To assess in a cross-sectional study the impact of including dental professionals in the multidisciplinary treatment team of head and neck squamous cell carcinoma (HNSCC) patients on the long-term oral health status.

Oral health status, dental care behaviours, and oral health-related quality of life were assessed based on a clinical and radiographic examination, interview, and medical records in patients treated for HNSCC ≥ 6months ago. This patient group ('cohort 2') was treated in a multidisciplinary treatment team including dental professionals and compared to a group of HNSCC patients previously treated at the same university, but without dental professionals included in the multidisciplinary treatment team ('cohort 1').

Cohort 2 consisted of 34 patients, who had received a dental check-up and if necessary, treatment by dental professionals prior to the initiation of cancer treatment. This cohort showed significantly improved oral hygiene habits and a better periodontal health status compared to cohort 1. However, cohort 2 still presented high demand for treatment due to active carious lesions; only a few, statistically insignificant improvements were detected compared to cohort 1.

Including dental professionals in the multidisciplinary treatment team of HNSCC patients has a positive impact on patient oral health status-primarily in terms of periodontal disease-6months and longer after finishing cancer therapy.

The study aims to investigate the shear bond strength (SBS) between silicate ceramic restorations and ceramic brackets after different pretreatments and aging methods.

Leucite (LEU) and lithium disilicate (LiSi) specimens were pretreated with (i) 4% hydrofluoric acid + silane (HF), (ii) Monobond Etch&Prime (MEP), (iii) silicatization + silane (CoJet), and (iv) SiC grinder + silane (SiC). Molars etched (phosphoric acid) and conditioned acted as comparison group. SBS was measured after 24h (distilled water, 37°C), 500 × thermocycling (5/55°C), and 90days (distilled water, 37°C). Data was analyzed using Shapiro-Wilk, Kruskal-Wallis with Dunn's post hoc test and Bonferroni correction, Mann-Whitney U, and Chi
test (p < 0.05). The adhesive remnant index (ARI) was determined.

LEU pretreated with MEP showed lower SBS than pretreated with HF, CoJet, or SiC. LiSi pretreated with MEP resulted in lower initial SBS than pretreated with HF or SiC. After thermocycling, pretreatment using MEP led to lower SBS than with CoJet. Within LiSi group, after 90days, the pretreatment using SiC resulted in lowest SBS values. After HF and MEP pretreatment, LEU showed lower initial SBS than LiSi. After 90days of water storage, within specimens pretreated using CoJet or SiC showed LEU higher SBS than LiSi. Enamel presented higher or comparable SBS values to LEU and LiSi. With exception of MEP pretreatment, ARI 3 was predominantly observed, regardless the substrate, pretreatment, and aging level.

MEP pretreatment presented the lowest SBS values, regardless the silicate ceramic and aging level. Further research is necessary.

Multicolor detection of ALP in saliva is ideal for a point-of-care diagnosis; however, this approach is very challenging since spectral responses over wavelengths of several tens of nanometers have thus far remained difficult to achieve. In this work, a colorimetric biosensor for ALP assay has been developed based on ALP affinity to dephosphorylate glucose phosphate into glucose, which has the affinity to deposit Ag nanoshells onto Au nanobipyramids with a multicolor response. This approach provides a blue shift of localized surface plasmon resonance (LSPR) as large as 190 nm corresponding to distinctive color changes, from yellowish brown to red based on the thickness of the formed Ag shell around the Au nanobipyramids. The change in the LSPR has been conducted for highly sensitive quantitative bioassay of ALP with a detectable multicolor change with linear dynamic range of 0.1-20 U/L and low limit of detection (LOD) of 0.085 U/L. Furthermore, the developed multicolor ALP biosensor exhibits high selectivity with high recovery of 98.6% demonstrating its reliability and suitability for a point-of-care diagnosis.Leprosy reactions are immune processes that cause neural damage in individuals with leprosy. As periodontitis is an infectious disease related to its development, specific antibodies to periodontal pathogens must be evaluated to better understand the humoral mechanisms underlying this relationship. Therefore, the objective of this study was to standardize an immunoassay to measure IgA specific to P. gingivalis antigens in the saliva of individuals with leprosy. An ELISA checkerboard titration was performed. A validation test involving 53 individuals with leprosy, 24 with and 19 without periodontitis, was conducted and a ROC curve constructed to calculate sensitivity and specificity. The coefficient of the optical densities was 2.21 and 2.66 for P. gingivalis crude extract and the recombinant protein HmuY, respectively. Sensitivity and specificity for the P. gingivalis crude extract were 66.7% and 73.7%, respectively, and for HmuY, were 62.5% and 52.6%, respectively. We assessed the utility of formalin-fixed paraffin-embedded (FFPE) tissues, which are readily available, for evaluation of high-risk HPV infection in oral cancer. For protocol standardization, we used 20 prospectively collected paired FFPE and fresh tissues of histopathologically confirmed oral cancer cases reported in Oral Medicine department of a dental hospital for comparative study. Only short PCRs (~ 200 bp) of DNA isolated using a modified xylene-free method displayed a concordant HPV result. For HPV analysis, we used additional 30 retrospectively collected FFPE tissues. DNA isolated from these specimens showed an overall 23.4% (11/47) HPV positivity with detection of HPV18. Comparison of HPV positivity from dental hospital FFPE specimens with overall HPV positivity of freshly collected oral cancer specimens (n = 55) from three cancer care hospitals of the same region showed notable difference (12.7%; 7/55). Further, cancer hospital specimens showed HPV16 positivity and displayed a characteristic difference in reported sub-sites and patient spectrum. Overall, using a xylene-free FFPE DNA isolation method clubbed with short amplicon PCR, we showed detection of HPV-positive oral cancer in dental hospitals.
The value of metabolomics in multi-systemic mitochondrial disease research has been increasingly recognized, with the ability to investigate a variety of biofluids and tissues considered a particular advantage. Although minimally invasive biofluids are the generally favored sample type, it remains unknown whether systemic metabolomes provide a clear reflection of tissue-specific metabolic alterations.

Here we cross-compare urine and tissue-specific metabolomes in the Ndufs4 knockout mouse model of Leigh syndrome-a complex neurometabolic MD defined by progressive focal lesions in specific brain regions-to identify and evaluate the extent of common and unique metabolic alterations on a systemic and brain regional level.

Untargeted and semi-targeted multi-platform metabolomics were performed on urine, four brain regions, and two muscle types of Ndufs4 KO (n≥19) vs wildtype (n≥20) mice.

Widespread alterations were evident in alanine, aspartate, glutamate, and arginine metabolism in Ndufs4 KO mice; while brain-region specific metabolic signatures include the accumulation of branched-chain amino acids, proline, and glycolytic intermediates. Furthermore, we describe a systemic dysregulation in one-carbon metabolism and the tricarboxylic acid cycle, which was not clearly reflected in the Ndufs4 KO brain.

Our results confirm the value of urinary metabolomics when evaluating MD-associated metabolites, while cautioning against mechanistic studies relying solely on systemic biofluids.The pollution of Iranian agricultural soils with trace elements (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, and Zn) was assessed and compared with other agricultural soils around the world. Experimental data on trace element content in Iranian agricultural areas in the west and north were collected from the literature 39 studies covered a total of 46 agricultural sites of 17 provinces in Iran, in order to characterize their patterns of accumulation of trace elements. Two pollution indices, namely, the pollution index (PI) and the integrated pollution index (IPI), were used to evaluate trace element accumulation. The data revealed a remarkable variation in trace element content among soils in different areas. Exploratory data analyses (EDAs) showed that a number of trace elements (Pb, Cu, and Zn) are asymmetrically distributed and scattered. Surveys indicated that 45.5% of the studied samples had elevated PI values for Cd, 13.0% for Cu, and 16.7% for Pb, clearly indicating an anthropogenic contribution of these three elements. The IPI of the agricultural soils also indicated that most areas are classified as having moderate and high pollution. Higher contents of trace elements (except for Mn) were found in some cities of the Isfahan, Hamadan, and Tehran provinces. Excessive application of conventional and organic fertilizers, pesticides, animal manure, and sewage sludge for enhancing crop production is responsible for high trace element content in Iran's agricultural soils. This in turn, through the food chain, is a threat to human health. Analysis of the correlation between trace elements exhibited that Cu, Pb, and Zn (Cd, Pb and Zn) were very closely associated with each other, showing that their prevalent sources are common and the efforts to regulate them linked in common actions. We consider this evaluation as a viable approach to other similar areas in the Middle East and beyond, which could be used by environmental scientists for risk assessment and decision making.
