greenhouse. The total 12 treatments which contained the combination of four light intensity (150, 250, 350 and 450 μmol · m-2 · s-1) and three nutrient solution concentration (NSC) (1/4, 1/2, 3/4 strength NSC) were established for investigation of lettuce growth and quality in a growth chamber. The combination of light intensity and NSC exhibited significant effects on photosynthetic pigment, nutritional quality, mineral content and antioxidant capacity. That a higher light intensity were readily accessible to higher chlorophyll a/b showed in lettuce of treatment of 350 μmol · m-2 · s-1 × 3/4NSC and 450 μmol · m-2 · s-1 × 1/4NSC. Lower total N contents, higher content of soluble protein, vitamin C, soluble sugar and free amino acid exhibited in lettuce under treatment of 250 and 350 μmol · m-2 · s-1 × 1/4NSC or 3/4NSC. With increasing NSC and LED irradiance, the content of total P and K in lettuce increased and decreased, respectively. The highest and lowest total Ca content were found in treatment of 150 μmol · m-2 · s-1 × 1/4NSC and 450 μmol · m-2 · s-1 × 1/4NSC, respectively, and higher content of total Mg and Zn was observed under 250 μmol m-2 s-1 × 1/4NSC and 150 μmol · m-2 · s-1 × 3/4NSC, respectively. The antioxidant contents generally decreased with increasing NSC level. The higher antioxidant content and capacity occurred in lettuce of 350 μmol · m-2 · s-1 × 1/4NSC treatment. The interaction of 350 μmol · m-2 · s-1 × 1/4NSC might be the optimal condition for lettuce growth in plant factory.An amendment to this paper has been published and can be accessed via a link at the top of the paper.The strengths and limits of isotopically labelled Engineered Nanoparticles (spiked ENPs) spread in biological media have been assessed. Multi-spiked CdSe/ZnS quantum dots (QDs), measuring 7 nm and coated with thioglycolic acid (TGA), were synthesized and enriched in 68Zn, 77Se and 111Cd. These QDs were dispersed at very low concentrations (0.1 to 5000 ppt) in diverse biological matrices (synthetic saliva, synthetic urine, plasma and Dulbecco's phosphate buffered saline - DPBS growth medium) and the isotopic compositions were determined by HR-ICP-MS. The initial QDs concentrations were calculated to assess the limit of quantification (QD-LOQ) according to the matrix and the isotopically enriched element. The obtained results demonstrated the advantages of the isotopic labelling method in order to work at very low concentrations the QD-LOQ values for the spiked Zn, Cd and Se originated from the QDs were 10, 0.3 and 6 ppt, respectively, which is below the conventional LOQ of the HR-ICP-MS used (30, 3 and 60 ppt for Zn, Cd and Se, respectively). Conversely, in complex matrices such as saliva, urine, plasma and DPBS growth medium, the QD-LOQ values increased significantly, with values ranging from 16 to 32 ppt for Cd, 446 to 10598 ppt for Zn and 1618 to 8317 ppt for Se. These QD-LOQs are dependent on factors as the elemental background concentration already present in the matrices, and the dilution factor. CRC occurs sporadically in the majority of cases, indicating the predominant cause of the disease are environmental factors. Diet-induced changes in gut-microbiome are recently supposed to contribute on epidemics of CRC. This study was aimed to investigate the association of metagenomics and metabolomics in gut extracellular vesicles (EVs) of CRC and healthy subjects. A total of 40 healthy volunteers and 32 patients with CRC were enrolled in this study. Metagenomic profiling by sequencing 16 S rDNA was performed for assessing microbial codiversity. We explored the small molecule metabolites using gas chromatography-time-of-flight mass spectrometry. In total, stool EVs were prepared from 40 healthy volunteers and 32 patients with CRC. Metagenomic profiling demonstrated that bacterial phyla, particularly of Firmicutes and Proteobacteria, were significantly altered in patients with colorectal cancer. In summary, the present findings suggest that gut flora dysbiosis may result in alternation of amino acid metabolism, which may be correlated with the pathogenesis of CRC.We evaluated the changes in visual field mean deviation (VF MD) and retinal nerve fibre layer (RNFL) thickness in glaucoma patients undergoing trabeculectomy. One hundred patients were examined with VF and spectral-domain optical coherence tomography (OCT) before trabeculectomy and 4 follow-up visits over one year. Linear mixed models were used to investigate factors associated with VF and RNFL. VF improved during the first 3 months of follow-up (2.55 ± 1.06 dB/year) and worsened at later visits (-1.14 ± 0.29 dB/year). RNFL thickness reduced by -4.21 ± 0.25 µm/year from 1st month of follow-up. Eyes with an absence of initial VF improvement (β = 0.64; 0.30-0.98), RNFL thinning (β = 0.15; 0.08-0.23), increasing intraocular pressure (IOP; β = -0.11; -0.18 to -0.03) and severe glaucoma (β = -10.82; -13.61 to -8.02) were associated with VF deterioration. Eyes with VF deterioration (β = 0.19; 0.08-0.29), increasing IOP (β = -0.09; -0.17 to -0.01), and moderate (β = -6.33; -12.17 to -0.49) or severe glaucoma (β = -19.58; -24.63 to -14.52) were associated with RNFL thinning. Changes in RNFL structure and function occur over a 1-year follow-up period after trabeculectomy. link3 Early VF improvement is more likely to occur in patients with mild/moderate glaucoma, whereas those with severe glaucoma show greater decline over one year. Our findings indicate that progression is observable using OCT, even in late-stage glaucoma.This study evaluated stereopsis and other visual functions in patients with idiopathic macular hole (MH), and sought to identify vision-related parameters that affect stereopsis. In this prospective, consecutive, comparative study, 39 eyes of 39 patients with unilateral idiopathic MH were included. At baseline and at 6 months after MH surgery, we evaluated stereopsis, with the Titmus stereo test (TST) and TNO stereotest (TNO), best-corrected visual acuity (BCVA), letter contrast sensitivity, severity of metamorphopsia, as assessed using M-CHARTS, and extent of aniseikonia, by the new aniseikonia test. Preoperative stereopsis (log) in patients with MH were 2.72 ± 0.53 (range 1.9-4.1) in the TST and 2.82 ± 0.65 (range 1.8-3.9) in the TNO. Preoperative TST was significantly correlated with letter contrast sensitivity (p less then 0.05), but not with the other visual functions. TNO showed significant correlation with letter contrast sensitivity (p less then 0.05) and aniseikonia (p less then 0.005).
