The developed innovative coatings show great applicative potential as a new active packaging for perishable foods.This study aimed to understand how the microstructure of gelled foods impacts the diffusion of a volatile antimicrobial compound and its efficacy at different depths from the surface. Carvacrol-loaded polylactic acid film was used to inhibit the growth of Pseudomonas fluorescens in WPI-carrageenan gels during storage at 4 °C. The diffusion of antimicrobials was increased in gels having larger average pore size. The antimicrobial efficacy of the antimicrobial packaging was dependent on the diffusion of carvacrol within the gels. The final concentration of carvacrol in the top layer was more than 4 fold higher than that in the middle layer and more than 13-fold higher than that in the bottom layer, resulting in a more effective inhibition in the top layer than those in the middle and bottom layers. Our study demonstrates the importance of considering the diffusion of antimicrobials in solid/semi-solid foods in the antimicrobial packaging design.Hyperstable cysteine-rich peptides (CRPs) represent an underexplored superfamily of bioactives in functional foods. An example is wolfberry of the Lycium barbarum family. Previously, we discovered a CRP, designated α-lybatide, from L. barbarum bark. Herein, we report the discovery of β-lybatide, a novel carboxypeptidase inhibitor belonging to a different CRP family from the wolfberry plant. Proteomic and transcriptomic analyses showed that β-lybatide contains 36 amino acids with six cysteine residues. NMR spectroscopy revealed that β-lybatide displays a knottin-like structure that renders it highly resistant to thermal, chemical and enzymatic degradation, conditions important for keeping its structural integrity in gastrointestinal tract. Biochemical assays showed that β-lybatide is a potent carboxypeptidase inhibitor which could contribute to the wolfberry biological activities. Bioinformatics analysis revealed an additional 49 β-lybatide-like plant carboxypeptidase inhibitors. Together, our results show that β-lybatide is the first and the smallest plant-derived hyperstable carboxypeptidase inhibitor discovered from a functional food.Adulteration of food ingredients, particularly replacement of high-value milk with low-cost milk, affects food safety. For rapid and accurate identification of the possible adulterating milk species in an unknown sample, a centrifugal microfluidic chip-based real-time fluorescent multiplex loop-mediated isothermal amplification (LAMP) assay was developed to simultaneously detect milk from cow, camel, horse, goat, and yak. Using precoated primers in different reaction wells, the centrifugal microfluidic chip markedly simplified the detection process and reduced false-positive results. The entire amplification was completed within 90 min with a genomic detection limit of 0.05 ng/µL in cow, camel, horse, and goat milk and 0.005 ng/µL in yak milk. Using simulated adulterated samples for validation, the detection limit for adulterated milk samples was 2.5%, satisfying authentication requirements, as the proportion of adulterated milk higher than 10% affects economic interests. Therefore, this simple, centrifugal, microfluidic chip-based multiplex real-time fluorescent LAMP assay can simultaneously detect common milk species in commercial products to enable accurate labeling.A ligand film with citric acid (CA) on the surface as a biochelator was prepared via photografting. Polypropylene film was photochemically brushed by immobilizing glycidyl methacrylate onto the film surface (PP-g-GMA) in the presence of benzophenone. The ligand film (PP-g-GMA-g-CA) was developed via a ring-opening reaction between PP-g-GMA and CA. The amount of green biomass has significantly decreased, whereas the chlorophylls a and b have not been affected. The biggest changes (increase/decrease) have been observed in the contents of p-coumaric and chlorogenic acid, quercetin and quercitrin.A method for early quantification of unripe macaw fruits oil content using near-infrared spectroscopy (NIR) and partial least squares (PLS) is presented. After harvest, the fruit takes about 30 days to reach its maximum oil accumulation. The oil content was quantified thirty days after harvest using Soxhlet extraction. PLS models were built using NIR spectra of shell obtained five days after harvest (Shell5). The Shell5 model was compared with models built using NIR spectra of the shell (Shell30) and mesocarp thirty days after harvest (Pulp30). Ordered predictors selection was used to select the most informative variables. The best models presented root mean square error of prediction and correlation coefficient of prediction of 4.87% and 0.89 for Shell5; 5.83% and 0.85 for Shell30; 4.76% and 0.92 for Pulp30. Thus, the anticipated prediction of oil content could reduce the time and costs of macaw palm quality control and storage.
