Programmed cell death is a central process in the control of tissue development, organismal physiology, and disease. We recently interrogated placental ferroptosis in placental dysfunction in human and mouse pregnancy, dissected its relevance to placental injury, and validated the role of glutathione peroxidase-4 in guarding placental trophoblasts against ferroptotic injury. We also uncovered a role for the phospholipase PLA2G6 (PNPLA9) in attenuating trophoblast ferroptosis. Here, we summarize current data on trophoblast ferroptosis, and the role of several proteins and microRNAs as regulators of this process. Our text offers insights into new opportunities for regulating ferroptosis as a means for protecting placental trophoblasts against lipotoxic injury.
Ovarian cancer is the most lethal gynecologic malignancy due to the tumor's acquisition of chemoresistance to platinum-based chemotherapy. To solve this problem, we conducted RNAi-based large-scale screening and determined that tyrosine kinase with immunoglobulin-like and EGF-like domains 1 (TIE-1) is a key molecule involved in the platinum resistance of ovarian cancer cells. Recently, a variety of studies have investigated that small extracellular vesicles (sEVs) contribute to the communication between cancer cells, including the development of chemoresistance in ovarian cancer. The purpose of our study is to determine if sEVs-derived TIE-1 is involved in the chemoresistance of ovarian cancer cells.

TIE-1-overexpressed TOV112D cells, termed TOV112D
cells, were established, and sEVs were isolated from TOV112D
cells supernatants by ultracentrifugation. We assessed cisplatin sensitivity in recipient cells with TOV112D
-derived sEVs by cell-Titer Glo kit. We also asked whether sEV-derived TIE-1 suppressed the DNA damage response in recipient cells and evaluated the DNA damage response by counting cells positive for DNA damage foci.

. We showed that sEV-derived TIE-1 decreased chemosensitivity to cisplatin by suppressing the DNA damage response in recipient cells.

Male breast cancer (MBC) accounts for 1% of all breast cancers and there is a paucity of data on factors impacting the treatment strategies and outcomes. We sought to use a large national database to examine trends and predictors of the use of adjuvant radiation (Adj-RT), as well as any association with outcome.

We queried the National Cancer Database (NCDB) for patients with stages I-III MBC treated with surgery (breast conservation surgery-BCS or mastectomy-MS) within 180 days of diagnosis (years 2004-2015). Multivariable logistic regression identified predictors of adj-RT receipt. Multivariable Cox regression evaluated predictors of survival. Propensity matching for adj-RT was used to account for indication biases.

We identified 6,217 patients meeting the eligibility criteria (1457 BCS vs. 4760 MS). The majority of patients were Caucasian (85%) and in an age range of 50-80 years (74%). Although adj-RT was omitted for 30% of BCS patients, the utilization was higher compared to MS (OR=26, p-value=0.001). The predictors of adj-RT use included African-American race, more advanced stage, higher grade, presence of lymphovascular invasion, and ER/Her-2 positivity for the entire cohort and increased age, urban location and higher income for BCS. Adj-RT was associated with lower mortality in the propensity matched model (overall HR for BCS=0.28, p-value<0.001; overall HR for MS=0.62, p-value=0.001).

These increased VEGF levels are considered to having a positive contribution on neurological side effects. Neurotoxicity that is related with the usage of sunitinib malate for two weeks will be presented in this case report.Dissolution fluxes of stable (127I) and radioactive (129I) isotopes of iodine from a brackish lake sediment beside a spent nuclear fuel reprocessing plant in Japan were evaluated through two kinds of experiments incubation using a sediment core sample for 24 h, and observation of 127I and 129I concentrations in sedimentary pore water. For 127I, the dissolution flux evaluated in the incubation experiment was comparable with that obtained from the vertical gradient of 127I concentration in pore water in the observation experiment. This suggests that degradation of organic matter in the surface sediment is an important source of dissolved 127I found in the water. For 129I, the dissolution flux estimated in the incubation experiment showed negative values, indicating the transfer of 129I to the sediment from the overlying water (i.e., absorption). Moreover, the flux evaluated from the observation experiment was positive. learn more This result suggests that degradation of organic matter in the surface sediment is scarcely important to the supply of 129I from the sediment to the water in the studied lake. The mean concentrations of both radionuclides in the soil were clearly greater than the UNSCEAR estimated worldwide average while that of the rice with values of 11.7 ± 4.3 and 3.0 ± 1.3 Bq/kg in field 1 and 8.5 ± 4.2 and 4.3 ± 1.6 Bq/kg in field 2 for 238U and 232Th respectively, were well below the worldwide average. The radionuclide transfer factors (TFs) across both depths ranged from 0.03 to 0.67 and 0.01 to 1.20 for 238U and 232Th respectively. The overall geometric means and geometric standard deviations of the TF estimated in field 1 were respectively 0.22 and 1.52 for 238U and 0.18 and 4.21 for 232Th (soil,0-4cm); 0.43 and 1.80 for 238U and 0.18 and 3.79 for 232Th (soil,4-8cm) whereas in field 2 the values were respectively 0.12 and 2.62 for 238U and 0.06 and 4.15 for 232Th (soil,0-4cm); 0.11 and 1.85 for 238U and 0.12 and 4.15 for 232Th (soil,4-8cm). These enzymes perform conversion of d-amino acids to corresponding imino acids followed by conversion into α-keto-acids. In the present study Mtb-Ra recombinants with DAO knockout (KO) and knockout complemented with DAO over-expressing plasmid (KOC) were constructed. The growth studies showed loss of growth of KO in medium containing glycerol as a primary carbon source. Substituting glycerol with acetate or with FBS addition, restored the growth. Growth was also restored in complemented strain (KOC). KO showed increased permeability to hydrophilic dye EtBr and reduced biofilm formation. Also, its survival in macrophages was low. Phagosome maturation studies suggested enhanced colocalization of KO, compared to WT, with lysosomal marker cathepsin D. Also, an increased intensity of Rab5 and iNOS was observed in macrophages infected with KO, compared to WT and KOC. The in vivo survival studies showed no increase in CFU of KO. This is the first study to show functional relevance of DAO encoded by MRA_1916 for Mtb-Ra growth on glycerol, its permeability and biofilm formation. Also, this study clearly demonstrates that DAO deletion leads to Mtb-Ra failing to grow in macrophages and in mice.
