Secondary objectives were to compare the composition of the NP microbiota between calves by age, clinical respiratory score (CRS), and previous antibiotic therapy. Holstein heifer calves (n = 50) from a southern Wisconsin dairy were enrolled at either 3 or 6 wk of age; 4 calves were sampled at both time points. Antibiotic treatment history was also collected for the 30 d before enrollment. For the purpose of this study, pneumonia was defined as having lobar pneumonia, as diagnosed by US, in at least 1 lung lobe. Following examination by CRS and US, a deep nasopharyngeal swab was obtained for 16S rRNA amplicon sequencing. Alpha diversity was reduced in calvespreviously treated with antibiotics.Genetic groups, also called unknown or phantom parents groups, are often used in dairy cattle genetic evaluations to account for selection that cannot be accounted for by known genetic relationships. With the advent of genomic evaluations, the theory of genetic groups was extended to the so-called single-step genomic BLUP (ssGBLUP). In short, genetic groups can be fitted in ssGBLUP through regression effects, or by including them in the pedigree and computing the adequate combined pedigree and genomic relationship matrix. In this study, we applied the so-called Quaas and Pollak transformation to a system of equations for single-step SNP BLUP (ssSNPBLUP), such that genetic groups can thereafter be included in the pedigree. The example in this study showed that including the genetic groups in the pedigree for ssSNPBLUP allowed reduced memory burden and computational costs in comparison to genetic groups fitted as covariates.This study aimed to investigate the survival of the foodborne pathogen Escherichia coli O157H7 in white-brined cheeses as influenced by the presence of Lactobacillus reuteri. The white cheeses were made from pasteurized bovine milk inoculated with E. coli O157H7 (cocktail of 3 strains) to achieve ∼5 log10 cfu/g with absence or presence of Lb. reuteri (∼6 log10 cfu/g). Cheese samples were brined in 10% or 15% NaCl solution and stored at 10°C and 25°C for 28 d. The white-brined cheeses were assessed for salt content, pH, water activity (Aw), and numbers of E. coli O157H7, Lb. reuteri, nonstarter lactic acid bacteria (NSLAB), yeasts, and molds. Results showed that E. coli O157H7 survived in cheese stored in both brine solutions at 10°C and 25°C regardless of the presence of Lb. reuteri. A substantial reduction was observed in cheese stored in 10% NaCl brine at 25°C, followed by cheese stored in 15% NaCl brine at 10°C by 2.64 and 2.16 log10 cfu/g, respectively, in the presence of Lb. reuteri and by 1.02 and 1.87 log10 cfu/g, respectively, in the absence of Lb. reuteri under the same conditions. The pathogen in brine solutions survived but at a lower rate. Furthermore, the growth of Lb. reuteri and NSLAB were enhanced or slightly decreased in cheese and brine by 28 d, respectively. The salt concentrations of cheese ranged from 4 to 6% and 5 to 7% (wt/wt), during 28-d ripening in 10 and 15% brine, respectively. Values of pH and Aw slightly increased at d 1 after exposure to brine and reached 4.69 to 6.08 and 0.91 to 0.95, respectively, in all treatments. We also aimed to determine whether shelter provision during the prepartum period influenced blood analytes related to energy metabolism, body cleanliness, and health status. In this study, 2 cohorts of 12 multiparous Holstein prepartum cows were tested in winter. Twenty-five days before their expected calving date, the cows in each cohort were paired and randomly assigned to an open paddock without shelter or to one with access to an artificial shelter until calving. Shelter use, lying time, number of lying bouts, duration of lying bouts, rumination time, and feeding time were continuously recorded during the 3 wk before calving. Cows were assessed weekly for body cleanliness throughout the 3-wk prepartum period. A blood sample was taken from the coccygeal vein of each cow at wk -3, -2, -1, 1,in daily rumination time, β-hydroxybutyrate concentrations, or postpartum health disorders. Despite the small number of dairy cows used in this study, these findings suggest that having access to a shelter during the prepartum period increases lying time, improves body cleanliness, and reduces adipose mobilization. Therefore, it is important to provide a protected area for the welfare of prepartum dairy cows exposed to winter climate conditions.Two experiments were conducted to evaluate the effect of isopropyl ester of 2-hydroxy-4-(methylthio) butanoic acid (HMBi) on lactation performance of dairy cows. Experiment 1 evaluated the effect of HMBi in diets with 15.3% crude protein (CP) and with different proportions of rumen-degradable and undegradable protein. Variation in rumen-degradable and undegradable protein was achieved by replacing raw with heated soybeans. Experiment 2 was an on-farm trial to evaluate HMBi with a large number of observations and using a farm-formulated diet (17.2% CP). In experiment 1, 20 Holsteins at 100 ± 41 d in milk were allocated to 5 replicated 4 × 4 Latin squares with 21-d periods. Treatments were formed by a 2 × 2 factorial arrangement of raw or heated soybeans with or without HMBi. Paper capsules with HMBi were orally administered twice daily to each cow. Dosage of HMBi was 7.6 g of digestible Met/cow per day. There was no interaction between soybean type and HMBi. Heat-treated soybeans increased the yields of milk, teract with HMBi supplementation.The purpose of this research was to investigate the effects of replacing soybean meal (SBM) with a fermented corn gluten-wheat bran mixture (FCWM) on nutrient digestibility, lactation performance, plasma metabolites, ruminal fermentation, and bacterial communities in Holstein cows. Nine healthy multiparous (parity = 3) Holstein cows with similar body weights (624 ± 14.4 kg), days in milk (112 ± 4.2), and milk yields (31.8 ± 1.73 kg; all mean ± standard deviation) were used in a replicated 3 × 3 Latin square design with 3 periods of 28 d. Cows were fed 1 of 3 dietary treatments in which FCWM replaced SBM as follows basal diet with no replacement (0FCWM); 50% replacement of SBM with FCWM (50%FCWM); and 100% replacement of SBM with FCWM (100%FCWM). The diets were formulated to be isocaloric and isonitrogenous. The results showed that the total-tract digestibility of dry matter and crude protein increased linearly with increased dietary FCWM, and we found a trend for increased total-tract neutral detergent fiber ccaceae decreased linearly. Overall, the replacement of SBM with FCWM altered the composition of the ruminal bacterial community and improved nutrient digestibility, lactation performance, and ruminal fermentation in cows, providing a data reference for the use of FCWM in dairy production.Intact seaweed or seaweed extracts are used as feed supplements to improve the gut microbiome in young animals. Seaweeds provide functional polysaccharides, and they are a good source of vitamins, minerals, and phenolic compounds, all of which are relevant for immune system development. However, literature on the effects of dried seaweed supplementation on immune system development is limited, especially in calves. This experiment aimed to study the effect of feeding milk supplemented with Ulva lactuca, Ascophyllum nodosum, or Saccharina latissima on the systemic immune status of preweaning dairy calves. Forty male Holstein calves with birth body weight 41 ± 4 kg and plasma Brix percentage ≥8.7% at d 2 after birth were used in this study. Calves were fed 4 L of cow milk twice a day (total 8 L/d). From d 2 to d 28, calves in the control group (n = 10) received milk without seaweed supplementation. Over the same period, experimental calves received milk supplemented with Ulva lactuca (SW1; n = 10), Ascophyllum ontrol and seaweed-supplemented calves. Feeding milk supplemented with dried seaweed increased plasma concentrations of variables related to the innate immune response in preweaning dairy calves.Automated monitoring devices (AMD) have become more affordable, and consequently more popular among dairy producers. We hypothesized that the addition of AMD-generated health alerts to a health-screening program improves survival, milk production, and reproductive success. In addition, we hypothesized that cows diagnosed with clinical disease that have AMD alerts are at greater risk of culling, lower milk production, and decreased risk of pregnancy than cows without AMD alerts. Holstein cows (nulliparous = 282, parous = 328) were enrolled at -60 ± 3 d (d 0 = calving), when they were fitted with an AMD and assigned randomly to 1 of 2 health-screening strategies (1) control AMD alerts not provided to farm personnel; and (2) automated device AMD alerts provided to farm personnel. Twice daily, study personnel determined which cows had AMD alerts (health index ≤79, rumination less then 200 min/d, or difference between current rumination and the average of the 3 preceding days less then 0) and provided the informduced risk of pregnancy after the first 2 services (54.5 ± 3.0 vs. 46.2 ± 3.2%). Cows diagnosed with a clinical disease without AMD alerts had reduced risk of removal from the herd by 150 DIM (5.7 ± 2.0 vs. 19.0 ± 3.3%), greater risk of pregnancy after the first 2 services (49.6 ± 4.5 vs. 33.6 ± 3.9%), and greater milk by 22 wk postpartum (6.7 ± 0.2 vs. 5.3 ± 0.2 × 103 kg) than cows diagnosed with a clinical disease that had an AMD alert. Adding AMD-generated health alerts to the health screening of postpartum cows in a herd with an existing screening program did not improve survival, milk yield, or reproductive success. In addition, AMD alerts in cows diagnosed with a clinical disease may be indicative of the future success of such cows.
